Abstract (German)
Die Lipoxygenasen Lox-5, Lox-12 und Lox-15 sowie die Cytochromoxidasen Cyp4A1, Cyp4A2, Cyp4A3, Cyp2C23 und Cyp2C11, die unter anderem eine sehr wichtige Rolle im Arachidonsäure-Stoffwechsel spielen, übernehmen eine Reihe von entscheidenden Funktionen, wie z.B. die Vermittlung von Entzündungsreaktionen, strukturelle Veränderungen bei Diabetes, Regulierung des Blutdrucks und viele weitere Prozesse bei anderen Erkrankungen. Daher ist deren Lokalisation in den einzelnen Nephronsegmenten des Cortex sowie der äußeren und inneren Medulla von enormer Bedeutung. Rattennieren wurden hierfür in Cortex, äußerer und innerer Medulla mikrodisseziert. Die mikrodissezierten Nephronsegmente wurden nach Kollagenase-Verdau gesammelt und die mRNA-Verteilung wurde durch RT-PCR untersucht.
So konnte man in den Versuchen die Lox-12 vor allem in Glom- entdecken. Die Signale in mTAL, cTAL, und CCD/CT in diesen Versuchen waren jedoch sehr schwach, so dass diese wohl daher vernachlässigt werden können. Lox-15-Expressionen fanden sich dagegen in cTAL, DCT, CCD/CT, OMCD und IMCD mit zunehmender Expressionsstärke in Richtung der Sammelrohre. Die Lox-5-mRNA konnte man hauptsächlich in Glom+ und Glom- entdecken. Die Signale in PCT, mTAL, cTAL, DCT, CCD/CT, OMCD und IMCD waren jedoch sehr schwach, so dass auch diese vernachlässigt werden können. Sie können auch durch Verunreinigungen aus VR stammen, in denen tatsächlich die Lox-5-Expression nachgewiesen werden konnte. Was die Verteilung der Cyp4A1 angeht, konnten bei diesen Versuchen Expressionen der PCT in jeder Sammlung beobachtet werden. Gelegentliche sehr schwache Signale fanden sich außerdem in Glom-, dTL, mTAL und cTAL, welche durch Verunreinigungen von anderen Zellen stammen könnten, zumal die Cyp4A1-mRNA bereits in Epithelzellen der proximalen Tubuli und generell in Zellen anderer Organe entdeckt werden konnte. Cyp4A2 bzw. Cyp4A3 wurden dagegen gemeinsam untersucht, da die Primer homologe Bereiche der beiden Enzyme erkennen. Cyp4A2- bzw. Cyp4A3-Signale konnten in diesen Versuchen in PCT, Glom-, Glom+ und dTL entdeckt werden. Die Cyp2C23 mRNA dagegen konnte in Glom-, Glom+, PCT und in schwächerer Form in IMCD lokalisiert werden. Bezüglich der Verteilung der Cyp2C11 lassen die Versuche keine eindeutige Aussage zu, da die Ergebnisse der Sammlungen sehr unterschiedlich ausfielen. Demnach ist die Cyp2C11-mRNA eventuell in PCT und IMCD exprimiert. Vernachlässigbar schwache Signale wurden in CCD/CT und Glom+ gefunden.
Insgesamt betrachtet kann man durch Blockierung gewisser Abbauprodukte der Arachidonsäure, wie z.B. Prostaglandine, Leukotriene oder 20-HETE, die durch Cyclooxygenasen, Lipoxygenasen und Cytochromoxidasen freigesetzt werden, nicht nur Schmerzen hemmen, sondern auch Erkrankungen wie Asthma, Krebs oder Herz-Kreislauf-Erkrankungen beeinflussen. Schon daher zeigt sich die Bedeutung der Lokalisation dieser Enzyme.

Translation of the abstract (English)
The lipoxygenases 5-LOX, 12-LOX and 15-LOX as well as the cytochrome P450 enzymes Cyp4A1, Cyp4A2, Cyp4A3, Cyp2C23 and Cyp2C11, which play a very important role in arachidonic acid metabolism, among other things, fulfil a number of crucial functions, such as mediating inflammatory reactions, structural changes in diabetes, regulating blood pressure and many other processes in other diseases. Their localization in the individual nephron segments of the cortex and the outer and inner medulla is therefore of enormous importance. Rat kidneys were microdissected in the cortex, outer and inner medulla for this purpose. The microdissected nephron segments were collected after collagenase digestion and the mRNA distribution was analysed by RT-PCR.
In the experiments, 12-LOX was detected mainly in Glom-. However, the signals in mTAL, cTAL, and CCD/CT in these experiments were very weak, so that they can probably be neglected. In contrast, 15-LOX expression was found in cTAL, DCT, CCD/CT, OMCD and IMCD with increasing expression levels towards the collection tubes. 5-LOX mRNA was mainly detected in Glom+ and Glom-. However, the signals in PCT, mTAL, cTAL, DCT, CCD/CT, OMCD and IMCD were very weak, so that these can also be neglected. They may also originate from impurities from VR in which 5-LOX expression could actually be detected. Regarding the distribution of Cyp4A1, PCT expression could be observed in each collection in these experiments. Occasional very weak signals were also found in Glom-, dTL, mTAL and cTAL, which could originate from contamination from other cells, especially since the Cyp4A1 mRNA has already been detected in epithelial cells of the proximal tubules and generally in cells of other organs. Cyp4A2 and Cyp4A3, on the other hand, were investigated together, as the primers recognise homologous regions of both enzymes. Cyp4A2 and Cyp4A3 signalling was detected in PCT, Glom-, Glom+ and dTL in these experiments. In contrast, the Cyp2C23 mRNA could be localized in Glom-, Glom+, PCT and in a weaker form in IMCD. With regard to the distribution of Cyp2C11, the experiments do not allow a clear statement to be made, as the results of the collections were very different. Accordingly, the Cyp2C11 mRNA is possibly expressed in PCT and IMCD. Negligible weak signals were found in CCD/CT and Glom+.
Overall, by blocking certain degradation products of arachidonic acid, such as prostaglandins, leukotrienes or 20-HETE, which are released by cyclooxygenases, lipoxygenases and cytochrome oxidases, it is possible not only to inhibit pain, but also to influence diseases such as asthma, cancer or cardiovascular diseases. This alone demonstrates the importance of the localization of these enzymes.
