In verschiedenen Tiermodellen konnte bereits die Beteiligung des Zytokins Interleukin-3 in inflammatorischen Autoimmunerkrankungen belegt werden. In verschiedenen Tiermodellen konnte bereits die Beteiligung des Zytokins Interleukin-3 in inflammatorischen Autoimmunerkrankungen belegt werden. Die Daten zur Rolle des Zytokins in humanen Autoimmungeschehen sind jedoch bisher spärlich. Daher beschäftigt sich diese Arbeit mit der Expression von IL-3 und seinem Rezeptor in Patienten mit Systemischem Lupus erythematodes, Psoriasis-Arthritis oder Granulomatose mit Polyangiitis. Es konnte eine signifikant höhere Ausschüttung des Zytokins in erkrankten Personen im Vergleich zu gesunden Kontrollprobanden beobachtet werden, die überdies eine positive Korrelation mit der Krankheitsaktivität aufwies. Ferner wurden IL-3 produzierende Zellen näher untersucht, wobei CD8+ T-Zellen als Hauptproduzenten identifiziert werden konnten. Diese Untersuchungen wurden zum Vergleich auch mit weiteren Zytokinen (IFNy, IL-2, IL-17, GM-CSF) durchgeführt. Ferner konnte eine deutliche Herabregulierung des IL-3 Rezeptors auf Basophilen Granulozyten und plasmazytoiden Dendritischen Zellen als Reaktion auf die erhöhte basale IL-3 Freisetzung durch T-Zellen gezeigt werden. Die Ergebnisse dieser Arbeit belegen die relevante Rolle von IL-3 im Krankheitsprogress der genannten Erkrankungen. Die Blockierung von IL-3 oder seinem Rezeptor stellt somit einen möglichen neuen Ansatzpunkt für die Therapie dieser rheumatologischen Erkrankungen dar.

The involvement of the cytokine interleukin-3 in inflammatory autoimmune diseases has already been demonstrated in various animal models. However, data on the role of the cytokine in human autoimmune events have been sparse. Therefore, this work deals with the expression of IL-3 and its receptor in patients with systemic lupus erythematosus, psoriatic arthritis, or granulomatosis with polyangiitis. A significantly higher release of the cytokine was observed in diseased subjects compared to healthy control subjects, which moreover showed a positive correlation with disease activity. Furthermore, IL-3 producing cells were investigated in more detail, with CD8+ T cells being identified as the main producers. These studies were also performed with other cytokines (IFNy, IL-2, IL-17, GM-CSF) for comparison. Furthermore, a significant down-regulation of the IL-3 receptor on basophil granulocytes and plasmacytoid dendritic cells in response to increased basal IL-3 release by T cells was demonstrated. The results of this work demonstrate the relevant role of IL-3 in the disease progression of the above diseases. Thus, blocking IL-3 or its receptor represents a potential new target for the therapy of these rheumatologic diseases.
