The acid-sensing ion channels ASIC1 and ASIC2, as well as the transient receptor potential vanilloid channels TRPV1 and TRPV4, are proton-gated cation channels that can be activated by low extracellular pH (pHe), which is a hallmark of the tumor microenvironment in solid tumors. However, the role of these channels in the development of skin tumors is still unclear. In this study, we investigated the expression profiles of ASIC1, ASIC2, TRPV1 and TRPV4 in malignant mela- noma (MM), squamous cell carcinoma (SCC), basal cell carcinoma (BCC) and in nevus cell nevi (NCN). We conducted immunohistochemistry using paraffin-embedded tissue samples from pa- tients and found that most skin tumors express ASIC1/2 and TRPV1/4. Striking results were that BCCs are often negative for ASIC2, while nearly all SCCs express this marker. Epidermal MM some- times seem to lack ASIC1 in contrast to NCN. Dermal portions of MM show strong expression of TRPV1 more frequently than dermal NCN portions. Some NCN show a decreasing ASIC1/2 expres- sion in deeper dermal tumor tissue, while MM seem to not lose ASIC1/2 in deeper dermal portions. ASIC1, ASIC2, TRPV1 and TRPV4 in skin tumors might be involved in tumor progression, thus being potential diagnostic and therapeutic targets.

Die Protonen-sensitiven Ionenkanäle ASIC1 und ASIC2 sowie die Transient-Receptor-Potential-Vanilloid-Kanäle TRPV1 und TRPV4 sind protonengesteuerte Kationenkanäle, die durch einen niedrigen extrazellulären pH-Wert (pHe) aktiviert werden können, der ein Kennzeichen des Tumormikromilieus in soliden Tumoren ist. Die Rolle dieser Kanäle bei der Entwicklung von Hauttumoren ist jedoch noch unklar. In dieser Studie untersuchten wir die Expressionsprofile von ASIC1, ASIC2, TRPV1 und TRPV4 in malignen Melanomen (MM), Plattenepithelkarzinomen (SCC), Basalzellkarzinomen (BCC) und in Nävuszellnävi (NCN). Wir führten immunhistochemische Färbungen durch und stellten fest, dass die meisten Hauttumore ASIC1/2 und TRPV1/4 exprimieren. Wesentliche Ergebnisse unserer Studie sind, dass BCCs oft keine Expression von ASIC2 aufweisen, allerdings fast alle SCCs. Während ASIC1 in epidermalen Anteilen des MM teilweise nicht exprimiert wird, weisen im Gegensatz dazu die epidermalen Anteile des NCN eine positive Expression auf. Dermale Gewebe des MM zeigen häufiger eine starke Expression von TRPV1 als dermale NCN-Gewebe. In einigen NCNs nimmt die ASIC1/2-Expression mit zunehmender Tiefe im dermalen Tumorgewebe ab, während MMs in tieferen dermalen Anteilen ASIC1/2 weiterhin gleichmäßig exprimieren. ASIC1, ASIC2, TRPV1 und TRPV4 in Hauttumoren könnten an der Tumorprogression beteiligt sein und somit potenzielle diagnostische und therapeutische Ziele darstellen.
