Zusammenfassung
MIA (Melanoma Inhibitory Activity) ist ein wichtiges Protein, das an der Progression und Metastasierung des malignen Melanoms beteiligt ist. MIA wird von Melanomzellen exprimiert und migrationsabhängig am distalen Zellpol sekretiert. Im Extrazellularraum fördert es die Ablösung der Tumorzellen von der Extrazellulären Matrix (Extracellular matrix, ECM) und dadurch die Migration und Invasion von Melanomzellen.
Im Rahmen dieser Arbeit wurden die gerichtete Sekretion von MIA und die Prozessierung des Signalpeptids analysiert. Dafür wurde zunächst die Lokalisation und Sekretion von MIA in transient mit pCMX PL1 MIA transfizierten HeLa Zellen, die keine endogene MIA Expression aufweisen, untersucht. Die Analysen zeigten, dass MIA auch in diesen Zellen migrationsabhängig im hinteren Zellteil akkumuliert und sekretiert wird. Die Sekretion von MIA konnte zudem in transfizierten HEK Zellen, die ebenfalls keine endogene MIA Expression besitzen, nachgewiesen werden. Da bereits bekannt ist, dass MIA als Dimer funktionell aktiv ist, wurde des Weiteren untersucht, ob die Dimerisierung auch wichtig für den gerichteten Transport bzw. die Sekretion des Proteins ist. Die Analyse einer nicht-dimerisierungsfähigen MIA Mutante zeigte, dass die Dimerisierung für diese Prozesse nicht notwendig ist und die Sekretion im Vergleich zu Wildtyp MIA sogar erhöht ist. Um den Einfluss des Signalpeptids von MIA auf den gerichteten Transport und die Sekretion zu prüfen, wurde zudem eine MIA Mutante ohne Signalpeptid untersucht. Die Experimente konnten bestätigen, dass das Signalpeptid von MIA obligatorisch für die Sekretion des Proteins ist. Die Lokalisation von MIA scheint jedoch nicht zwingend durch das Signalpepitd definiert zu sein. So konnte auch MIA ohne Signalpeptid im hinteren Teil migrierender Zellen detektiert werden. Die Untersuchung eines chimären Proteins, bestehend aus dem Signalpeptid des membranständigen E Cadherins und dem maturen MIA Protein, zeigte darüber hinaus, dass MIA auch mit diesem Signalpeptid von transfizierten Zellen sekretiert wird. Zudem akkumuliert diese MIA Mutante ebenso wie Wildtyp MIA und MIA ohne Signalpeptid im hinteren Zellteil, was die Annahme stützt, dass die Information der intrazellulären Lokalisation von MIA nicht im Signalpeptid kodiert ist, sondern eventuell durch weitere Eigenschaften des maturen Proteins bestimmt wird.
In Western Blot Analysen von Proteinextrakten mit pCMX PL1 MIA transfizierten HeLa-, HEK und Mel Ei Zellen, konnte neben der Bande des maturen MIAs eine weitere Bande detektiert werden, die der Größe von MIA mit Signalpeptid entspricht. Um die Prozessierung des Signalpeptids und dessen Einfluss auf die Sekretion von MIA zu analysieren, wurden verschiedene MIA Signalpeptid Mutanten kloniert und getestet. Durch die Mutagenesestudie konnten zwei verschiedene Gruppen von Mutanten identifiziert werden. Die erste Gruppe wies das gleiche Bandenmuster wie Wildtyp MIA auf und wird von transfizierten HeLa und HEK Zellen sekretiert. Die zweite Gruppe wies ein verändertes Bandenmuster auf und wurde zudem nicht sekretiert. Beide Gruppen von MIA Mutanten wiesen zwar eine migrationsabhängige Akkumulation im hinteren Zellteil auf, unterschieden sich allerdings teilweise in ihrer relativen Verteilung in der Zelle von Wildtyp MIA. Zudem wurde in dieser Arbeit durch die Modulierung des intrazellulären Vesikeltransports mittels siRNA Technologie und Inhibitoren die Prozessierung des Signalpeptids weiter untersucht. Die Experimente mit transient transfizierten HeLa Zellen zeigten ein verändertes Bandenmuster von MIA in Western Blot Analysen, die darauf schließen lassen, dass das Signalpeptid nicht schon ko translational beim Eintritt in das Endoplasmatische Reticulum abgespalten wird, sondern die Prozessierung zu einem späteren Zeitpunkt erfolgen kann.

Summary
MIA (Melanoma Inhibitory Activity) is a very important protein, involved in melanoma progression and metastasis. In melanoma cells, MIA is expressed and migration-associated secreted at the cell rear. In the extracellular space MIA promotes the detachment of tumor cells from the ECM (Extracellular matrix) and thereby increases the migratory and invasive potential of melanoma cells.
This thesis analyzes the directed secretion of MIA and the processing of the signal peptide. Therefore, HeLa-cells without endogenous MIA expression were transiently transfected withpCMX-PL1-MIA and subsequently the localization and secretion of MIA were examined. The analysis revealed a migration-associated accumulation and secretion of MIA in the cell rear. The secretion was also shown in transfected HEK-cells, which do not express MIA, too. It is already known, that MIA is functionally active as a dimer. To elucidate if the dimerization is also important for transportation and secretion of the protein, a non-dimerizable mutant was investigated. The study showed that these processes are not dependent of the dimerization and that the secretion of monomeric MIA is increased compared to wild type MIA. Additionally, the impact of the signal peptide was analyzed using a MIA-mutant without signal peptide. We found, that this structure is mandatory for protein secretion. On the other side, the intracellular localization of MIA seems not necessarily to be determined by the signal peptide, as MIA without signal peptide could be detected migrations-associated at the cell rear, as well. Moreover, examination of a chimeric protein, consisting of the signal peptide from membrane-bound E-Cadherin and the mature part of MIA, revealed an ongoing secretion of the protein. Besides, the mutant still localizes at the rear of migration cells, emphasizing the idea that the intracellular transport of MIA is not encoded in the signal peptide but in additional structures of the mature protein.
Western blot analysis of protein extratcs from HeLa, HEK and Mel Ei cells, each transfected with pCMX-PL1-MIA, revealed the existence of protein band with the size of MIA and bound signal peptide. To analyze the processing and influence of the signal peptide on the secretion of MIA, several new mutants were cloned and tested. By mutagenesis two different groups of mutants were identified. The first group showed the same expression pattern and secretion like wild type MIA. The second group showed an altered pattern and was not secreted anymore. Both groups showed a migration-associated accumulation of MIA in the cell rear, but partly the relative distribution compared to wild type MIA changed. Furthermore, the processing of the signal peptide was investigated by modulation of the intracellular vesicle transport using RNAi-technology and inhibitors. The experiments with transiently transfected HeLa cells showed a changed band pattern of MIA in Western blot analysis. The findings indicate, that the signal peptide of MIA is not cleaved co-translationally but may occur at a later stage.
