Abstract (German)

Verschiedene Amin-substituierte α-Methylen-γ-Butyrolactone wurden durch Michael-Addition an die Doppelbindung des Lactons synthetisiert. Als Amine dienten sowohl einfache Amine (Diethylamin, Propylamin), als auch die Seitenkette eines an der Hauptkette Cbz-geschützten Lysins.
Das Eliminierungsverhalten der Amine in einer Retro-Michael-Addition unter verschiedenen Bedingungen in wässrigem Medium wurde untersucht. So stellte sich die Substitution an der Position 5 des Lactonrings als notwendig heraus, um Nebenreaktionen im Zuge der Eliminierung, wie eine Ringöffnung, zu verhindern.

Der Einfluss des Substitutionsmusters der Amine sowie des pH-Wertes der Pufferlösung auf das Eliminierungsverhalten wurde untersucht.
Wurde das exo-Methylen-Lacton an eine Lysinseitenkette von Cbz-Lys-OH gebunden, so stellte sich eines am Seitenkettenstickstoff methyl-substituiertes Derivat als das geeignetste System heraus, um an ein Carrierpeptid gebunden zu werden.

Des Weiteren wurde die Cytotoxizität der verwendeten Amin-Addukte mit denen der nackten Lactone verglichen und die Eignung als Prodrug und Kandidat für die Anbindung an ein Carrierpeptid bestätigt.

Die exemplarische Anbindung dieser Addukte in einer Festphasenpeptidsynthese an ein Glycin als letztem Schritt der Beladung eines CPP (an den N-Terminus) wurde durchgeführt.

Translation of the abstract (English)

Different amine-substituted α-Methylene-γ-Butyrolactones were synthesized by Michael-addition to the double bond of the lactone. As amines served simple amines (diethyl amine, propyl amine) as well as the side chain of a lysine which was cbz-protected at the main chain amino group.
The elimination behavior of the amines in a retro-michael-addition under different conditions under aqueous conditions was investigated. Substitution on the 5-position of the lactone ring proved to be necessary in order to prevent side reactions such as ring opening.

The influence of the substitution pattern of the amines as well as the pH-value of the buffer solution on the elimination behavior was investigated.
When the exo-methylene-lactone was bound to the side chain of Cbz-Lysine-OH, it turned out that a derivative which was methylated at the side chain amine was best suited for being bound to a carrier peptide.

Furthermore, the cytotoxicity of the amino-adducts used was compared with these of the naked lactones and the applicability as prodrug and candidate for binding to a carrier peptide was confirmed.

Binding of the derivative to glycine in a solid phase peptide synthesis as the last step of loading onto the N-terminus of a carrier peptide was carried out.
