Vector-borne diseases transmitted by mosquitoes are a major cause of morbidity and mortality in the world by transmitting infections such as malaria, dengue, yellow fever, West Nile fever or chikungunya, to more than 700 million people each year. Without existing vaccines for each disease, skin repellents are one of the most effective defense against mosquitoes for an individual protection. N,N-diethyl-m-methylbenzamide (DEET) is an efficient and commonly used active since decades. New molecules have appeared on the market and proved their repellent action mostly comparable to DEET in regards to different vectors such as para-Menthane-3,8-diol (PMD). This molecule has interesting advantages for repellent products such as a high repellent activity against mosquitoes, a potential natural origin and a low toxicity. The aim of this project is to investigate and to improve repellent products based on para-Menthane-3,8-diol.
A first part of this thesis focused on the study of PMD, from its synthesis to its introduction into new formulation systems allowing to improve the repellent action against mosquitoes. A green and simple method to produce PMD from the essential oil of Eucalyptus citriodora is reported in Chapter 2. The repellent activity of the synthesized active is investigated against Aedes aegypti mosquitoes and shows a high repellent action on Human volunteers. The majority of repellent products on the market is composed of mixed surfactant systems or hydro-alcoholic bases, which present several drawbacks for the consumers (skin irritation, reducing time of protection, unpleasant odor or feeling). In Chapter 3 surfactantless and classical microemulsions containing PMD were studied in order to extend the lasting protection against mosquitoes. A second study in Chapter 4 involved the development of an oil in water PMD repellent lotion with environmentally acceptable additives. The main goal was to reach a high protection activity comparable to the reference product on the market, the Autan® Protection Plus from SC Johnson composed of KBR 3023. Finally, the complexation of PMD into cyclodextrins (CDs) was investigated in Chapter 5 in order to solubilize PMD in water and enable its controlled release over time.
In a second part, a new water-soluble repellent active derived from PMD was considered. The low water solubility of PMD restrains the development of simple repellent formulations and the synthesis of the active so called PMD-Succinate (PMD-S) is reported in Chapter 6. In order to determine the nature of the new active, especially its surface activity as a surfactant or a hydrotrope, physical and chemical properties were studied in Chapter 7. This study is completed in Chapter 8 by the evaluation of the cytotoxicity of PMD-S on HeLa and Keratinocyte cells as well as those of all the commonly repellent actives used in the market. Finally, an improved synthesis method of PMD-S was investigated with green considerations and an acceptable yield in Chapter 9.
In a last part (Chapter 10) and independently of the work on PMD and its derivative, a study of 19 Corsican (French island) essential oils was carried out in order to determine their protection activity against mosquitoes as well as their possible integration into repellent products.

Vektorübertragene Krankheiten, die durch Moskitos verbreitet werden, sind eine der Hauptursachen für Morbidität und Mortalität in der Welt, indem sie Infektionen wie Malaria, Denguefieber, Gelbfieber, West-Nil-Fieber oder Chikungunya bei mehr als 700 Millionen Menschen pro Jahr auslösen. Ohne Impfstoffe für diese Krankheiten sind wirksame Hautabwehrmittel eine der effizientesten Maßnahmen gegen Moskitos und für einen individuellen Schutz. N,N-diethyl-m-methylbenzamide (DEET) ist ein seit Jahrzehnten wirksamer und weit verbreiteter Wirkstoff. Inzwischen gibt es neue Moleküle, die in Bezug auf ihre abstoßende Wirkung mit DEET vergleichbar sind, wie para-Menthan-3,8-diol (PMD). Dieses Molekül hat neben seiner hohe Repellent-Wirkung gegen Mücken weitere interessante Vorteile, wie zum Beispiel einen potentiellen natürlichen Ursprung und eine geringe Toxizität. Ziel der vorliegenden Arbeit ist es deshalb, Repellentprodukte mit para-Menthan-3,8-diol zu untersuchen und zu verbessern.
Der erste Teil dieser Arbeit beschäftigt sich mit der Untersuchung von PMD, von der Synthese bis zur Einführung in neue Formulierungssysteme, die es erlauben, die abstoßende Wirkung gegen Moskitos zu verbessern. Eine grüne und einfache Methode zur Herstellung von PMD aus dem ätherischen Öl von Eukalyptus citriodora wird in Kapitel 2 beschrieben. Die abstoßende Aktivität des synthetisierten Wirkstoffs wird gegen Aedes aegypti-Mücken untersucht und zeigt eine hohe Effizienz bei menschlichen Probanden. Die Mehrzahl der auf dem Markt erhältlichen Repellentprodukte besteht aus gemischten Tensidsystemen oder hydroalkoholischen Basen, die für den Verbraucher mehrere Nachteile mit sich bringen (Hautreizung, Verkürzung der Schutzzeit, unangenehmer Geruch oder Gefühl). In Kapitel 3 werden tensidfreie und klassische PMD-haltige Mikroemulsionen untersucht, um den dauerhaften Schutz vor Mücken zu erweitern. Eine weitere Studie in Kapitel 4 befasst sich mit der Entwicklung einer Öl-in-Wasser-Lotion von abweisendem PMD mit umweltverträglichen Zusätzen. Das Hauptziel war es, eine hohe Schutzwirkung zu erreichen, vergleichbar mit dem Referenzprodukt auf dem Markt, dem Autan® Protection Plus von SC Johnson, KBR 3023 als Aktivsubstanz enthaltend. Schließlich wurde die Komplexierung von PMD in Cyclodextrine (CDs) in Kapitel 5 untersucht, um PMD in Wasser zu lösen und seine kontrollierte Freisetzung über die Zeit zu ermöglichen.
In einem zweiten Teil wurde ein neuer wasserlöslicher Repellent-Wirkstoff aus PMD in Betracht gezogen. Die geringe Wasserlöslichkeit von PMD hemmt die Entwicklung einfacher Repellent-Formulierungen. Die Synthese des aktiven und wasserlöslichen, so genannten PMD-Succinat (PMD-S) wird in Kapitel 6 beschrieben. Um die Art des neuen Wirkstoffs, insbesondere seine Oberflächenaktivität als Tensid oder Hydrotrop, zu bestimmen, wurden in Kapitel 7 physikalische und chemische Eigenschaften untersucht. Diese Studie wird in Kapitel 8 durch die Evaluierung der Zytotoxizität von PMD-S auf HeLa- und Keratinozytenzellen sowie der aller marktüblichen Repellent-Wirkstoffen vervollständigt. Schließlich wurde eine verbesserte Synthesemethode von PMD-S mit grünen Überlegungen und einer akzeptablen Ausbeute untersucht (Kapitel 9).
In einem letzten Teil (Kapitel 10) und unabhängig von der Arbeit an PMD und seinen Derivaten wurde eine Studie mit 19 ätherischen Ölen aus Korsika (französische Insel) durchgeführt, um ihre Schutzwirkung gegen Mücken sowie ihre mögliche Integration in Repellentprodukte zu bestimmen.
