Connexine sind Intramembranproteine und Bausteine von Gap Junctions, welchen eine große Bedeutung für die interzelluläre Kommunikation zugemessen wird und deren Eigenschaften durch die Art ihrer Zusammensetzung bestimmt werden, womit Connexinen in unterschiedlichen
Organsystemen verschiedene, teilweise kritische Funktionen zukommen. In der Niere konnte für Connexin 43 eine Beeinflussbarkeit der Progression der chronischen Nierenerkrankung demonstriert werden. Connexin 40 (Cx40) andererseits ist essenziell für die Regulation der Ausschüttung von Renin, einem ebenfalls für die Pathophysiologie von Nierenerkrankungen relevanten Mechanismus. Ziel dieser Arbeit war die Untersuchung der Bedeutung von Cx40 bei der Entstehung und Progression der Nierenfibrose, dem Endstadium der meisten chronischen Nierenerkrankungen. Dazu wurde an der Maus die Expression von Cx40 in der basalen Wildtyp-Niere, in zwei fibrotischen Nierenmodellen, induziert durch Adenin oder unilaterale Ureterobstruktion, sowie einem Nierenmodell mit Knockout des Cx40-Gens in PDGFRβ-exprimierenden Zellen auf Proteinebene mittels konventioneller Immunfluoreszenz (IF) und auf mRNA-Ebene mittels in-situ-Hybridisierung (ISH, RNAscope® von ACD Bio) analysiert.
Eine Expression von Cx40 wird mittels IF und ISH im basalen und fibrotischen Nierengewebe im intraglomerulären Mesangium (IGM) und im angrenzenden juxtaglomerulären Apparat (JGA) sowie im Endothel aller renalen Gefäße, mit Ausnahme der intraglomerulären Kapillarschlingen, nachgewiesen. Im fibrotischen Nierengewebe ist die Cx40-Expression in Endothelzellen der Vasa
recta und im inneren Mark verstärkt. Allein die IF liefert darüber hinaus Hinweise auf eine Expression von Cx40 in Tubuluszellen des basalen und fibrotischen Nierenmarks. Nur mittels ISH wurde eine ausgeprägte Rekrutierung von Renin-mRNA-exprimierenden Zellen entlang der Vasa recta im Bereich
der äußeren Medulla dargestellt. Im Modell mit Cx40-Knockout in PDGFRβ-exprimierenden Zellen zeigte sich eine Rekrutierung von Renin-exprimierenden myoepithelialen Zellen entlang des Vas afferens sowie in Form von juxtaglomerulären α-SMA-negativen Zellen.
Eine Expression von Cx40 durch Myofibroblasten konnte in keinem Modell dargestellt werden, was gegen eine direkte Involvierung des Connexins in die zur Fibrose führenden Signalwege spricht, auch wenn die sichtbare Veränderung der Expressionslevel zwischen basalem und fibrotischem Modell
eine Beteiligung an der Gewebsreaktion, wie etwaigen Kompensationsmechanismen, nahelegt. Da im Knockout-Modell über die oben genannten Beobachtungen hinaus keine Allgemeinveränderungen wie insbesondere eine pathologische Fibrosierung nachgewiesen werden konnte, ergibt sich erneut kein Hinweis auf eine elementare Bedeutung von Cx40 für die Organisation von mesenchymalen Zellen in der Niere. Eine vervollständigende Untersuchung dieses Knockout-Modells nach Fibroseinduktion inklusive ISH wurde durch uns noch nicht durchgeführt. Die Rekrutierung von Renin-exprimierenden Zellen im fibrotischen und im Knockout-Modell betont erneut die essenzielle Bedeutung von Cx40 für die Regulation der Renin-Ausschüttung, welche ihrerseits einen Mediator zur
Entstehung und Progression der chronischen Nierenerkrankung darstellt, der Cx40 als Objekt weiterer Studien Bedeutung verleiht.

Connexins are intermembrane proteins and components of gap junctions which are attributed with significant importance for intercellular communication and whose properties are determined by their connexin composition. Therefore, connexins fulfil different, partially critical functions in various organ systems. For connexin 43 studies could demonstrate an influence on the progression of
chronic kidney disease. Connexin 40 (Cx40) on the other hand is essential for the regulation of renin secretion, another mechanism important for the pathophysiology of chronic kidney disease. The aim of this study was to research the role of Cx40 in the development of renal fibrosis, the final stage of
most variants of chronic kidney disease. Therefore, the expression of Cx40 in the basal wild-type kidney, in two types of fibrotic kidney models, respectively induced through adenine or unilateral ureteral obstruction, and a kidney model after knockout of the Cx40 gene in PDGFRβ expressing cells on protein level were analysed with conventional immunofluorescence (IF) and on mRNA level by insitu-hybridisation (ISH, RNAscope® by ACD Bio) in mice.
An expression of Cx40 is detected in basal and fibrotic kidney tissue in the intraglomerular mesangium (IGM) and in the adjacent juxtaglomerular apparatus (JGA), as well as in the endothelium of all renal blood vessels except the intraglomerular capillary loop by IF and ISH. In fibrotic kidney tissue expression of Cx40 in endothelial cells of the vasa recta and in the inner medulla is increased. Only IF indicates Cx40 expression in tubular cells of the basal und fibrotic kidney tissue. Only through ISH on the other hand a significant recruitment of renin mRNA expressing cells alongside the vasa recta in the outer medulla area is depicted. In the kidney model with knockout of Cx40 in PDGFRβ-expressing cells a recruitment of renin expressing myoepithelial cells alongside the vas afferens as well as in the form of juxtaglomerular α-SMA negative cells is shown.
An expression of Cx40 by myofibroblasts could not be shown in any kidney model which contradicts a direct involvement of the connexin in the signal pathways that lead to kidney fibrosis. Still the visible changes in expression between basal and fibrotic kidney model suggest a contribution to the tissue’s
reaction, like potential compensation mechanisms. The lack of general alterations of tissue like pathological fibrotic remodelling in the knockout model again does not hint to a fundamental significance of Cx40 for the organisation of mesenchymal cells in the kidney. A complete study of this
knockout model has not yet been realised by us. The recruitment of renin expressing cells in the fibrotic and the knockout model emphasizes again the essential impact of Cx40 on the regulation of renin secretion, which on its own is a mediator for the development and progression of chronic kidney disease and therefore gives significance to Cx40 as an object of further investigation.
