Abstract (German)

In dieser Arbeit konnte eine relative Nebenniereninsuffizienz an durch Kollagen II immunisierten und arthritischen Ratten gezeigt werden. Diese konnten hierbei trotz erhöhtem relativen Nebennierengewicht keine höheren Plasmacorticosteronkonzentrationen als gesunde Tiere aufbauen. In dieser Arbeit konnte eine relative Nebenniereninsuffizienz an durch Kollagen II immunisierten und arthritischen Ratten gezeigt werden. Diese konnten hierbei trotz erhöhtem relativen Nebennierengewicht keine höheren Plasmacorticosteronkonzentrationen als gesunde Tiere aufbauen. In vitro zeigten die Nebennieren im Gewebeverbund einen zeitabhängigen Effekt auf IL-1β-Stimulation, während ACTH keine Corticosteronsekretion induzieren konnte. So konnte IL-1β nur anfangs (Tag 5) die Corticosteronsekretion bei CIA-Tieren stimulieren. IL-1β vermochte ebenso erst zu späteren Versuchszeitpunkten (Tag 55) den Sauerstoffverbrauch der Nebennieren zu senken. Es wurde nachgewiesen, dass ACTH- und IL-1β-Inkubation die Nebennieren zu vermehrter Zytokinproduktion veranlasst. Hier fiel v.a. eine starke Zunahme der Proteinexpression von CINC-1 und CINC-3 unter IL-1β-Stimulation sowohl bei CIA- als auch bei Kontrolltieren auf. Auf ultrastruktureller Ebene zeigte sich während der Hochphase der Arthritis an Tag 28 eine Verhältnisumkehr von sER zu Zytoplasma zu Gunsten der CIA-Tiere.

Translation of the abstract (English)

To further investigate the relative adrenal dysfunction in experimental arthritis, corticosteron concentration in the plasma, cytokine secretion and oxygen consumption of the adrenal gland was compared between control animals and animals with collagen II-induced arthritis (CIA). To further investigate the relative adrenal dysfunction in experimental arthritis, corticosteron concentration in the plasma, cytokine secretion and oxygen consumption of the adrenal gland was compared between control animals and animals with collagen II-induced arthritis (CIA). Also a quantitative analysis of cell compartments (smooth endoplasmic reticulum and mitochondria) in arthritic and control animals was realized by electron microscopic images of the adrenal gland.
In this paper the relativ adrenal insufficiency in collagen II-induced arthritic rats could be reconfirmed. The CIA rats could not - despite augmented relative adrenal weights – establish a higher secretion of corticosteron in the plasma than healthy control animals. In vitro incubation of the adrenal glands with ACTH (adrenocorticotropic hormone) could also not augment corticosteron secretion (widely consistent with former investigations). However interleukin 1 beta (IL-1β) could stimulate corticosteron secretion at day 5 of the experiment and inhibit the oxygen consumption of the adrenal glands at day 55 of the experiment.
Incubation of the adrenal glands of both control animals and CIA animals with IL-1β results in an enormous expression of the protein CINC (cytokine-induced neutrophile chemoattractant) 1 and 3. These cytokines could be at least partially responsible for an altered and insufficient response to elevated systemic cytokine levels while chronic inflammation in rheumatoid arthritis.
The relation of smooth endoplasmic reticulum / cytoplasm shows a change from higher levels of this ratio in control animals (day 1 and day 5) to higher levels in arthritic animals in day 28 (most intense inflammation) after inducing arthritis. This suggests that enzymatic steps of the steroidogenesis might be blocked with a consecutive congestion of corticosteron precursors and thus a dilation of smooth endoplasmic reticulum and mitochondria (where most of the steps of the steroidogenesis take place).
