The 18-kDa translocator protein (TSPO) is a protein of the outer mitochondrial membrane. Due to its location, it has been associated with key cellular functions. It has been shown that TSPO expression is elevated in a range of brain pathologies, including the most aggressive brain tumor glioblastoma (GB), making TSPO a possible therapeutic target. To formulate a first characterization of GRT16085N (GRT16), the effects of the ligand were compared to established TSPO agonists, namely XBD173 and Etifoxine. Modulation of TSPO was examined in a human, mesenchymal glioma stem cell line (BTIC13) with respect to cytotoxicity, cytokine expression, mitochondrial respiration, generation of ROS and apoptosis. The aim of our study was to evaluate the effects of GRT16 in order to develop translational pathways for the inhibition of pathophysiologically relevant mechanisms in patients with GB.
Our data imply that GRT16 does not affect cell viability by itself, but might promote the effect of the pro-apoptotic agent TRAIL. Furthermore, GRT16 significantly impaired mitochondrial respiration. Other areas of investigation remained relatively unaffected, urging further investigations to fully establish TSPO-mediated effects and the putative role of GRT16085N in possible drug development for GB.

Das Glioblastom stellt den aggressivsten malignen Tumor dar und bleibt bis heute eine therapeutische Herausforderung. Einer dieser Ansatzpunkte ist das bereits länger bekannte 18-kDa Translokationsprotein (TSPO), welches von Glioblastomen vermehrt exprimiert wird. Aufgrund seiner Lokalisation an der äußeren Mitochondrienmembran wurde das Protein mit zahlreichen zellulären Schlüsselfunktionen in Verbindung gebracht, die in der Pathogenese des Glioblastoms eine Rolle spielen.
Mit dieser Arbeit erfolgte eine erste Charakterisierung von GRT16 hinsichtlich Zytotoxizität, Zytokinexpression, mitochondriale Zellatmung, ROS Synthese und Apoptose in einer humanen Hirntumor Stammzelllinie (BTIC13) vom mesenchymalen Typ. Ziel dieser Studie ist es, durch ein besseres Verständnis des Wirkprofils des TSPO Liganden GRT16, dazu beizutragen, translationale Wege zur Hemmung pathophysiologisch relevanter Mechanismen bei Glioblastompatienten zu entwickeln.
Unsere Daten legen nahe, dass GRT16 die Zellviabilität allein nicht beeinflusst, allerdings die Wirkung des pro-apoptotischen Wirkstoffs TRAIL fördert. Darüber hinaus beeinträchtigt GRT16 die mitochondriale Atmung signifikant. Andere Funktionen blieben weithin unbeeinträchtigt durch die Liganden. Weitere Untersuchungen sind erforderlich, um TSPO-vermittelte Effekte vollumfänglich zu erfassen und die mögliche Rolle von GRT16085N als therapeutischer Ansatzpunkt genauer zu definieren.
