Abstract (German)

Das Protein Melanoma Inhibitory Activity (MIA) wird als Serum Tumormarker des malignen Melanoms diskutiert. Bisher waren jedoch wichtige präanalytische, analytische und postanalytische Aspekte der MIA Bestimmung mittels Quantikine ELISA Human MIA Immunoassay (R&D Systems Inc., USA) unzureichend geklärt. Somit lag das Ziel der vorliegenden Studie in der Untersuchung der wichtigen präanalytischen Faktoren der Probenstabilität und Interferenzanfälligkeit. Zudem sollten die analytischen Leistungsmerkmale des Quantikine ELISA Human MIA Immunoassay bewertet werden. Zur Verbesserung der postanalytischen Interpretation wurden laborinterne Grenzwerte indirekt anhand eines großen Datensatzes statistisch berechnet. Ergänzend wurde ein Methodenvergleich mit einem zweiten human MIA ELISA (Roche Diagnostics GmbH, Deutschland) durchgeführt.
Zusammenfassend waren MIA Serumproben bei Raumtemperatur 1 Tag stabil. Für Lagerung und Versand waren MIA Proben bei 4-6 °C oder -20 °C im 14-tägigen Untersuchungszeitraum stabil. Die analytischen Leistungsmerkmale des Quantikine ELISA Human MIA Immunoassay eignen sich für die Diagnostik. Die MIA ELISA von R&D Systems und Roche Diagnostics haben eine unterschiedliche Wertelage, korrelieren jedoch gut. Neu bestimmte indirekte MIA Referenzbereiche können bei der Interpretation der Serum MIA Konzentration unterstützen.

The protein melanoma inhibitory activity (MIA) is being discussed as a serum marker for melanoma. However, up until now important preanalytical, analytical and postanalytical aspects of MIA quantification by Quantikine ELISA Human MIA Immunoassay (R&D Systems Inc., USA) were not fully clarified. The aim of this study was therefore to evaluate important preanalytical aspects of sample stability and assay interferences. Furthermore, analytical performance of the Quantikine ELISA Human MIA Immunoassay was evaluated. To improve postanalytical interpretation intra-laboratory reference limits were determined from a big dataset using an indirect statistical method. Additionally, an analytical method comparison with another human MIA ELISA (Roche Diagnostics GmbH, Germany) was performed.
In conclusion, serum samples for MIA quantification were stable up to 1 day at room temperature. For storage und shipping, MIA samples stored at 4-6 °C or -20 °C were stable during the maximum studied period of 14 days. Analytical performance of the Quantikine ELISA Human MIA Immunoassay was suitable for diagnostic purposes. Measurements by R&D Systems and Roche Diagnostics MIA ELISA did not show agreement, but the measurements correlated well. New MIA reference limits determined with an indirect method can aid in the interpretation of serum MIA levels.
