Abstract (German)
Das Renin-Angiotensin-System (RAAS) spielt eine entscheidende physiologische Rolle bei der Regulation des Salz- und Wasserhaushalts sowie des Blutdrucks. Darüber hinaus trägt es maßgeblich zur Progression von Herz-Kreislauf-Erkrankungen und Nierenerkrankungen bei. Es gibt zahlreiche Hinweise dafür, dass das lokale RAAS bei diesen Erkrankungen aktiviert wird. In dieser Arbeit wurde herausgearbeitet, dass Renin nicht nur in den juxtaglomerulären Zellen, sondern auch in den interstitiellen Zellen der Niere exprimiert wird. Die Renin-produzierenden interstitiellen Zellen scheinen unabhängig von den juxtaglomerulären Zellen reguliert zu werden und könnten daher möglicherweise eine bedeutende Rolle bei der Regulation des lokalen RAAS in der Niere spielen. Da Renin produzierende interstitielle Zellen den PDGFR β exprimieren, wurde eine PDGFR β-Cre-Mauslinie verwendet, um eine zellspezifische Deletion von Renin in interstitiellen Zellen (PDGFR β-Cre/Ren KO) zu induzieren. Bereits unter physiologischen Bedingungen zeigen sich deutliche Unterschiede zwischen den beiden Genotypen. PDGFR β Ren KO Mäusen weisen geringere Renin-, Prorenin-und Aldosteron-Konzentrationen auf, sowie einen niedrigeren Blutdruck als PDGFR β Ren WT Mäuse. Insgesamt haben PDGFR β Ren KO Mäuse durch den interstitiellen Renin KO ein abgeschwächtes RAAS. Während die Zahl der Renin exprimierenden interstitiellen Zellen unter Kontrollbedingungen eher gering war, stieg sie bei PDGFR β Ren WT Mäusen unter einer Adeninnephropathie und einer diabetischen Nephropathie deutlich an (Renin-mRNA-in-situ-Hybridisierung). Wie erwartet, wird die Renin mRNA in den interstitiellen Zellen von PDGFR β Ren KO Mäusen weder unter Kontrollbedingungen noch unter Schädigungsbedingungen nachgewiesen. Die Adeninnephropathie und diabetische Nephropathie sind bei beiden Genotypen mit einer Suppression der Plasma-Renin-Konzentration verbunden. Im Gegensatz dazu ist die Plasmakonzentration von enzymatisch inaktivem Prorenin, das mit einem verstärkten Fortschreiten der Nierenerkrankung einhergeht, bei PDGFR β Ren WT Adenin Mäusen im Vergleich zu gesunden Kontrollmäusen um das Dreifache erhöht. PDGFR β Ren KO Adenin Mäuse verzeichnen keinen Prorenin Anstieg im Plasma. Unter diabetischen Bedingungen hingegen wird bei beiden Genotypen ein Anstieg von Prorenin im Plasma beobachtet, was durch die gleichzeitige Aktivierung des Tubulus-Renins unter diabetischen Bedingungen erklärt werden könnte. Um gezielt die Auswirkungen des interstitiellen Renins zu untersuchen, erweist sich daher das Adenin-Modell als geeigneter. Die PDGFR β Ren KO Adenin Mäuse zeigen hinsichtlich glomerulärer Filtrationsrate (GFR) sowie in Bezug auf die Expression von Fibronektin und F4/80 mRNA eine geringere Nierenschädigung im Vergleich zu den PDGFR β Ren WT Adenin Mäusen. Zusammenfassend lässt sich festhalten, dass die Expression von Renin in den PDGFRβ-positiven interstitiellen Zellen unter Diabetes und einer Adeninnephropathie stimuliert wird und dies zu den zirkulierenden Renin- und Prorenin-Spiegeln im Plasma beiträgt. Die gezielte Deletion von Renin in den PDGFR β-positiven Zellen bei PDGFR β Ren KO Adenin Mäuse verhindert den Anstieg von Prorenin im Plasma und führt zu einer Verbesserung der Nierenschädigung. Dies weist darauf hin, dass das interstitielle Renin ein krankheitsfördernder Faktor sein könnte.

The renin-angiotensin system (RAAS) plays a crucial physiological role in the regulation of salt and water balance as well as blood pressure. It also contributes significantly to the progression of cardiovascular diseases and kidney diseases. There are numerous indications that the local RAAS is activated in these diseases. In this study it was shown that renin is expressed not only in the juxtaglomerular cells, but also in the interstitial cells of the kidney. The renin-producing interstitial cells appear to be regulated independently of the juxtaglomerular cells and could therefore possibly play an important role in the regulation of the local RAAS in the kidney. Since renin-producing interstitial cells express the PDGFR β, a PDGFR β-Cre mouse line (PDGFR β-Cre/Ren KO) was used to establish a cell-specific deletion of renin in interstitial cells. Even under physiological conditions there are clear differences between the two genotypes. PDGFR β Ren KO mice have lower renin, prorenin and aldosterone concentrations as well as lower blood pressure than PDGFR β Ren WT mice. Overall PDGFR β Ren KO mice have an attenuated RAAS due to the interstitial renin KO. While the number of renin-expressing interstitial cells was rather low under control conditions, it increased significantly in PDGFR β Ren WT mice under adenine nephropathy and diabetic nephropathy (renin mRNA in situ hybridization). As expected, renin mRNA is not detected in the interstitial cells of PDGFR β Ren KO mice under either control or damage conditions. Adenine nephropathy and diabetic nephropathy are associated with suppression of plasma renin concentration in both genotypes. In contrast, the plasma concentration of enzymatically inactive prorenin, which is associated with increased renal disease progression, is increased threefold in PDGFR β Ren WT adenine mice compared to healthy control mice. PDGFR β Ren KO adenine mice do not show an increase in prorenin in plasma. Under diabetic conditions, however, an increase of prorenin in plasma is observed in both genotypes, which could be explained by the simultaneous activation of tubule renin under diabetic conditions. The adenine model is therefore more suitable for specifically investigating the effects of interstitial renin. The PDGFR β Ren KO adenine mice show less renal damage in terms of glomerular filtration rate (GFR) and the expression of fibronectin and F4/80 mRNA compared to the PDGFR β Ren WT adenine mice. In conclusion, renin expression is stimulated in PDGFRβ-positive interstitial cells under diabetes and adenine nephropathy, and this contributes to circulating plasma renin and prorenin levels. Targeted deletion of renin in the PDGFR β-positive cells in PDGFR β Ren KO adenine mice prevents the increase of prorenin in plasma and leads to amelioration of renal injury. This indicates that interstitial renin may be a disease-promoting factor.
