Abstract (German)

PD-1 und sein Ligand PD-L1 spielen eine wichtige Rolle beim Tumor Immunescape. Zytotoxische T-Lymphozyten bilden bei Erkennung einer Zieltumorzelle eine immunologische Synapse im sogenannten Kiss of Death. Diese Arbeit untersucht die Lokalisation von PD-1 und deren Einfluss auf die T-Zell-Funktionalität während der Interaktion Zwischen T-Zelle und Tumor. Experimente mit PD-L1.Ig auf Beads erbrachten wegen sterischer Bindungshemmung der stimulatorischen Proteine keine validen Ergebnisse. Daher wurde stattdessen ein murines TCR transgenes Tumormodell etabliert und PD-1 defiziente, Wildtyp und PD-1 transgene T-Zellen charakterisiert. In Anwesenheit eines B7-Kostimulus fand sich bei repetitiver Stimulationen keine PD-1-Abhängigkeit bezüglich Aktivierung bzw. Exhaustion. In Abwesenheit des Kostimulus hingegen zeigte sich eine PD-1/PD-L1 vermittelte T-Zell-Inhibition in Abhängigkeit der PD-1 Konzentration auf T-Zellen. PD-L1 auf T-Zellen führte nicht zu einer inter-lymphozytären T-Zell-Hemmung. Immunfluoreszenz-mikroskopisch stellte sich die PD-1 Expression in Clustern nach T-Zell-Aktivierung, sowie die Akkumulation mit dem T-Zell-Rezeptor und dem MTOC an der Immunsynapse während der T-Zell-Tumor-Interaktion dar. PD-L1 auf T-Zellen erfährt dagegen keine Polarisierung. Während der Interaktion findet auch keine PD-L1-Polarisierung von Seiten des Tumors im Sinne eines Gegenangriffs statt. Es zeigte sich vor allem in exhausted T-Zellen eine Tendenz zur PD-1 vermittelten Hemmung der MTOC-Translokation und der Polarisierung zytotoxischer Granula.

Translation of the abstract (English)

PD-1 and its ligand PD-L1 play an important role in tumor immunescape mechanism. Cytotoxic T lymphocytes interact with the target tumor cell in forming the immunosynapse, the Kiss of Death. We examined the localization of PD-1 during the Kiss of Death and its influence on T cell functionality. Experiments using PD-L1.Ig on beads delivered no reliable data due to steric inhibition of costimulating factors onto the beads. Instead, we established a murine TCR transgenic tumor model and characterized PD-1 deficient, wildtype and PD-1 transgenic T cells. Repetitive stimulation in presence of B7-1 costimulus via P815.B71 did not lead to PD-1 dependent alteration of exhaustion. Co-culture of primed T cells with PD-L1 expressing melanoma cells led to impaired T cell functions of PD-1tg T cells compared to WT T cells, whereas PD-1ko T cells showed improved immune functions. Activation results in clustered expression of PD-1. PD-1 clusters orientate with the T cell receptor and the MTOC towards the immunological synapse during the Kiss of Death. PD-L1 allocation on the T cell surface is not altered during T cell-target interaction. PD-L1 is not moved on the tumor surface, excluding a counter attack of the tumor cell. PD-1 might influence the efficiency of MTOC and granzyme polarization during the Kiss of Death.
