Abstract (English)
Cyclooxygenase 2 is a key enzyme in the production of prostaglandins. The widespread application of Cox-2 selective inhibitors to treat pain and inflammation has shown severe renal side effects for these drugs. But the exact expression sites for Cox-2 and its normal function in the kidney is not yet clear.
Utilizing a novel in-situ hybridization technique Cox-2 could be distinctly located in two developmentally different cell populations of the murine kidney. In the cortex only specialized macula densa cells of the cTAL express Cox-2. In the renal medulla, interstitial fibroblast-like cells, characterized by the expression of PDGFR-β and TNC, produce Cox-2 mRNA.
Stimulating Cox-2 expression in the kidneys of mice, through a high dietary sodium intake, Cox-2 expression was induced only in the inner medulla and the inner stripe of the outer medulla. This recruitment of Cox-2 expressing cells only took place in interstitial PDGFR-β+/TNC+ cells. In mice deprived of water for 24h the recruitment pattern and cell type was the same, but the total number of Cox-2 expressing cells was on average lower than in high salt treated mice. The model of genetic stabilization of the HIF pathway in PDGFR-β+/iCre Vhlfl/fl mice not only showed that Cox-2 is a hypoxia inducible gene, but that an overlap between Cox-2 and EPO expression in the same cell is possible. The locally very restricted inducible expression of medullary interstitial Cox-2 led me to classify these cells as a functional subpopulation of the large pool of PDGFR-β+ interstitial cells. No Cox-2 expression could be detected in non-interstitial cells or in interstitial cells of the cortex in the conditions investigated in the scope of this work.
The medullary Cox-2+ cells, as well as most cells involved in the severe phenotype of global Cox-2 deficient mice, namely mesangial cells and vascular smooth muscle cells derive from the FoxD1+ stromal progenitor compartment. The role of Cox-2 during nephrogenesis was evaluated with a selective deletion in this compartment with FoxD1+/Cre Cox2fl/fl mice. The mice with a targeted Cox-2 deletion in the stromal compartment, but intact Cox-2 function in the cells of the macula densa, showed no developmental or functional defects. In FoxD1+/Cre Cox2fl/fl mice the recruitment of renin on a low salt stimulus was normal unlike in the mice with a total deletion of Cox-2.
In adult mice, the strong medullary induction of Cox-2 with a high dietary sodium intake implicates a functional connection between Cox-2 and the salt handling capability of the kidney. The lack of an adequate model made the distinction between the functions of the different Cox-2 expression sites difficult. With the use of PDGFR-β+/iCre Cox2fl/fl mice, it was possible to study the functional consequences of the isolated deletion of Cox-2 only in medullary interstitial cells under different conditions. This model showed that missing Cox-2 expression in the renal medulla led to reduced concentrating ability for sodium with a compensatory rise in urine volume and persistent hypertension under a high salt diet.
Experiments performed, concerning the role of Cox-2 in PDGFR-β+ cells during fibrosis, showed a dependence on the fibrotic model used. In the adenine-induced nephropathy Cox-2 seems to play some role in the induction of Col1a1 mRNA expression. But a deletion of Cox-2 in PDGFR-β+ cells had no effect on the expression of fibrotic markers in the UUO model.
The characterization of markers for Cox-2 expressing cells in the kidney is the basis for a targeted deletion in sub-compartments and their functional investigation. While Cox-2 expression in medullary interstitial cells is not essential for normal kidney development, it is necessary for blood pressure homeostasis during a high salt challenge.

Cyclooxygenase-2 (Cox-2) ist das Schlüsselenzym in der Produktion von Prostaglandinen. Die weitreichende Anwendung von Cox-2 selektiven Inhibitoren zur Behandlung von Schmerzen und Entzündungen hat schwere renale Nebenwirkungen aufgezeigt. Jedoch sind die genauen Expressionsorte für Cox-2 und deren basale Funktion in der Niere noch nicht eindeutig geklärt.
Unter Verwendung einer neuen Methode der in-situ Hybridisierung konnte Cox-2 eindeutig in zwei Zellpopulationen der Mausniere lokalisiert werden. Im Cortex der Niere wird Cox-2 ausschließlich von Zellen der Macula densa exprimiert und im Nierenmark wird es nur von PDGFR-β+ und TNC+ interstitiellen Zellen exprimiert.
Durch eine Stimulation der Cox-2 Expression mit Hilfe einer Hochsalzdiät konnte gezeigt werden, dass neu rekrutierte Cox-2 exprimierende Zellen ausschließlich PDGFR-β+/TNC+ Zellen der inneren und äußeren Medulla waren. Bei Mäusen mit eingeschränktem Zugang zu Trinkwasser war der Zelltyp der rekrutierten Cox-2 exprimierenden Zellen identisch, aber die Zahl der rekrutierten Zellen war geringer.
Durch eine genetische Stabilisation des HIF-Signalweges konnte gezeigt werden, dass Cox-2 ein Hypoxie-induzierbares Gen ist. In den interstitiellen Zellen des Cortex konnte keine Cox-2 Expression detektiert werden.
Eine globale Deletion des Cox-2 Gens zeigt in Mäusen einen schweren renalen Phänotyp, der viele Abkömmlinge der FoxD1 positiven Stromavorläuferzellen betrifft. Die Rolle der Cox-2 in diesem Kompartiment während der Nephrogenese wurde mithilfe einer zellspezifischen Deletion von Cox-2 (FoxD1+/Cre Cox2fl/fl) in Mäusen untersucht. In diesen FoxD1+/Cre Cox2fl/fl Mäusen konnte kein Phänotyp festgestellt werden. Durch die intakte Expression der Cox-2 in den Zellen der Macula densa in diesem Mausmodell war die Rekrutierung von Reninzellen während eines Niedrigsalz-Stimulus unverändert.
Durch eine zellspezifische Deletion von Cox-2 in den PDGFR-β+ interstitiellen Zellen der Nieren (PDGFR-β+/iCre Cox2fl/fl) von adulten Mäuse wurde die Funktion der medullären Cox-2 während einer Hochsalzdiät untersucht. In diesen Mäusen konnte eine verminderte Konzentration von Natrium im Urin festgestellt werden. Durch einen Anstieg des Urinvolumens und des Blutdrucks wurde die Ausscheidung von Natrium aber konstant gehalten.
In Experimenten zur Funktion von Cox-2 in der Nierenfibrose konnte eine Abhängigkeit vom verwendeten Modell festgestellt werden. In dem Modell der Adenin-induzierten Fibrose konnte bei Deletion von Cox-2 in PDGFR-β+/iCre Cox2fl/fl Mäusen eine verminderte mRNA Expression von Kollagen1a1 gemessen werden. Hingegen war der Verlauf der Fibrose im Modell der unilateralen Ureterokklusion bei PDGFR-β+/iCre Cox2fl/fl Mäusen unverändert.
Durch die eindeutige Lokalisation von Cox-2 und die Identifikation von Zellmarkern konnte eine spezifische Deletion durchgeführt werden, um nur das Kompartiment der medullären Cox-2 Expression zu untersuchen. Eine intakte Funktion der Cox-2 Expression in den medullären Zellen der Niere ist für die basale Funktion der Niere nicht notwendig, aber sie ist essentiell für die Homöostase des Blutdrucks bei erhöhter Natriumzufuhr über die Nahrung.
