In dieser Arbeit wurden die Expression von Semaphorin 3A (Sema3A) und seines Rezeptors Neuropilin-1 (NRP1) sowie deren Einfluss auf artikuläre Chondrozyten bei Gonarthrose untersucht. Sema3A wie auch NRP1 werden in Knieknorpel von Arthrosepatienten im Vergleich zu Knieknorpel von Probanden ohne Arthrose verstärkt exprimiert. So weisen OA-Knorpelzellen verglichen mit Nicht-OA-Knorpelzellen erhöhte Level an NRP1- und Sema3A-mRNA sowie eine signifikant vermehrte Proteinbiosynthese von Sema3A auf. Sema3A wird im Gegensatz zu NRP1 mit zunehmender Passage der OA-Chondrozyten weniger exprimiert. Zudem zeigen OA-Chondrozyten nach Kultivierung im 3D-System im Vergleich zum 2D-System eine verringerte Expression von Sema3A und NRP1, bei Nicht-OA-Chondrozyten hat das Kultursystem keinen derartigen Einfluss. Sema3A könnte daher im Zusammenhang mit Dedifferenzierungsprozessen vermehrt exprimiert werden.
Nach dem transienten Knockdown von NRP1 zeigte sich, dass NRP1 keinen Einfluss auf das Apoptoseverhalten von OA-Chondrozyten hat, jedoch katabole Stoffwechselprozesse fördert. So stimuliert NRP1 die Expression von MMP13. Dadurch werden Vorgänge wie der Abbau der extrazellulären Matrix, im Speziellen des Kollagennetzwerks, die insbesondere über MMP13 vermittelt werden, unterhalten. Bei MMP13 und VEGFA handelt es sich zudem um Hypertrophiemarker, deren vermehrte Expression charakteristisch für das Initialstadium von Arthrose ist und dessen Entstehungsprozess fördert und unterhält.
Unsere Versuche beweisen des Weiteren, dass NRP1 die Adhäsionsfähigkeit von OA-Chondrozyten, möglicherweise über eine verringerte Expression von Integrinen, herabsetzt und die Proliferation der Chondrozyten anregt, wodurch es in arthrotisch verändertem Knorpel eine wichtige Rolle beim chondrocyte cloning einnehmen könnte. NRP1 wird im OA-Knorpel von Liganden aktiviert, welche insgesamt eine vermehrte MMP13-Expression induzieren. Eine Blockade von NRP1 in arthrotischem Knorpel würde zu einer Verminderung der Proliferation und möglicherweise auch der Migration führen und dadurch dem als chondrocyte cloning bezeichneten Versuch des Gelenkknorpels, geschädigte Areale zu reparieren, entgegenwirken. Zum anderen könnte eine solche Inhibition aber, insbesondere durch die verringerte Expression von MMP13, die katabolen Stoffwechselmechanismen eingrenzen, welche zum Abbau der Knorpelmatrix führen, und das Fortschreiten von Arthrose verhindern.

In this work, the expression of semaphorin 3A (Sema3A) and its receptor neuropilin-1 (NRP1) and their influence on articular chondrocytes in gonarthrosis were investigated. Sema3A as well as NRP1 are increasingly expressed in knee cartilage of osteoarthritis patients compared to knee cartilage of persons without osteoarthritis. Thus, OA cartilage cells show increased levels of NRP1 and Sema3A mRNA as well as a significantly increased protein biosynthesis of Sema3A compared to non-OA cartilage cells. In contrast to NRP1, Sema3A is expressed less with increasing passage of OA chondrocytes. In addition, after cultivation in the 3D system, OA chondrocytes show a reduced expression of Sema3A and NRP1 compared to the 2D system; in non-OA chondrocytes the culture system has no such influence. Sema3A could therefore be expressed more in connection with dedifferentiation processes.
After the transient knockdown of NRP1, it was shown that NRP1 has no influence on the apoptosis rate of OA chondrocytes, but promotes catabolic metabolism. Thus, NRP1 stimulates the expression of MMP13. This maintains processes such as the degradation of the extracellular matrix, in particular the collagen network, which are mediated in particular by MMP13. MMP13 and VEGFA are also hypertrophy markers whose increased expression is characteristic of the initial stage of osteoarthritis and promotes and maintains its development.
Our experiments further demonstrate that NRP1 reduces the adhesion capacity of OA chondrocytes, possibly via a reduced expression of integrins, and stimulates chondrocyte proliferation, which could enable it to play an important role in chondrocyte cloning in arthritic cartilage. In OA cartilage, NRP1 is activated by ligands that induce an increased expression of MMP13. Blocking NRP1 in arthritic cartilage would lead to a reduction in proliferation and possibly also migration, thereby counteracting the attempt of articular cartilage to repair damaged areas, which is known as chondrocyte cloning. On the other hand, however, such inhibition, especially through the reduced expression of MMP13, could limit the catabolic mechanisms that lead to the degradation of the cartilage matrix and prevent the progression of osteoarthritis.
