Abstract (German)
In der vorliegenden Arbeit wurde die Wirkung von KAP auf die Funktionalität neutrophiler Granulozyten untersucht. Polymorphkernige neutrophile Granulozyten (PMN) stellen die erste Verteidigungslinie des Immunsystems gegen eindringende Pathogene dar, indem sie eine rasche und effektive Antwort auf Infektionsherde ermöglichen. Die Rolle der PMN, insbesondere ihre Fähigkeit, den Prozess der Phagozytose zu initiieren und Entzündungsreaktionen zu modulieren, ist zum Beispiel bei der Wundheilung von großer Bedeutung. Wenn gasförmiger Materie weiter Energie zugeführt wird, entsteht Plasma. KAP bezeichnet jenes medizinisch nutzbare Plasma, welches durch eine Temperatur unter 40 °C für den Einsatz am vitalen Gewebe geeignet ist. Die wirksamen Bestanteile von KAP sind mit Ionen und Elektronen, UV- und IR- Strahlung, sichtbarem Licht, elektromagnetischen Feldern, Wärmestrahlung und reaktiven Spezies sehr vielfältig. Im medizinischen Kontext sind neben der UV-Strahlung und dem elektrischen Feld vor allem die im KAP enthaltenen reaktiven Sauerstoffspezies (ROS) und reaktiven Stickstoffspezies (RNS), von Bedeutung. Die antimikrobielle Wirkung von KAP sowie seine potenzielle Rolle bei der Förderung der Wundheilung und Modulation der Immunantwort bieten vielversprechende Perspektiven für
therapeutische Anwendungen. Insbesondere die Fähigkeit von KAP, die Produktion von ROS zu stimulieren, könnte entscheidend für die Beeinflussung der zellulären Funktionen von neutrophilen Granulozyten sein. In dieser Studie wurden zunächst neutrophile Granulozyten aus dem Blut gesunder Proband*innen isoliert. Anschließend erfolgte die Behandlung des
Zellkulturmediums mit KAP durch zwei verschiedene Plasmaquellen. Die PMN selbst waren nicht direkt einer Plasmabehandlung ausgesetzt. Der Kontakt der PMN zu der PTS (plasma treated solution) kam in einem speziellem μ-Slide Chemotaxis® (siehe 2.2.5) zustande. Für jede einzelne Versuchsdurchführung stand unbehandeltes Zellkulturmedium als Kontrolle zur Verfügung. Das μ-Slide Chemotaxis® ermöglichte durch seine verschiedenen Kammern eine
modifizierte Befüllung und somit die Durchführung von drei verschiedenen
Versuchsaufbauten, die jeweils separat ausgewertet wurden.
Ziel war es, die Auswirkungen von KAP auf die zelluläre Funktionalität der PMN zu untersuchen. Mittels Life-Cell-Imaging wurden die Bewegungen und Verhaltensweisen der neutrophilen Granulozyten über sieben Stunden automatisiert unter einem Mikroskop beobachtet. Die so gewonnenen Bilddaten wurden mit Hilfe verschiedener Software objektiviert und statistisch ausgewertet. Von Interesse waren Effekte von KAP auf die Zellmigration und die ROS-Produktion sowie die NETose als Indikatoren für die Aktivierung der neutrophilen Granulozyten. In den Ergebnissen zeigte sich abhängig von der
Behandlungsdauer der PTS bei beiden Plasmaquellen eine signifikant früher einsetzende ROS- Produktion. Die NETose setzte bei Verwendung von MicroPlaSter® signifikant früher ein, bei plasma care® blieb sie unverändert. Die Migration der PMN blieb bei beiden Plasmaquellen weitgehend unverändert. Die PTS zeigte keine chemoattraktive Wirkung auf die PMN.
Die Studie liefert wichtige Einblicke in die potenzielle Rolle von KAP in der Immunmodulation und Wundheilung. Die Fähigkeit von KAP, spezifische zelluläre Reaktionen in PMN zu induzieren, unterstreicht das therapeutische Potenzial dieser Technologie in der medizinischen Praxis. Zukünftige Forschungen sollten sich darauf konzentrieren, die molekularen Mechanismen hinter den beobachteten Effekten weiter zu entschlüsseln und die optimale
Dosierung sowie die Feineinstellungen von KAP in Abhängigkeit von der verwendeten Plasmaquelle zu bestimmen, um die Effizienz und Sicherheit der Anwendung zu maximieren.
Zusammenfassend zeigt diese Arbeit, dass kaltes atmosphärisches Plasma eine signifikante Wirkung auf die Funktionalität neutrophiler Granulozyten hat. Abwägend könnte die Modulation der Immunantwort durch KAP eine innovative Möglichkeit bieten die Wundheilung zu fördern und die Abwehr gegen mikrobielle Infektionen zu verstärken. Hierzu sind weitere Untersuchungen erforderlich, um das volle Potenzial von KAP in der Medizin zu
erschließen und praktische Anwendungen für die klinische Praxis zu entwickeln.

This study investigated the effect of cold atmospheric plasma (CAP) on the functionality of neutrophilic granulocytes. Polymorphonuclear neutrophils (PMNs) are the first line of defense of the immune system against invading pathogens by enabling a rapid and effective response to infection sites. The role of PMNs, particularly their ability to initiate the process of phagocytosis and modulate inflammatory reactions, is of great importance, for example, in wound healing.
When gaseous matter is further supplied with energy, plasma is formed. CAP refers to medically usable plasma that is suitable for application on vital tissue due to its temperature being below 40°C. The active components of CAP are highly diverse and include ions and electrons, UV and IR radiation, visible light, electromagnetic fields, heat radiation, and reactive species. In the medical context, in addition to UV radiation and the electric field, the reactive oxygen species (ROS) and reactive nitrogen species (RNS) present in CAP are of particular importance. The antimicrobial effect of CAP, as well as its potential role in promoting wound healing and modulating the immune response, offer promising prospects for therapeutic applications. In particular, the ability of CAP to stimulate ROS production could be crucial in influencing the cellular functions of neutrophilic granulocytes.
In this study, neutrophilic granulocytes were first isolated from the blood of healthy volunteers. The cell culture medium was then treated with CAP using two different plasma sources. The PMNs themselves were not directly exposed to plasma treatment. The contact between the PMNs and the plasma-treated solution (PTS) was established in a special μ-Slide Chemotaxis® (see 2.2.5). For each experiment, untreated cell culture medium was used as a control. The μ-Slide Chemotaxis® allowed for a modified filling through its various chambers, enabling the conduct of three different experimental setups, each of which was evaluated separately.
The goal was to investigate the effects of CAP on the cellular functionality of PMNs. Using live-cell imaging, the movements and behaviors of the neutrophilic granulocytes were automatically observed under a microscope for seven hours. The resulting image data were objectively quantified and statistically analyzed using various software tools. The effects of CAP on cell migration, ROS production, and NETosis were of particular interest as indicators of neutrophil activation.
The results showed that depending on the treatment duration of the PTS, both plasma sources led to a significantly earlier onset of ROS production. NETosis occurred significantly earlier with the use of MicroPlaSter®, while it remained unchanged with plasma care®. The migration of PMNs remained largely unaffected by both plasma sources. The PTS showed no chemoattractive effect on PMNs.
The study provides important insights into the potential role of CAP in immune modulation and wound healing. The ability of CAP to induce specific cellular reactions in PMNs highlights the therapeutic potential of this technology in medical practice. Future research should focus on further elucidating the molecular mechanisms behind the observed effects and determining the optimal dosage and fine-tuning of CAP depending on the plasma source used, in order to maximize the efficiency and safety of its application.
In conclusion, this study shows that cold atmospheric plasma has a significant effect on the functionality of neutrophilic granulocytes. Weighing the evidence, modulation of the immune response by CAP could offer an innovative way to promote wound healing and enhance defense against microbial infections. Further studies are needed to unlock the full potential of CAP in medicine and to develop practical applications for clinical practice.
