Abstract (German)
In der vorliegenden Arbeit wurde untersucht, welche Auswirkungen chronisch-psychosozialer Stress auf das Immunsystem besitzt. CSC induziert unreife, myeloide Zellen, die regulatorisch das Immunsystem beeinflussen und in der Literatur als myeloide Suppressorzellen (myeloid-derived supressor cells; MDSC) bekannt sind. Lymphozyten, B- und T-Zellen, waren nach CSC in sekundär lymphatischen Organen reduziert, wohingegen MDSC prozentual anstiegen. Ferner stieg das suppressive Verhalten monozytischer MDSC aus dem Knochenmark nach CSC um ein Vielfaches an. In einer Reihe von Versuchen wurde untersucht, wie MDSC im zeitlichen Verlauf von CSC in Knochenmark und Milz akkumulierten. Ein Teil dieser Arbeit beschäftigte sich mit den molekularen Mechanismen, die solch eine Akkumulation von MDSC begünstigten. Dazu wurde sowohl die Rolle von inflammatorischen Zytokinen, wie dem Tumornekrosefaktor (tumor necrosis factor; TNF) oder Interleukin 6 (IL-6), als auch die Rolle der beiden Haupt-Stresshormone Adrenalin und Corticosteron näher untersucht. Es wurde deutlich, dass die Aktivierung des TNF-Rezeptors Typ 2 (TNFR2) durch TNF für die Akkumulation von MDSC wesentlich ist. Hinsichtlich der beiden Haupt-Stresshormone zeigte sich, dass Adrenalin nur teilweise an der Akkumulation von MDSC beteiligt ist, während Glucocorticoide eine bedeutende Rolle für diesen Stress-vermittelten Effekt zu spielen scheinen. Behandlung naiver Tiere mit einem synthetischen Glucocorticoid erzeugte die gleichen Effekte, wie sie nach chronischem Stress festgestellt wurden. MDSC wurden ursprünglich in Patienten gefunden, die an einer Krebs Erkrankung leiden, und es besteht eine direkte Korrelation bezüglich der Anzahl an MDSC und der Tumor-Größe. Aufgrund der Tatsache, dass nach chronischem Stress ein vermehrtes Auftreten von MDSC nachgewiesen wurde, stellte sich die Frage, ob chronischer Stress Tumor-Wachstum begünstigt. In dieser Arbeit wird gezeigt, dass das Wachstum eines Fibrosarkoms nach CSC verstärkt war. Das gesteigerte Wachstum scheint vor allem durch eine erhöhte Angiogenese in der frühen Phase des Tumor-Wachstums vermittelt zu sein. Eine Vor-Behandlung naiver Tieren mit einem synthetischen Glucocorticoid zeigte jedoch keinen Einfluss auf das Tumorwachstum. Dies deutet darauf hin, dass neben Glucocorticoiden noch weitere Faktoren dafür verantwortlich sind, dass nach CSC das Tumorwachstum verstärkt war.

In the following thesis it was investigated, what is the effect of chronic-psychosocial stress on the immune system. CSC induced immature, myeloid cells, which act immune-regulatory and in the literature are known as myeloid-derived suppressor cells (MDSC). Lymphocytes, B- and T cells were reduced in secondary lymphatic organs, whereas MDSC were increased. Furthermore, it was shown that the suppressive capacity of monocytic MDSC from the bone marrow was increased following CSC. In a series of experiments the time point was investigated, when MDSC will accumulate in spleen and bone marrow. One part of this work was dealing with the molecular mechanisms, leading to an accumulation of MDSC. Therefore the role of both inflammatory cytokines like tumor necrosis factor (TNF) or Interleukin 6 (IL-6) and the two main stress hormones adrenalin and corticosterone was closer investigated. It was clear that activation of the TNF receptor type 2 (TNFR2) via TNF is essential for the accumulation of MDSC. Regarding the two major stress hormones it came out, that adrenalin is partially responsible for the accumulation of MDSC, whereas glucocorticoids played an important role at this effect. Treatment of naïve animals with synthetic glucocorticoids resulted in the same phenotype like chronic stress. MDSC were initially found in patients suffering from tumor and there is a direct correlation between the number of MDSC and tumor size. Based on the fact, that chronic stress induces an increased number of MDSC, it was hypnotized, if chronic stress increases tumor growth. Evidence supports hypothesis, it was shown, that chronic stress increases tumor growth. An increased angiogenesis in the early phase seems to be responsible for the increased tumor growth. Treatment animals with an glucocorticoid antagonist before CSC didn’t show an effect. This data show, that besides glucocorticoids, other factors are also responsible for these stress increased tumor growth.
