Dem humanen Gen icb-1 (“induced by contact to basement membrane 1”) kommt eine Rolle in in-vitro Proliferations- und Differenzierungs- sowie Wachstums- und Progressionsprozessen von gynäkologischen Tumorzellen zu.
Im Zuge der vorliegenden Arbeit wurde nun die Bedeutung von icb-1 für die Apoptose von Mamma-, Ovarial- und Endometriumkarzinomzellen untersucht. Hierfür wurde die apoptotische Aktivität von stimulierten Zellklonen mit icb-1-Knockdown und solchen mit normaler icb-1-Expression verglichen. Es wurden Krebszelllinien verwendet, die die genannten Tumorentitäten repräsentieren: MCF-7 (Mammakarzinom), SK-OV-3 (Ovarialkarzinom) und HEC-1B (Endometriumkarzinom). Vor Detektion der apoptotischen Aktivität wurden die Zellklone mit potenten Apoptose-induzierenden Zytostatika stimuliert. Die Apoptose wurde auf Zellebene direkt über Bestimmung der Caspase-3/7-Aktivität sowie indirekt über Proliferationsversuche gemessen. Auf mRNA-Ebene wurde die Expression Apoptose-assoziierter Gene nach Stimulation mittels Real-Time qPCR bestimmt.
Die Vermutung, dass icb-1 auch in die apoptotischen Prozesse von gynäkologischen Tumorzellen eingreift, konnte in den Versuchsreihen auf verschiedenen Ebenen bestätigt werden. Insbesondere bei den MCF-7-Klonen steigerte icb-1 die Empfindlichkeit gegenüber den meisten der eingesetzten Apoptoseinduktoren, es wirkte also proapoptotisch. Dass sich diese Ergebnisse teilweise auch bei den SK-OV-3- und HEC-1B-Klonen widerspiegelten, lässt die Vermutung zu, dass icb-1 nicht nur bei bestimmten Gewebetypen, sondern generell diese Wirkung entfaltet. Auf molekulargenetischer Ebene scheinen XAF1 (XIAP-associated factor 1) und XIAP (X-linked inhibitor of apoptosis protein) bei der Vermittlung der Apoptoseinduktion durch icb-1 eine zentrale Rolle zu spielen. Möglicherweise kommt in diesem Zusammenhang auch dem Caspase-unabhängigen Apoptoseinduktor AIF (apoptosis inducing factor) eine Bedeutung zu.
Mit seiner proapoptotischen Wirkung könnte icb-1 potentiell zur Verbesserung der Chemotherapiewirkung sowie zur Überwindung von Resistenzen gegenüber Zytostatika beitragen. Die gewonnen Erkenntnisse geben somit Anlass für weitere Studien bezüglich der Rolle von icb-1 als Tumorsuppressorgen und Chemosensitizer im Rahmen einer gerichteten Tumortherapie.

Translation of the abstract (English)

The human gene icb-1 (“induced by contact to basement membrane 1”) plays a part in the in-vitro proliferation, differentiation, growth and progression processes of gynaecological tumour cells.
This dissertation assesses the significance of icb-1 for the apoptosis of breast, ovarian und endometrial cancer cells by comparing the apoptotic activity of stimulated cell clones with icb-1-knockdown and those with normal icb-1-Expression, using the following cancer cell lines to represent these tumour entities: MCF-7 (breast cancer), SK-OV-3 (ovarian cancer) and HEC-1B (endometrial cancer). Prior to the measurement of apoptotic activity, the cell clones were stimulated with potent apoptosis-inducing cyctostatic drugs. Apoptosis was measured directly at cell level through assessing the caspase-3/7-activity, as well as indirectly through proliferation experiments. At mRNA-level, the expression of apoptosis-associated genes after stimulation was assessed through Real-time qPCR.
Serial experiments did confirm the hypothesis that icb-1 affects the apoptotic processes of gynaecologic tumour cells on a number of levels. With regards to the MCF-7-clones in particular icb-1 increased sensitivity to the used apoptosis-inducing drugs and thus acted in a pro-apoptotic way. The fact that these results were to some extent also mirrored with the SK-OV-3- and the HEC-1B-clones suggests that icb-1 may have this effect not just on some specific kinds of tissue, but more generally. On a molecular genetic level, XAF1 (XIAP-associated factor 1) and XIAP (X-linked inhibitor of apoptosis protein) appear to play a crucial role in the induction of apoptosis through icb-1. It is possible that in this context, the Caspase-independent apoptosis inductor AIF (apoptosis inducing factor) may be of significance.
On the basis of its pro-apoptotic effect, icb-1 could potentially help improve the efficacy of chemotherapy, as well as aid the overcoming of resistance towards cyctostatic drugs The work presented here could therefore form the basis for further studies into the role of icb-1 as a tumour suppressor gene and chemosensitizer as part of a directed tumour therapy.
