Abstract (German)

Im Mittelpunkt dieser Arbeit stehen mit den humanen Praeadipozyten die fibroblastenartigen Vorläuferzellen reifer Adipozyten, in welchen mit der Aromatase (Cytochrom P-450Arom) das Schlüsselenzym der Estrogenbiosynthese lokalisiert ist. Im Mittelpunkt dieser Arbeit stehen mit den humanen Praeadipozyten
die fibroblastenartigen Vorläuferzellen reifer Adipozyten, in welchen
mit der Aromatase (Cytochrom P-450Arom) das Schlüsselenzym der Estrogenbiosynthese
lokalisiert ist. Mit der pharmakologischen Klassifizierung eines Purinozeptors
auf humanen Praeadipozyten, der den potenzierenden Effekt von ATP bei
der Induktion von Aromatase durch Cortisol und PDGF (Platelet-Derived
Growth Factor) vermittelt, als Ausgangspunkt konnte die cDNA für die P2X1-Rezeptoruntereinheit
über den gesamten kodierenden Bereich nachgewiesen werden. Versuche
zur weiteren Charakterisierung des Purinozeptors auf molekularer Ebene
mittels Yeast Two Hybrid Assays scheiterten.
Unabhängig hiervon wurden humane Praeadipozyten als Expressionsort von
Komplementfaktoren des klassischen Aktivierungsweges identifiziert.
Die Faktoren C1s, C1r, C2 und C4 konnten auf mRNA-Ebene nachgewiesen
werden, während der Komplementfaktor C1q nicht gefunden wurde.
Die Serinproteasen C1s und C1r konnten zudem durch Western-Blot-Technik
sowohl im Zellysat als auch in sezernierter Form im zugehörigen
Zellkulturüberstand nachgewiesen werden, was ihre funktionelle Expression
nahelegt. Zur Untersuchung der differenzierungsabhängigen Expression
der in humanen Praeadipozyten nachweisbaren Komplementfaktoren wurden
semiquantitative RT-PCRs etabliert. Die Resultate belegen einen deutlichen
Rückgang der Genexpression der Komplementfaktoren C1s und C1r während
der Differenzierung primärkultivierter humaner Praeadipozyten, der bei
der Untersuchung von entsprechenden Zellysaten durch Western-Blot-Technik
Bestätigung fand. Durch Western-Blot-Technik konnte zudem gezeigt werden,
dass sich der dedifferenzierende Effekt von TNF (Tumor-Nekrose-Faktor)
auf reife Adipozyten in einer Steigerung der Expressions- und Sekretionsrate
für die Komplementfaktoren C1s und C1r niederschlägt.

Human preadipocytes, morphologically similar to fibroblasts, are the precursor cells of mature adipocytes and they synthesize cytochrome-p450-dependent aromatase, the key enzyme of estrogen biosynthesis. Human preadipocytes, morphologically similar to fibroblasts, are the
precursor cells of mature adipocytes and they synthesize cytochrome-p450-dependent
aromatase, the key enzyme of estrogen biosynthesis. Induction of aromatase
activity in vitro is observed in the presence of cortisol and PDGF
(platelet-derived growth factor) and is further increased by addition
of ATP. Starting from the pharmacological profile of the involved purinoceptor
a complementary DNA encoding the P2X1 receptor subunit was isolated from
human preadipocytes. Attempts to characterize the whole receptor complex
by yeast two-hybrid assays failed.
Further on the synthesis of several components of the classical complement
pathway by human preadipocytes was demonstrated. The components C1s, C1r,
C2 and C4 were detected by RT-PCR technique, whereas the complement factor
C1q could not be found. The serine proteases C1s and C1r could also be
detected by western blot analysis both in cell lysates and after secretion
in the culture medium. To monitor the differentiation dependent expression
of the detectable complement components a semiquantitative RT-PCR was
established. The results showed a marked decrease in the gene expression
of C1s and C1r during differentiation of human preadipocytes in vitro.
These observations were confirmed by western blot analysis of appropriate
cell lysates. Besides TNF-mediated reversal of adipocyte differentiation
could be demonstrated to be associated with an increase in the levels of C1s
and C1r proteins both in the cell lysate and in the culture medium.
