Abstract (German)

In der vorliegenden Arbeit wurde zunächst die Zytotoxizität der beiden Basis-monomere zahnärztlicher Komposite, Bisphenol A-Glycidyldimethacrylat (Bis-GMA) und Urethandimethacrylat (UDMA), des wichtigsten Comonomers Triethylenglycoldimethacrylat (TEGDMA), der beiden Monomere Methylmethacrylat (MMA) und 2-Hydroxyethylmethacrylat (HEMA) und der Ausgangssubstanzen zur Synthese des Bis-GMA, ...

In der vorliegenden Arbeit wurde zunächst die Zytotoxizität der beiden Basis-monomere zahnärztlicher Komposite, Bisphenol A-Glycidyldimethacrylat (Bis-GMA) und Urethandimethacrylat (UDMA), des wichtigsten Comonomers Triethylenglycoldimethacrylat (TEGDMA), der beiden Monomere Methylmethacrylat (MMA) und 2-Hydroxyethylmethacrylat (HEMA) und der Ausgangssubstanzen zur Synthese des Bis-GMA, Glycidylmethacrylat (GMA) und Bisphenol A (BPA) in V79-Zellen ermittelt. Mutagene Effekte der Substanzen wurden mit zwei verschiedenen Genmutationstests geprüft und quantifiziert. Der Salmonella/Mikrosomen-Test (Ames-Test) erkennt Basenpaarsubstitutionen und frameshift-Mutationen. Der V79/HPRT Test zeigt Punktmutationen, Insertionen und große Deletionen an.
Die Basismonomere Bisphenol A-Glycidyldimethacrylat (Bis-GMA) und Urethandimethacrylat (UDMA), das Monomer 2-Hydroxyethylmethacrylat (HEMA) und Bisphenol A nicht mutagen. GMA und TEGDMA hingegen induzierten Genmutationen bei direkter Prüfung. GMA bewirkte einen 2-3-fachen Anstieg der Mutantenzahlen in S. typhimurium TA97a und TA102, im Stamm TA100 stiegen die Mutantenzahlen sogar 8-fach. GMA war auch in V79-Zellen mutagen. Dieser Effekt wurde mit S9 vollständig aufgehoben. TEGDMA war im V79/HPRT-Test bei direkter Prüfung mutagen. In Anwesenheit von S9 wurde es wahrscheinlich durch eine Esteraseaktivität zu Methacrylsäure und Triethylenglycol gespalten.
Ein Deletionsscreening kodierender Exonsequenzen des hprt-Gens mit Polymerase Chain Reaction zeigte das TEGDMA-induzierte Mutationsspektrum. In 19 von insgesamt 25 analysierten Mutanten der Zellinie V79-UL waren keine Exonsequenzen nachzuweisen (Totaldeletionen), 5 Mutanten wiesen Partialdeletionen auf und lediglich ein Zellklon besaß das Wildtypmuster des hprt-Gens. Es wurden in keiner der spontan entstandenen V79-Mutanten Deletionen gefunden. Es ist zu vermuten, daß TEGDMA im Sinne einer Michael-Addition mit Purinbasen doppelsträngiger DNA reagierte und crosslinks ausbildete.

Translation of the abstract (English)

In the present study the cytotoxicity of two major monomers of dental composites, bisphenol A glycidyl dimethacrylate (Bis-GMA) and urethane dimethacrylate (UDMA) as well as the effects of the most important comonomer triethylene glycoldimethacrylate (TEGDMA), of the two monomers methyl methacrylate (MMA) and 2-hydroxyethyl methacrylate (HEMA) and of two chemicals used to synthesize Bis-GMA, ...

In the present study the cytotoxicity of two major monomers of dental composites, bisphenol A glycidyl dimethacrylate (Bis-GMA) and urethane dimethacrylate (UDMA) as well as the effects of the most important comonomer triethylene glycoldimethacrylate (TEGDMA), of the two monomers methyl methacrylate (MMA) and 2-hydroxyethyl methacrylate (HEMA) and of two chemicals used to synthesize Bis-GMA, glycidyl methacrylate (GMA) and bisphenol A (BPA), was determined in V79 cells. Mutagenic effects of the substances were then analyzed and quantified in vitro using two different gene mutation assays. The Salmonella/microsome test (Ames-Test) indicated base pair substitutions and frameshift-mutations. The V79/HPRT assay indicates induced point mutations, insertions and large deletions.
The major monomers bisphenol A glycidyl dimethacrylate (Bis-GMA) and urethane dimethacrylate (UDMA), the monomer 2-hydroxyethyl methacrylate (HEMA) and bisphenol A tested non mutagenic. GMA and TEGDMA induced gene mutations after direct testing. GMA caused a 2-3-fold increase of mutant numbers in S. typhimurium TA97a and TA102, and mutant numbers increased 8-fold in strain TA100. GMA was also tested mutagenic in V79 cells. This effect was completely absent in the presence of S9. TEGDMA was tested mutagenic in the V79/HPRT assay after direct testing. In the presence of S9 it was probably cleaved by a esterase activity creating methacrylic acid and triethylene glycol.
Deletion screening of exon sequences of the hprt gene by polymerase chain reaction gave insight into the TEGDMA-induced mutation spectra. No exon sequences were detected in 19 of a total of 25 mutants of the cell line V79-UL (total deletions), 5 mutants showed partial deletions and only one cell clone showed a wildtype pattern of the hprt gene. No deletions were found in spontaneously originated V79 mutants. It is assumed, that TEGDMA reacted with purine bases of double-stranded DNA via Michael addition resulting in crosslinks.
