Abstract (German)
Das Ziel der medizinischen Doktorarbeit war es, herauszufinden, ob die bei der Patientin detektieren Mutationen CACNA1HS475T und CACNA1HR788C bzw. ihre Kombination CACNA1HS475T/R788C ursächlich für einen Primären Hyperaldosteronismus sein können. Dafür wurden NCI Zellen mit Plasmiden transfiziert, welche zum einen genau diese mutierten Gene trugen und zum anderen die Sequenz für den WT CACNA1H. ...
Das Ziel der medizinischen Doktorarbeit war es, herauszufinden, ob die bei der Patientin detektieren Mutationen CACNA1HS475T und CACNA1HR788C bzw. ihre Kombination CACNA1HS475T/R788C ursächlich für einen Primären Hyperaldosteronismus sein können. Dafür wurden NCI Zellen mit Plasmiden transfiziert, welche zum einen genau diese mutierten Gene trugen und zum anderen die Sequenz für den WT CACNA1H. An diesen Zellen wurden anschließend Whole-Cell Patch Clamp Experimente durchgeführt und neben Strom-Spannungskurven auch die Zeit der Aktivierung und Inaktivierung des CACNA1H gemessen. Die Gruppen wurden untereinander verglichen und die Ergebnisse hinsichtlich statistischer Unterschiede untersucht. Es konnten zwar Unterschiede, zwischen WT CACNA1H und den mutierten Kanälen gefunden werden, diese waren aber nicht statistisch signifikant.
Aufgrund dieser Ergebnisse kann gefolgert werden, dass es unwahrscheinlich ist, dass die Mutationen CACNA1HS475T, CACNA1HR788C und CACNA1HS475T/R788C allein für die Entstehung des Primären Hyperaldosteronismus verantwortlich sind, wenn sie vielleicht auch dazu beitragen.
Translation of the abstract (English)
The goal of this work was to find out whether novel mutations of the voltage-dependent Ca2+-Channel CACNA1H – namely CACNA1HS475T, CACNA1HR788C or the combination of both mutations CACNA1HS475T/R788C – could lead to the development of Primary Hyperaldosteronism. For that Realtime-PCR and Whole-Cell-Patch-Clamp experiments were executed on transient transfected NCI-cells. Wildtype CACNA1H ...
The goal of this work was to find out whether novel mutations of the voltage-dependent Ca2+-Channel CACNA1H – namely CACNA1HS475T, CACNA1HR788C or the combination of both mutations CACNA1HS475T/R788C – could lead to the development of Primary Hyperaldosteronism.
For that Realtime-PCR and Whole-Cell-Patch-Clamp experiments were executed on transient transfected NCI-cells.
Wildtype CACNA1H cells were then compared to the cells expressing the differently mutated CACNA1H. There were no significant findings between the groups in the Realtime-PCR experiments regarding the expression of CYP11B2. Electrophysiology and kinetics of CACNA1H measured in Whole-Cell-Patch-Clamp experiments seem to be not affected by the mutations since there were no significant differences detectable between the groups.
Regarding these results the conclusion is that the novel mutations are not likely to lead to the development of Primary Hyperaldosteronism.
