NGAL wird von verschiedenen Tumorentitäten exprimiert und wird als potentieller Marker für die Tumorprogression betrachtet. Durch eine Einteilung in N1- und N2-Granulozyten wurden zwei Populationen beschrieben, die auf gegenteilige Weise in das Tumorgeschehen eingreifen können. Die vorliegende Arbeit sollte klären, ob und inwiefern NGAL die Funktion und Differenzierung der Granulozyten beeinflusst.
Granulozyten gesunder Blutspenderinnen und Blutspender wurden in den Überständen der Tumorzelllinien MDA-MB-231 und BxPC3 inkubiert. Vergleichend wurde eine Inkubation der Granulozyten mit rekombinantem NGAL in ähnlicher Konzentration durchgeführt. Mittels Live Cell Imaging und Durchflusszytometrie wurden spezifische Oberflächenepitope, der oxidative Burst, die NETose, die MPO-Freisetzung sowie das Migrationsverhalten untersucht. Dabei zeigte sich, dass die Tumorzellüberstände eine starke Aktivierung der Granulozyten bewirkten. Im Gegensatz dazu ergab die Inkubation mit NGAL keine signifikanten Unterschiede in den analysierten Parametern im Vergleich zur Kontrolle. Es konnte eine Tendenz zur Differenzierung in einen N2-Phä-notyp beobachtet werden, jedoch ohne statistische Signifikanz. Die Ergebnisse der Migrationsanalysen waren uneinheitlich. Dies deutet darauf hin, dass NGAL keine klare Wirkung als Chemoattraktans auf neutrophile Granulozyten ausübt.
Daneben wurde die Überlebensfähigkeit der Granulozyten unter in vitro-Bedingungen analysiert. Nach 48 h zeigte sich ein großer Anteil der Zellen vital, welcher durch Zugabe von Q-VD-OPh noch vergrößert wurde. Allerdings führte der Caspase-Inhibitor auch zu deutlichen Veränderungen in der ROS-Produktion und der Expression von Oberflächenmarkern. Zusätzlich zeigte sich nach 48-stündiger Inkubation mit NGAL eine signifikante Verringerung der Apoptose der Granulozyten.
Zusammenfassend lassen sich folgende Ergebnisse festhalten: Innerhalb der ersten zwei Stunden der Inkubation zeigten die Granulozyten keine signifikanten funktionellen Veränderungen durch NGAL. Durch NGAL wurde keine eindeutige Differenzierung in einen TAN-Phänotypen induziert. Zudem wurde keine signifikante Wirkung von NGAL als Chemoattraktans nachgewiesen. Es ergab sich eine signifikante Zunahme im Überleben neutrophiler Granulozyten bei Inkubation mit NGAL im Vergleich zu IFN-β.

Q-VD-OPh als Caspase-Inhibitor steigerte das Zellüberleben der Granulozyten nicht signifikant, bewirkte jedoch teils signifikante Veränderungen in deren Funktion.

The aim of this study was to investigate the influence of the protein NGAL on neutrophil granulocytes, particularly with regard to their role in the tumor microenvironment. NGAL is expressed by various tumor entities and is considered a potential marker of tumor progression. The classification of neutrophils into N1 and N2 granulocytes has described two populations that can exert opposing effects on tumor progression. The present study aimed to determine whether and to what extent NGAL influences the function and differentiation of granulocytes.

Granulocytes from healthy blood donors were incubated in the supernatants of the tumor cell lines MDA-MB-231 and BxPC3. For comparison, granulocytes were also incubated with recombinant NGAL at a similar concentration. Live-cell imaging and flow cytometry were used to investigate specific surface epitopes, oxidative burst, NETosis, MPO release, and migratory behavior. The results showed that the tumor cell supernatants induced strong activation of the granulocytes. In contrast, incubation with NGAL did not result in significant differences in the analyzed parameters compared with the control. A tendency toward differentiation into an N2 phenotype was observed, although this did not reach statistical significance. The results of the migration analyses were inconsistent. This suggests that NGAL does not exert a clear chemoattractant effect on neutrophil granulocytes.

In addition, the viability of the granulocytes under in vitro conditions was analyzed. After 48 h, a large proportion of the cells remained viable, and this proportion was further increased by the addition of Q-VD-OPh. However, the caspase inhibitor also caused pronounced changes in ROS production and the expression of surface markers. Furthermore, incubation with NGAL for 48 h resulted in a significant reduction in granulocyte apoptosis.

In summary, the following findings can be stated: During the first two hours of incubation, NGAL did not induce any significant functional changes in the granulocytes. NGAL did not induce a clear differentiation into a TAN phenotype. Moreover, no significant effect of NGAL as a chemoattractant was demonstrated. Incubation with NGAL resulted in a significant increase in neutrophil survival compared with IFN-β.

Q-VD-OPh, as a caspase inhibitor, did not significantly increase granulocyte survival but caused, in some cases, significant changes in granulocyte function.
