Abstract (German)

In der vorliegenden Promotionsarbeit wurden Zellen einer ARVC Patientin mit denen einer gesunden Kontrollpatientin und mit mutationskorrigierten Zellen der ARVC Patientin in Hinblick auf die Produktion von mitochondrialen Sauerstoffradikalen verglichen. Mit Hilfe der hiPSC-CM wurden Unterschiede zwischen den Zelllinien herausgearbeitet und mögliche Begründungen der veränderten mtROS Produktion geklärt. Dabei zeigte sich eine tendenziell erhöhte Bildung von mtROS bei der ARVC-Patientin.
Da die Bildung von Sauerstoffradikalen in den Mitochondrien komplex ist, lies sich diese Beobachtung nicht klar auf eine oder mehrere Ursachen beschränken. Es wurden wichtige mtROS bildende und antioxidative Proteine in den Zelllinien vergleichend untersucht.
Ursachen für die Unterschiede der mitochondrialen Sauerstoffradikalbildung zwischen den Zelllinien konnten in vorliegender Dissertationsarbeit herausgearbeitet werden. Zum einen konnte der Atmungskettenkomplex II als möglicher mtROS-Bildner bei der ARVC Patientin aufgedeckt werden. Auch haben die Unterschiede in der Ausprägung der Glukosetransporter SGLT1 und GLUT4 sowie des Antioxidans SOD2 mögliche Ursachen in der vermehrten mtROS Produktion der ARVC Zelllinien aufgebracht. Außerdem kann die Expression der NADPH Oxidasen 2 und 4 das unterschiedliche Verhalten der Zellen in Stresssituationen klären.

In the present thesis, cells from an ARVC patient were compared with those of a healthy control patient and with mutation-corrected cells from the ARVC patient with regard to the production of mitochondrial oxygen radicals. With the help of hiPSC-CM, differences between the cell lines were identified and possible reasons for the altered mtROS production were clarified. This revealed a tendency towards increased formation of mtROS in the ARVC patient.
Since the formation of oxygen radicals in the mitochondria is complex, this observation could not be clearly limited to one or more causes. Important mtROS-forming and antioxidant proteins in the cell lines were examined comparatively.
Causes for the differences in mitochondrial oxygen radical formation between the cell lines were identified in this dissertation. On the one hand, respiratory chain complex II was identified as a possible mtROS producer in the ARVC patient. Furthermore, the differences in the expression of the glucose transporters SGLT1 and GLUT4 as well as the antioxidant SOD2 may have led to increased mtROS production in the ARVC cell lines. In addition, the expression of the NADPH oxidases 2 and 4 can clarify the different behavior of the cells in stress situations.
