Abstract (German)
Die vorliegende Dissertation untersucht die Wirkung von molekularem Wasserstoff und Sevofluran auf die Funktionalität humaner neutrophiler Granulozyten. Ziel war es, die Einflüsse beider Substanzen auf zentrale Mechanismen wie Migration, Bildung reaktiver Sauerstoffspezies, Aktivierung des Enzyms Myeloperoxidase sowie die Ausbildung neutrophiler extrazellulärer Fallen zu vergleichen. Diese Arbeit untersucht die Hypothese, ob H₂ eine stärkere hemmende Wirkung auf PMNs zeigt als Sevofluran.
Die Effekte auf die Migration und Funktionalität der PMNs wurden mittels Live-Cell-Imaging analysiert. Die ROS-Produktion, MPO-Aktivität und NETose wurden über fluoreszenzbasierte Marker gemessen und quantitativ ausgewertet. Darüber hinaus wurden mithilfe der Durchflusszytometrie die mediane Fluoreszenzintensität sowie die Expression der Oberflächenmoleküle CD11b, CD62L und CD66b untersucht, die essenziell für die Zelladhäsion und die Immunantwort sind.
Die Ergebnisse hinsichtlich der Migration sollten aufgrund der verwendeten Lockstoffkonzentration als nicht relevant erachtet werden. Bei der ROS-Produktion wurde bei mit H₂ begasten Zellen eine längere Zeit bis zum Erreichen der TmaxROS gemessen, während Zellen, die mit Sevofluran begast wurden, diese deutlich schneller erreichte. Die Zeit bis zum Erreichen der ET50MPO wurde durch Begasen der Zellen mit H₂ signifikant verzögert, während Sevofluran keinen vergleichbaren Effekt zeigte. Die Zeit bis zum Erreichen der ET50NETose wurde bei Zellen, die mit H₂ begast wurden im Vergleich zu mit Sevofluran begasten Zellen ebenfalls hinausgezögert. Die Durchflusszytometrie zeigte eine statistisch signifikante Verringerung der Medianen Fluoreszenzintensität der mit H2 begasten Zellen im Vergleich zu normoxischen Kontrollversuchen mit Aktivierung durch PMA, fMLP, TNF-α sowie LPS.
Molekularer Wasserstoff zeigte eine deutliche modulierende Wirkung auf neutrophile Granulozyten, insbesondere durch die Verzögerung der TmaxROS, der ET50MPO und ET50NETose. Die Kombination aus Live-Cell-Imaging und Durchflusszytometrie ermöglichte eine umfassende Analyse dieser Effekte. Diese Eigenschaften könnten therapeutisch genutzt werden, um eine überschießende Immunantwort zu regulieren. Sevofluran zeigte ebenfalls Effekte, jedoch weniger ausgeprägt und schneller einsetzend. Die Ergebnisse unterstreichen das Potenzial von H₂ als adjuvantes Therapeutikum bei entzündlichen Erkrankungen.

This dissertation investigates the effects of molecular hydrogen and sevoflurane on the functionality of human neutrophil granulocytes. The aim of this study was to compare the influence of both substances on key mechanisms such as migration, the production of reactive oxygen species, activation of the enzyme myeloperoxidase and the formation of neutrophil extracellular traps. This work examines the hypothesis that H₂ exhibits stronger inhibitory effects on PMNs than sevoflurane.
The effects on PMN migration and functionality were analyzed using live-cell imaging. ROS production, MPO activity, and NETosis were measured using fluorescence-based markers and were evaluated quantitatively. Additionally, the median fluorescence intensity of ROS production and the expression of surface molecules CD11b, CD62L and CD66b was examined via flow cytometry.
The results concerning the migration of the PMNs should not be considered relevant due to the concentration of the used chemoattractant. Cells exposed to molecular exhibited a longer time to reach TmaxROS, whereas cells exposed to sevoflurane reached this point faster. The amount of time needed to reach ET50MPO was significantly delayed in cells exposed to H₂, whereas sevoflurane showed no comparable effect. Regarding ET50NETosis, the amount of time needed to reach this marker was prolonged in cells exposed to H₂ compared to cells exposed to sevoflurane. Flow cytometry showed a significant reduction in the median fluorescence intensity of ROS production when cells were exposed to molecular hydrogen compared to normoxic controls. This effect was noticeable when activated with either LPS, TNF-α, fMLP or PMA.
Molecular hydrogen demonstrated a modulatory effect on neutrophil granulocytes, particularly by delaying ROS production, MPO activity, and NETosis. The combination of live-cell imaging and flow cytometry allowed for a comprehensive analysis. These properties could be therapeutically utilized to regulate excessive immune responses. Sevoflurane also exhibited effects, though less pronounced and with a quicker onset. These findings underscore H₂’s potential as an adjuvant therapeutic in inflammatory diseases.
