Abstract (German)

Die Zielsetzung dieser Promotionsarbeit lag in der dezidierten In-Vitro-Erforschung der Direktwirkung der Antibiotika Meropenem, Cefuroxim, Moxifloxacin, Clarithromycin sowie dem Glukokortikoid Prednisolon auf das Zusammenspiel der Abwehrmechanismen neutrophiler Granulozyten. Die Zielsetzung dieser Promotionsarbeit lag in der dezidierten In-Vitro-Erforschung der Direktwirkung der Antibiotika Meropenem, Cefuroxim, Moxifloxacin, Clarithromycin sowie dem Glukokortikoid Prednisolon auf das Zusammenspiel der Abwehrmechanismen neutrophiler Granulozyten. Differierende Studien zeigten ein zum Teil widersprüchliches, ambivalentes Wirkspektrum der zu untersuchenden Medikamente auf die körpereigene Immunabwehr. Zum einen scheinbar eine Förderung sowie Verbesserung der Granulozytenaktivität, zum anderen aber auch einen Abfall der Neutrophilenzahl (Neutropenie). So ergab sich die explizite Forschungshypothese, ob neutrophile Granulozyten durch bestimmte Antibiotikaklassen bzw. das Glukokortikoid Prednisolon in ihrer Wirkweise in vitro inhibiert werden. Deshalb wurden Migrationslänge, Migrationsgeschwindigkeit, die Produktion von reaktiven Sauerstoffspezies sowie die Freisetzung des Enzyms Myeloperoxidase und von nukleärer DNA in Form von NET-Bildung der neutrophilen Granulozyten betrachtet. Durch Vergleich der eben genannten Parameter einmal in der Kontrolle ohne Medikamentenzusatz, im Vergleich zu mindestens zwei eingesetzten unterschiedlichen Antibiotika bzw. Prednisolon-Konzentrationen sollte die Hypothese bestätigt oder widerlegt werden. Insgesamt betrachtet konnte festgestellt werden, dass alle getesteten Medikamente neutrophile Granulozyten in ihrer Funktion bezüglich Migration, NETose, ROS-Produktion, ROS-Intensität und Myeloperoxidase-Freisetzung nicht inhibieren. Somit wurde durch diese Resultate die Forschungshypothese widerlegt.

The objective of this doctoral thesis was to conduct dedicated in vitro research into the direct effect of the antibiotics meropenem, cefuroxime, moxifloxacin, clarithromycin and the glucocorticoid prednisolone on the interaction of the defense mechanisms of neutrophil granulocytes. The objective of this doctoral thesis was to conduct dedicated in vitro research into the direct effect of the antibiotics meropenem, cefuroxime, moxifloxacin, clarithromycin and the glucocorticoid prednisolone on the interaction of the defense mechanisms of neutrophil granulocytes. Differing studies showed a partly contradictory, ambivalent spectrum of effects of the drugs under investigation on the body's own immune defense. On the one hand, there appears to be a promotion and improvement in granulocyte activity, but on the other hand there is also a drop in the neutrophil count (neutropenia). Thus, the explicit research hypothesis arose as to whether neutrophil granulocytes are inhibited in vitro by certain antibiotic classes or the glucocorticoid prednisolone. Therefore, migration length, migration speed, the production of reactive oxygen species as well as the release of the enzyme myeloperoxidase and of nuclear DNA in the form of NET formation of neutrophil granulocytes were examined. The hypothesis was to be confirmed or refuted by comparing the aforementioned parameters once in the control without the addition of medication, in comparison with at least two different antibiotics or prednisolone concentrations used. Overall, it was found that all the drugs tested did not inhibit neutrophil granulocyte function in terms of migration, NETosis, ROS production, ROS intensity and myeloperoxidase release. These results therefore disproved the research hypothesis.
