Abstract (English)
In eukaryotic cells, transcription and processing of mRNAs in the nucleus are separated from translation in the cytoplasm by the nuclear membrane. Transport of mRNA from the nucleus to the cytoplasm is facilitated through the nuclear pore complex. The heteromeric THO/TREX complex plays an important role in the export of mRNA, by recruiting the mRNA export receptor that translocates the mRNA through the nuclear pore complex. The composi-tion and function of the TREX complex in Arabidopsis thaliana is not well described.In this work the four Arabidopsis orthologs ALY1, ALY2, ALY3 and ALY4 of the human TREX sub-unit ALYREF, were characterized. All four proteins were found to interact directly with anoth-er TREX subunit UAP56, as seen in humans. ALY3 and ALY4 were found to localize in both nucleoplasm and nucleolus of root cells, whereas ALY1 and ALY2 only localized in the nucle-oplasm. A double mutant of aly1-1 and aly2-1 showed no phenotypes. The double mutant of aly3-1 and aly4-1 showed an early flowering phenotype and a bulk accumulation of poly(A) mRNA in the nucleus, indicating partial redundancy.
A member of the THO subcomplex of TREX, TEX1 was characterized. A knockdown mu-tant tex1-4 exhibited multiple pleiotropic phenotypes, like early flowering, additionally activat-ed axillary meristems and less elongated lateral roots. The tex1-4 mutant did not accumulate poly(A) mRNA but showed decreased protein synthesis rates. TEX1 was shown to copurify with the rest of the THO complex, UAP56, ALY3 and multiple proteins involved in splicing and transcription. MOS11, an Arabidopsis ortholog of the TREX associated human CIP29 protein, was also characterized. MOS11 was shown to copurify UAP56, and to be copurified by UAP56. The mos11-2 mutant on its own showed a mild flowering phenotype and accumu-lation of poly(A) mRNA. When mos11-2 was crossed with tex1-4 additional pleiotropic pheno-types were observed. Trichomes, auxillary meristems, inflorescences and flower organs were especially affected. TEX1 was found to regulate tasiRNA independent of MOS11, whereas MOS11 was found to regulate miR173 independently of TEX1. In the double mutant a synergistic accumulation of poly(A) mRNA was seen, correlating with the observed in-crease in phenotype severity. Therefore, this study provides evidence that the general com-position of TREX is conserved and that ALY3, ALY4, TEX1 and MOS11 are involved in mRNA export.

In Eukaryonten ist die Transkription und mRNA-Prozessierung im Zellkern, durch die Kernmembran von der Translation im Zytoplasma abgetrennt. mRNA- Transport aus dem Zellkern heraus in das Zytoplasma hinein wird durch den Kernporenkomplex erleichtert. Der heterodimerische THO/TREX-Komplex spielt eine wichtige Rolle im Export von mRNA, indem er die mRNA-Export Rezeptoren rekrutiert, die die mRNA durch den Kernporenkomplex leiten. Die Beschaffenheit und die Funktion des TREX-Komplex in Arabidopsis thaliana ist wenig erforscht. In dieser Arbeit wurden die vier Arabidopsis Orthologe zu der menschlichen TREX Untereinheit ALYREF - ALY1, ALY2, ALY3 und ALY4 - charakterisiert. Alle vier Proteine interagierten, wie im Menschen, direkt mit UAP56, einer weiteren TREX-Untereinheit. ALY3 und ALY4 wurden sowohl im Nucleoplasma als auch im Nukleolus von Wurzelzellen detektiert, wohingegen ALY1 und ALY2 nur im Nucleoplasma gefunden wurden. Eine aly1-1 und aly2-1 Doppelmutante zeigte keine Phänotypen. Die Doppelmutante von aly3-1 und aly4-1 zeigte einen frühblühenden Phänotyp und eine Ansammlung von Poly(A)-mRNA im Zellkern, was eine gewisse Redundanz indiziert.
Eine Untereinheit des TREX Unterkomplexes THO, TEX1, wurde ebenfalls charakterisiert. Eine knockdown Mutante, tex1-4, zeigte mehrere pleiotrope Phänotypen, wie z.B. einen früheren Blühzeitpunkt, zusätzliche aktivierte axillare Meristeme und weniger lange seitliche Wurzeln. Die tex1-4 Mutante akkumulierte nicht Poly(A)-mRNA, aber zeigte eine verminderte Proteinsynthese. Es wurde gezeigt, dass TEX1 mit dem Rest der THO Komplex, UAP56, ALY3 und mehreren an Splicing und Transkription beteiligten Proteinen aufgereinigt wurde. MOS11, ein Arabidopsis ortholog des menschlichen CIP29, welches ebenfalls dem Trex-Komplex zugeordnet wird, wurde ebenso charakterisiert. MOS11 reinigte UAP56 auf und vice versa. Die mos11-2 Mutante zeigte einen leichten Blüh-Phänotyp und Akkumulation von Poly(A)-mRNA. Als mos11-2 mit Tex1-4 gekreuzt wurde zeigten sich zusätzlichen pleiotrope Phenotypen. Trichome, axilläre Meristeme, Blütenstände und Blüten Organe waren besonders betroffen. Es wurde gezeigt, dass TEX1 tasiRNA unabhängig von MOS11 reguliert; MOS11 hingegen reguliert miR173, unabhängig von TEX1. In der Doppel-Mutante wurde eine synergistische Anhäufung von Poly(A)-mRNA beobachtet. Dies korreliert mit dem beobachteten stärkeren Ausprägungen des Phänotyps. Diese Studie zeigt, dass die allgemeine Komposition von TREX konserviert ist und dass ALY3, ALY4, TEX1 und MOS11 am mRNA-Export beteiligt sind..
