Der Einsatz von DNA-Impfstoffen zur Prävention von AIDS ist gegenwärtig durch Sicherheitsmängel und geringe Immunogenität der derzeit verwendeten Expressionsvektoren limitiert.
Ziel war es, anhand unterschiedlicher SIV-gag DNA Konstrukte die komplexe lentivirale Regulation zu verstehen und sie in Hinblick auf eine SIV-gag spezifische DNA-Vakzine zu umgehen.
Die Immunogenität der DNA-Vakzinekandidaten wurde im Balb/C-Mausmodell ausgetestet. Die stärkste Immunantwort wurde durch im. Immunisierung mit einer kodonoptimierten syngag Vakzine erzielt. Diese zeigte eine TH1-Polarisierung, mit hohen spezifischen IgG2a Titern, IFNg Sekretion und eine starker ZTL-Antwort.
Wirksamkeit / Schutz kodonoptimierter DNA-Vektoren wurden im SHIV-Makakenmodell analysiert. Optimierte DNA-Impfstoffkandidaten wurden in Kombination mit rekominanten viralen Vektoren verabreicht.
In einer Studie wurde zusätzlich der Einfluß von Adjuvantien (IL-12 u. IL-18) beurteilt. Nach iv. Challenge mit dem pathogenen Virusisolat SHIV 89.6P konnte in keiner der untersuchten Gruppen ein vollständiger Schutz erzielt werden. Allerdings wurde die initiale Viruslast in allen spezifisch immunisierten Gruppen stark reduziert,
ein niedrigerer 'Set Point' erzielt und die CD4-Zellzahl konstant gehalten. Die Gruppen mit Interleukingabe zeigten eine erhöhte Induktion von ZTLs und T-Helferzellen, aber auch schlechteren Schutz vor Infektion.
In einer zweiten Studie wurde die Wirksamkeit bezüglich eines rektalen Challenge sowie der Einfluß unterschiedlicher Immunisierungsrouten (im./id. vs. nasal) analysiert. 3 der 4 Tiere waren durch die nasale Immunisierung geschützt (vgl. im./id.:1/4).
Analog zu den kodonoptimierten DNA-Vektoren für die Primaten-Studien wurden im Rahmen dieser Arbeit auch DNA-Vektoren für humane Studien entwickelt, die auf einem endemisch vorkommenden HIV Primärisolat 97CN54 basieren. Diese werden Ende dieses Jahres in klinischen Studien an freiwilligen, HIV-negativen Probanden getestet.

Translation of the abstract (English)

The efficient application of DNA vaccines to prevent AIDS is limited by the lack of safety as well as weak immunogenicity of the currently used vector systems.Therefore the primary aim was a better understanding of the complex lentiviral regulation. Accordingly, different DNA reporter constructs were developed aiming at a SIV-gag DNA vaccine without regulatory limitations.
The immunogenicity of the different DNA vaccine candidates was evaluated in a Balb/C mouse model. The strongest immune response was attained by im. immunisation with a codon-optimized syngag vaccine inducing a TH-1 polarisation characterized by high specific IgG2a titers, significant IFNg -secretion and a strong CTL response.The SHIV macaque model was used to test efficacy and protection of codon optimized DNA vectors. The respective candidates were applied in a prime-boost regime in combination with recombinant viral vectors. In one study we additionally analysed the influence of adjuvans (IL-12 and IL-18) on efficiency. After intravenous challenge with the highly pathogenic isolate SHIV 89.6P non of the animals were completely protected usually indicated by erradification of the virus. Nevertheless all specific immunized monkeys showed a dramatically reduction of the initial virusload and of the 'set-point' of the infection. Further, the CD4 cell counts were stabil over time. The groups of monkeys, that received interleucines developed a higher CTL and T-Helper activity, silmutaneously the protection levels were reduced.
In a second study we analysed protection upon rectal challenge and the efficacy of different application modes (im./id. vs. nasal). 3 of 4 animals were protected after nasal and only 1 after im./id. immunization.
Analogues to the codon optimized DNA vectors developed for the non human primate studies we generated DNA vaccines based on the primary HIV isolate 97CN54. These vectors will be testet in clinical studies on HIV negativ volunteers at the end of this year.
