Abstract (German)
Seit seiner Identifikation als Entry-Rezeptor der SARS-Co-Viren gerät das Angiotensin Converting Enzyme 2 zunehmend in das Blickfeld biomedizinischer Forschung. Mit seiner Entdeckung im Jahr 2000 wurde das konventionelle RAAS um eine nicht kanonische Systemachse erweitert. Es spaltet Ang II und produziert weitere Angiotensinderivate, wie Ang(1-7) und Ang (1-9), welche als Liganden für seine beiden korrespondierenden Rezeptoren Mas-1 und AT2 fungieren. Es stellt dadurch einen essentiellen Regulator des Renin-Angiotensin-Aldosteron-Systems dar, dessen beiden Achsen in einem empfindlichen Gleichgewicht zueinander stehen, welches maßgeblich an der Aufrechterhaltung vieler physiologischer Funktionen im Organismus beteiligt ist.
Im Rahmen der Genese und Progression etlicher Erkrankungen kommt es zu einer Überaktivität des kanonischen Systemarms, welcher sowohl systemisch als auch lokal im Gewebe nachteilige Effekte vermittelt. In diesen pathologischen Zuständen agiert das nicht-kanonische RAAS als zentraler Mediator protektiver antagonistischer Effekte. Diese sind in erster Linie vasodilatativer, antioxidativer, antithrombotischer, antiproliferativer und antiinflammatorischer Art.
Die Datenlage hinsichtlich der Lokalisation des ACE2 erweist sich als sehr ambivalent. Da der Expressionsort und die Funktionalität eines Proteins eng miteinander verknüpft sind, ist es von großem Interesse, mehr über seine genaue Organ- und Zellverteilung zu erfahren. Ziel dieser Dissertation war es deshalb das Schlüsselenzym des nicht kanonischen RAAS sowie seine wichtigsten Rezeptoren in ausgewählten murinen Geweben genauer zu lokalisieren.
Dazu wurden zunächst quantitative mRNA- und Proteinanalysen mittels Real-Time qPCR bzw. Western Blot durchgeführt, um einen Eindruck über die interorganische Distribution der untersuchten Enzyme und Rezeptoren zu gewinnen. Haupttranskriptions- und Expressionsort des ACE2 war dabei die Niere, weshalb auf diese in den nachfolgenden Versuchen besonderes Augenmerk gelegt wurde. Bemerkenswert war außerdem, dass die Komponenten des nicht-kanonischen RAAS in manchen Geweben teilweise in ähnlicher oder sogar höherer Ausprägung präsent waren als die des kanonischen RAAS, was angesichts der Prädominanz der kanonischen Systemachse im pathophysiologischen Verständnis vieler Erkrankungen besonders relevant erscheint.
Im nächsten Schritt wurde durch Fluoreszenz-in-situ-Hybridisierung und Immunfluoreszenzfärbung versucht, den Komponenten des nicht-kanonischen RAAS wiederum auf mRNA- und Proteinebene zelluläre Strukturen zuzuordnen.
In Herz, Leber und Nebenniere konnte ACE2 am ehesten in Fibroblasten bzw. fibroblastoiden Zellen des Interstitiums nachgewiesen werden. Insbesondere in kardialem und hepatischem Gewebe legen diese Beobachtungen in Kombination mit Befunden aus vorherigen Studien nahe, dass es dort durch die Degradation von Ang II fibrotisch-degenerativen Prozessen entgegenwirken kann. In der murinen Lunge ist ACE2 am apikalen Pol des respiratiorischen Epithels der Bronchien lokalisiert.
Die ACE2-Level in der Niere waren die höchsten unter den untersuchen Organen. Dort tritt das nicht-kanonische Schlüsselenzym vorwiegend im Bürstensaum des proximalen Tubulus in Erscheinung, wo es eine Kolokalisation mit Mas-1 und seinem kanonischen Analogon ACE aufweist. Mas ist weiterhin in Mesangialzellen und der Muscularis der renalen Gefäße exprimiert. Dort könnte es EZM-Expansion im Rahmen einiger Nierenerkrankungen hemmen und die renale Durchblutung positiv beeinflussen.
Die erhobenen Befunde bezüglich der Lokalisation der Enzyme und Rezeptoren des nicht-kanonischen RAAS unterstützen seine postulierten protektiven Effekte auf unterschiedliche Organsysteme. Künftig könnten seine Komponenten deshalb eine wichtige Rolle in klinischen Therapieverfahren einnehmen, etwa bei arterieller Hypertonie, Herzinsuffizienz, thrombotisch-ischämischen Ereignissen, fibrotisch degenerativen und neoplastischen Organveränderungen und nicht zuletzt Infektionskrankheiten wie Covid-19. Gerade vor dem Hintergrund der erst kürzlich überwundenen Pandemie und nun endemischen SARS-CoV-2-Situation ist es von immenser Bedeutung, weitere Lokalisationsstudien zum Thema ACE2 durchzuführen und genauer zu eruieren, welche Bedeutung dem Enzym im Rahmen dieser Multisystemerkrankung zukommt.

Since its identification as the entry receptor of SARS-Co-viruses, angiotensin converting enzyme 2 has increasingly become the focus of biomedical research. With its discovery in 2000, a non-canonical system axis was added to the conventional RAAS. It degrades Ang II and produces further angiotensin derivatives, such as Ang(1-7) and Ang (1-9), which act as ligands for its two corresponding receptors Mas-1 and AT2. It is therefore an essential regulator of the renin-angiotensin-aldosterone system, the two axes of which are in a delicate balance with each other and play a key role in maintaining many physiological functions in the organism.
As part of the genesis and progression of a number of diseases, there is an overactivity of the canonical system arm, which mediates adverse effects both systemically and locally in the tissue. In these pathological conditions the non-canonical RAAS acts as a central mediator of protective antagonistic effects. These are primarily vasodilatory, antioxidant, antithrombotic, antiproliferative and anti-inflammatory.
The data situation regarding the localization of ACE2 proves to be very ambivalent. Since the expression site and the functionality of a protein are closely linked, it is of great interest to learn more about its exact organ and cell distribution. The aim of this thesis was therefore to localize the key enzyme of the non-canonical RAAS and its most important receptors more precisely in selected murine tissues.
To this end, quantitative mRNA and protein analyses were first carried out using real-time qPCR and Western blot, respectively, to gain an impression of the interorganic distribution of the enzymes and receptors investigated. The main transcription and expression site of ACE2 was the kidney, which is why particular attention was paid to this in the subsequent experiments.
It was also remarkable that the components of the non-canonical RAAS were present in some tissues to a similar or even higher degree than those of the canonical RAAS, which seems particularly relevant in view of the predominance of the canonical system axis in the pathophysiological understanding of many diseases.
In the next step, fluorescence in situ hybridization and immunofluorescence staining were used to assign cellular structures to the components of the non-canonical RAAS, again at the mRNA and protein level.
In the heart, liver and adrenal gland, ACE2 was most likely to be detected in fibroblasts or fibroblastoid cells of the interstitium. Especially in cardiac and hepatic tissue in particular, these observations in combination with findings from previous studies suggest that the degradation of Ang II may counteract fibrotic-degenerative processes. In the murine lung ACE2 is localized at the apical pole of the respiratory epithelium of the bronchi. ACE2 levels in the kidney were the highest among the organs studied. There, the non-canonical key enzyme appears predominantly in the brush border of the proximal tubule, where it shows colocalization with Mas-1 and its canonical analog ACE. Mas is also expressed in mesangial cells and the muscularis of renal vessels. There it may inhibit ECM expansion in some renal diseases and positively influence renal blood flow.
The findings regarding the localization of the enzymes and receptors of the non-canonical RAAS support its postulated protective effects on various organ systems. In the future, its components could therefore play an important role in clinical therapeutic procedures, for example in arterial hypertension, heart failure, thrombotic-ischemic events, fibrotic degenerative and neoplastic organ disorders and, last but not least, infectious diseases such as Covid-19. Especially against the background of the recently overcome pandemic and now endemic SARS-CoV-2 situation, it is of immense importance to carry out further localization studies on ACE2 and to find out more precisely what significance the enzyme has in the context of this multisystem disease.
