Abstract (German)
Der Nukleolus der Bäckerhefe ist ein Subkompartiment des Zellkerns, welches um die ribosomale DNA auf Chromosom XII assembliert. Neben ribosomaler DNA und RNA beinhaltet der Nukleolus eine Vielzahl an Proteinen, welche für Transkription ribosomaler Gene, Prozessierung der Primärtranskripte und auch Zellzyklusregulation von Bedeutung sind (Taddei und Gasser, 2012; Pederson, 2011). Ribosomale DNA wird in der Mitose symmetrisch auf Mutter und Tochter verteilt, der umgebende Zellkern hingegen wird asymmetrisch verteilt (Enserink und Kolodner, 2010; Jorgensen et al., 2007). Daher ergibt sich die Frage, welches Verhalten nukleoläre Proteine in der Mitose zeigen. Das Ziel dieser Arbeit war, einen tieferen Einblick in das mitotische Segregationsverhalten nukleolärer Proteine zu erhalten. Hierfür wurden endogene GFP-Fusionen von verschiedenen nukleolären Proteinen durch konfokale Fluoreszenzmikroskopie analysiert.
Es konnte festgestellt werden, dass nukleoläre Proteine in der Mitose drei verschiedene Segregationsmuster vorweisen. Von der RNA Polymerase I Untereinheit Rpa135, den Prozessierungsfaktoren Nop56 und Nsr1, sowie der RENT-Komponente Net1 erhielt der Nukleolus der Mutter im Zuge der mitotischen Segregation einen höheren Proteinanteil als der Nukleolus der Tochter. Die Verteilung dieser Proteine entsprach somit der des nukleoplasmatischen Proteins mCherry-NLS. Das rDNA bindende Protein Fob1 hingegen zeigte eine gleichmäßige Verteilung auf Mutter und Tochter, was ebenso für das stabil an DNA bindende Kernhiston Hta2 zutraf. Das rDNA-Bindeprotein Hmo1 wiederum, welches mit transkribierten Regionen der rDNA assoziiert, zeigte ein besonderes Verteilungsverhalten in der Mitose. Von diesem Protein erhielt der Nukleolus der Tochter den höheren Anteil und damit zeigte Hmo1 eine der Kernsegregation und den meisten anderen nukleolären Proteinen genau gegenläufige Asymmetrie.
Als grundlegender Punkt musste getestet werden, ob die asymmetrische Kernteilung einen entscheidenden Einfluss auf die Segregation nukleolärer Proteine nimmt. Sowohl Rpa135 als auch Hmo1 zeigten bei Bleichexperimenten effiziente nukleoläre Retention während der Anaphase. Zudem wurde in einer Δrpa12 Mutante ein gegenläufiger Effekt auf die Segregation nukleolärer Proteine und Nukleoplasma beobachtet. Nach diesen Ergebnissen sollte die Art der Assoziation eines nukleolären Proteins mit rDNA entscheidend für das Segregationsverhalten sein. Des Weiteren scheint Transkription durch RNA Polymerase I während der Anaphase in S.cerevisiae stattzufinden und die mitotische Segregation von nukleolären Proteinen zu beeinflussen.
Da die Chromosomentrennung in der Bäckerhefe deutlich asymmetrisch verläuft und eine spezielle Mechanik aufweist (Renshaw et al., 2010), wurde vermutet, dass dies der ausschlaggebende Faktor sein könnte. Bestätigt wurde dies durch in den Mutanten cdc14-3 und Δcin8 gemachte Beobachtungen. Hier konnte eine deutlich veränderte Trennungsmechanik ausgemacht werden und die Proteine Rpa135 und Hmo1 wurden wesentlich symmetrischer verteilt. Geometrie und Mechanik der Chromosomentrennung sollten also die mitotische Segregation nukleolärer Proteine bestimmen. Zudem konnte durch diese Experimente ausgeschlossen werden, dass der Knospenhals als mögliche Barriere die mitotische Segregation nukleolärer Proteine beeinflusst.
Das Zurückbleiben von Rpa135, Nop56 und Nsr1 im Nukleolus der Mutter kann durch effiziente nukleoläre Retention der Proteine in Verbindung mit dynamischer rDNA-Assoziation und der asymmetrischen Chromosomentrennung erklärt werden. Gleichverteilung wie im Falle von Fob1 sollte durch stabile DNA-Bindung zustande kommen. Das besondere Verhalten von Hmo1 allerdings kann mit diesen Argumenten nicht erklärt werden, weshalb Struktur-Funktionsanalysen für Hmo1 durchgeführt wurden. Hierbei wurden verschiedene Derivate von Hmo1 hinsichtlich subzellulärer Lokalisation und Nukleinsäurebindung mittels EMSA-Studien charakterisiert. rDNA-Bindung stellte sich als grundlegend, jedoch nicht hinreichend für normale Lokalisation und Segregation heraus. Zudem stellte Oligomerisierung eine Grundvoraussetzung dar. Des Weiteren zeigte Hmo1 in Situationen niedriger transkriptioneller Aktivität eine massive nukleoläre Anreicherung, welche ebenso von Oligomerisierung abhängig war und die Aktivität von RNA Polymerase I war notwendig für die Etablierung der Asymmetrie von Hmo1. Daher wird vermutet, dass das besondere Segregations-verhalten von Hmo1 durch seine intrinsischen Eigenschaften im Zusammenhang mit einer durch RNA Polymerase I verursachten Verdrängung zustande kommt. Während der Anaphase wird ein Schwesterchromatid aus dem Nukleolus der Mutter gezogen, wobei speziell das für die Tochter bestimmte Chromatid starke Streckungs- und Kompaktierungsereignisse durchläuft. Dadurch wird dieses vermutlich deshalb von RNA Polymerase I depletiert, da das Enzym dynamisch mit rDNA wechselwirkt und somit im Nukleolus der Mutter zurückgehalten wird. Innerhalb einer elongierten Anaphasestruktur könnte dann eine Verschiebung von Hmo1 in die töchterlichen Bereiche erfolgen, welche von Polymerase depletiert sind. Hier könnte Hmo1 durch rDNA-Bindung und Oligomerisierung massiv anreichern.
Vermutlich werden durch Geometrie und Mechanik der rDNA-Trennung unterschiedliche rDNA-Chromatinzustände etabliert. Der Mutternukleolus verfügt nach der Trennung über eine höhere Menge an RNA Polymerase I und Prozessierungsfaktoren als der Tochternukleolus und befindet sich vermutlich in einem transkriptionell hochaktiven Zustand. Der Tochternukleolus hingegen verfügt über eine höhere Menge an Hmo1 und befindet sich vermutlich in einem transkriptionell weniger aktiven Zustand.

The budding yeast nucleolus is a subnuclear compartment which assembles around ribosomal DNA located on Chromosome XII. In addition to ribosomal DNA and RNA, the nucleolus comprises various proteins, which are necessary for transcription of ribosomal genes, processing of the primary transcripts and cell cycle regulation (Taddei and Gasser, 2012; Pederson, 2011). Like any chromosomal DNA, rDNA is equally distributed between mother and daughter cells during mitosis (Enserink and Kolodner, 2010). On the contrary, the surrounding nucleoplasm is asymmetrically inherited (Jorgensen et al., 2007). Therefore, the question arises how nucleolar proteins are distributed in mitosis. The aim of this study was to get insight into the mitotic segregation behaviour of nucleolar proteins and the underlying mechanisms. To this end, endogenous GFP-fusions of nucleolar proteins were analyzed by confocal fluorescence microscopy.
The basic observation was that nucleolar proteins show three different mitotic segregation patterns. In case of the RNA polymerase I subunit Rpa135, the processing factors Nop56 and Nsr1, as well as the RENT component Net1, the mother nucleolus obtained a larger portion than the daughter nucleolus. This resembled the segregation behaviour of the nucleoplasmic protein mCherry-NLS. However, the rDNA binding protein Fob1 showed equal distribution, which was also the case for stably DNA bound core histone Hta2. Astonishingly, the rDNA binding protein Hmo1, which associates with transcribed rDNA regions, showed special segregation behaviour. For this protein, the daughter nucleolus obtained a larger portion than the mother nucleolus. Therefore, Hmo1 showed converse segregation compared to nucleoplasm and most other analyzed nucleolar proteins.
As a basic matter, a possible influence of the segregation of nucleoplasm on nucleolar protein segregation had to be tested. Several experiments showed that nuclear division is not the crucial factor for nucleolar protein segregation. In bleaching experiments, Rpa135 as well as Hmo1 displayed nucleolar retention and therefore no efficient exchange between two nucleolar structures in a nucleoplasmic continuum during anaphase. Furthermore, a differential effect on nuclear and nucleolar protein segregation could be observed in a Δrpa12 mutant. The conclusion was drawn that the nature of rDNA association should determine the mode of mitotic segregation for a nucleolar protein. The fact that a nucleolus specific segregation effect was observed in Δrpa12 suggests that rDNA transcription takes place during mitosis and that the organizing function of RNA polymerase I for the nucleolus plays a role for nucleolar protein segregation.
The fact that the process of chromosome segregation is highly asymmetric in budding yeast and shows special mechanics (Renshaw et al., 2010) led to the conclusion that this could be the crucial factor for nucleolar protein segregation. Confirmation came from experiments conducted in cdc14-3 and Δcin8 mutants. In those strains, geometry and mechanics of nucleolar segregation were severely affected and in addition Rpa135 and Hmo1 showed quite equal segregation. So geometry and mechanics of chromosome segregation should be crucial for nucleolar protein segregation. Furthermore, this experiments excluded the bud neck as a potential barrier influencing nucleolar segregation.
Mother retention can be explained with efficient nucleolar retention of the protein, dynamic rDNA association, and asymmetric chromosome segregation. This should be the case for the RNA polymerase I subunit Rpa135 and the processing factors Nop56 and Nsr1. The equal distribution of Fob1 should be caused by stable DNA binding. However, the special behaviour of Hmo1 cannot be explained with those arguments. Therefore, structure function analyses for Hmo1 were conducted to elucidate the mechanisms of nucleolar localization and mitotic segregation. Several derivatives of Hmo1 were characterized by microscopy and EMSAs. rDNA binding turned out to be essential, but not sufficient for normal localization and segregation. In addition, oligomerization is a requirement for normal behaviour of the protein. Hmo1 showed massive nucleolar enrichment in situations of low RNA polymerase I activity and therefor, oligomerization was a prerequisite. Furthermore, high RNA polymerase I activity was necessary for the establishment of Hmo1 asymmetry during mitosis. Therefore, the conclusion was drawn that the intrinsic properties of Hmo1 and a displacement mechanism caused by polymerase I could work together to establish the special segregation behaviour. During anaphase, one sister chromatid is being pulled out of the mother nucleolus in direction of the bud. During this process, only the daughter destined chromatid experiences massive stretching and recoiling. This sister chromatid gets depleted from RNA polymerase I supposedly because of the dynamic rDNA association of the enzyme and resulting mother retention. Within the elongated structure, Hmo1 could move to daughter located regions which are depleted from RNA polymerase I. Here, it should be able to bind massively through direct rDNA binding and oligomerization. After complete segregation, no exchange can occur between the two nucleoli and the asymmetries are finally established.
Therefore, geometry and mechanics of rDNA segregation seem to cause the establishment of differential rDNA chromatin states in mother and daughter cells. The mother nucleolus gets a larger portion of RNA polymerase I as well as processing factors and is supposedly in a highly transcriptional state. On the contrary, the daughter nucleolus gets a larger portion of Hmo1 and is supposedly in a state of low transcriptional activity.
