Abstract (German)
Eine enantioselektive Totalsynthese von Xanthatin wird beschrieben. Diese Synthese umfasst eine stereoselektive Cyclopropanierung, sp3-sp3-Kreuzkupplung, Ringschlussmetathese und stereoselektive allylische Oxidation. Ebenfalls wurde eine Reihe unnatürlicher Xanthatin Derivate hergestellt, um Auswirkungen auf die biologische Aktivität zu überprüfen. Die Substanzen wurden gegen verschiedene ...
Eine enantioselektive Totalsynthese von Xanthatin wird beschrieben. Diese Synthese umfasst eine stereoselektive Cyclopropanierung, sp3-sp3-Kreuzkupplung, Ringschlussmetathese und stereoselektive allylische Oxidation. Ebenfalls wurde eine Reihe unnatürlicher Xanthatin Derivate hergestellt, um Auswirkungen auf die biologische Aktivität zu überprüfen. Die Substanzen wurden gegen verschiedene Krebszelllinien, Pilze und Bakterien getestet. Auch wurden entzündungshemmende Eigenschaften über den iNOS Weg bestimmt. Es konnten 4 Substanzen identifiziert werden, die entzündungshemmende Eigenschaften aufweisen sowie 6 Substanzen, die gegen Mucor hiemalis aktiv sind. Keine der Substanzen war gegen Bakterien aktiv. Die Meisten Derivate konnten gute Aktivitäten gegen L-929 Mausfibroblasten, A-549 humanen Lungenkrebs und KB3-1 humanen Gebärmutterhalskrebs aufweisen.
Translation of the abstract (English)
The dissertation describes a new enantioselective synthesis of the sesquiterpene lactone Xanthatin using a stereoselective cyclopropanation, sp3-sp3 cross coupling, ring closing metathesis and stereoselective allylic oxidation. Furthermore the derivatization of Xanthatin to several unnatural Xanthanolides is described to evaluate specific functional groups in biological activity tests. ...
The dissertation describes a new enantioselective synthesis of the sesquiterpene lactone Xanthatin using a stereoselective cyclopropanation, sp3-sp3 cross coupling, ring closing metathesis and stereoselective allylic oxidation. Furthermore the derivatization of Xanthatin to several unnatural Xanthanolides is described to evaluate specific functional groups in biological activity tests. Substances were tested against different mammalian cancer cell lines, fungi and bacteria as well as anti inflammatory effects via iNOs pathway were tested. While all of the substances were mostly inactive against bacteria, 6 derivatives could be found to have inhibition against Mucor hiemalis. 4 substances were identified to have significant anti inflammatory activities and most of the substances had good activities against L-929 mouse fibroblasts, A-549 human lung cancer and KB3-1 human cervix carcinoma.
