Antimikrobielle Peptide sind wichtige Bestandteile des angeborenen Immunsystems. Neben ihrer antimikrobiellen Wirkung spielen sie eine wichtige Rolle bei der Modulation einer Immunantwort infolge einer Entzündungsreaktion. Diverse Studien zeigen dabei sowohl pro- als auch antiinflammatorische Eigenschaften der antimikrobiellen Peptide. In der vorliegenden Arbeit konnte gezeigt werden, dass das β-Defensin HBD2 indirekt mit dem P2X7R über die Freisetzung von ATP interagiert und dieser Signalweg zu einer synergistischen und proinflammatorischen Zytokinproduktion in Monozyten und Makrophagen nach TLR-Stimulation führt. Des Weiteren erfolgte im Rahmen dieser Arbeit die Generierung einer hReg3α transgenen Mauslinie. Dabei führte eine Überexpression des hReg3α zu einer Veränderung des Darmmikrobioms, die sich negativ auf die Erholungsfähigkeit nach einer akuten DSS-induzierten Kolitis auswirkte. Auch im untersuchten Tumormodell induzierte die Expression von hReg3α ein erhöhtes Tumorwachstum.

Antimicrobial peptides are important components of the innate immune system. Beside their antimicrobial functions they play an important role in modulating an inflammatory reaction. In this study we report that pre-stimulation of monocytes and macrophages with the β-defensin HBD2 results in a synergistic, enhanced expression of pro-inflammatory cytokines induced by TLR ligand re-stimulation. Thereby, HBD2 indirectly interacts with the P2X7R by the release of ATP.
In addition we generated a hReg3α transgenic mouse line. The overexpression of hReg3α resulted in an altered gut microbiota composition, which negatively affected the regeneration after a DSS-induced colitis. The expression of hReg3α induced an enhanced tumor growth in the examined tumor model.
