DIRAS3 ist ein Protein aus der ras-Familie, dessen genaue Funktion bis dato unbekannt ist. Die Transkription unterliegt einem maternalen Imprinting Mechanismus, der in Zusammenspiel mit der in Gliomen, und insbesondere Oligodendrogliomen, häufigen 1p/19q-Kodeletion zu einer biallelischen Inaktivierung des DIRAS3-Gens führt. Die 1p/19q-Kodeletion ist in Gliomen ein positiver prognostischer und prädiktiver Marker für das Ansprechen auf PCV-Chemotherapie (Procarbazin, Lomustin, Vincristin). Frühere Untersuchungen, wie auch eine Metaanalyse von Expressionsdaten im Rahmen dieser Arbeit, konnten zeigen, dass auch eine niedrige DIRAS3-Expression in Gliomen mit einem besseren Gesamtüberleben der Gliompatienten einhergeht.
In dieser Arbeit wurden die funktionellen Effekte der DIRAS3-Expression anhand eines stabil transfizierten shRNA-DIRAS3-knockdown Modells in den Gliomzelllinien U251 und HS683 untersucht. Dabei wurden Migrations- und Invasionsverhalten, Proliferations- und Apoptoseverhalten sowie Chemotherapieresistenz gegenüber den Chemotherapeutika Temozolomid, Procarbazin, Lomustin und Vincristin der shRNA Klone in funktionellen in vitro Assays getestet. Weiter wurde über einen Antikörperarray ein Phosphoproteinprofil der shRNA-Klone erstellt und es erfolgte eine statische Ploidiebestimmung zur Beurteilung des Zellzyklus. Hierbei zeigte sich unter knockdown-Bedingungen insgesamt eine erniedrigte Zellviabilität mit reduzierter Migrationsfähigkeit und verstärkter Apoptose sowie eine geringere Resilienz gegenüber Lomustin. Weiter ergaben sich Hinweise auf eine stoffwechselabhängige Funktion von DIRAS3 mit der Regulation des Stoffwechselschalters AMPKα1/2 und eine zellzyklusregulatorische Funktion von DIRAS3 über p53.
Der nach der Expressionsdatenanalyse zu vermutende onkogene Effekt von DIRAS3 in Gliomen wird somit durch die funktionellen Testergebnisse untermauert, wobei der genaue Wirkmechanismus von DIRAS3 und seiner Interaktionspartner auf Proteinebene noch weitergehend untersucht werden muss.

The DIRAS3 protein is a member of the ras-family. To date the exact function of DIRAS3 remains unknown. The genomic sequence is located in the chromosomal region 1p31.3. Due to an imprinting mechanism, gene expression from the maternal allele is silenced by CPG island methylation. In gliomas and particularly in oligodendrogliomas 1p/19q codeletion occurs frequently, causing a biallelic inactivation of DIRAS3. 1p/19q-codeletion is a positive prognostic marker for overall survival (OS) as well as a predictive marker for therapy-response to PCV-treatment (procarbazine, lomustine, vincristine). Biallelic inactivation and therefore low expression of DIRAS3 has been associated with a better OS in glioma previously and could be confirmed in a metaanalysis of DIRAS3 expression data in various glioma datasets in this work.
This work investigated the functional effects of DIRAS3 expression using stably transfected shRNA-DIRAS3-knockdown clones of the glioma cell lines U251 and HS683. Tumor cell migration, invasion as well as proliferation and apoptosis were tested by execution of functional in vitro assays. Furthermore, we used proteome profiling arrays to investigate the phosphorylation profile of the shRNA clones and performed FACS-based static ploidy measurements for cell cycle assessment. Taken together, the functional assays indicated a reduced cell-viability marked by a lowered migration capacity, increased rate of apoptosis and an improved chemoresponse to lomustine treatment. Furthermore, we found evidence for a regulatory function of DIRAS3 on cell-metabolism via AMPKα1/2 as well as on cell cycle control via p53.
In summary, these data corroborate an oncogenic function of DIRAS3 in gliomas, as indicated by the DIRAS3 transcriptional analyses.
