Zusammenfassung (Deutsch)
Das für P. furiosus etablierte genetische System wurde verwendet, um verschiedene Komponenten des Transkriptionsapparates und des Stoffwechsels von P. furiosus zu analysieren und im Zuge dieser Anwendungen das Spektrum der verfügbaren genetischen Methoden zu erweitern.
Mit Hilfe eines Shuttle-Vektor-Systems wurden mutierte RpoA"-Untereinheiten der RNAP in P. furiosus produziert. Dadurch wurde bewiesen, dass es bei P. furiosus möglich ist auch defekte, mutierte Proteine zu produzieren und durch die simultan vorhandenen wt Proteine die Vitalität der Zellen zu gewährleisten. Assemblierte RNAPs mit integrierter, modifizierter RpoA"-Untereinheit konnten aus den Zellen isoliert und für Struktur-Funktions-Analysen eingesetzt werden. Diese bestätigten die Inaktivierung der RNAP durch die Deletion der Tip-Region des Trigger Loops bzw. die reduzierte Aktivität aufgrund einer Aminosäuresubstitution an Position 87.
Bei der Charakterisierung des putativen Transkriptionsregulators TrmBL2 wurde ein P. furiosus-Deletionsstamm generiert, welcher für Wachstums- und Genexpressionsanalysen eingesetzt wurde. Auch erfolgten in vitro Untersuchungen mit TrmBL2, welches über die Integration einer chromosomalen Proteintagsequenz direkt aus P. furiosus gereinigt wurde. Die erhaltenen Daten erbrachten keinen Hinweis auf eine Funktion von TrmBL2 als Transkriptionsregulator des Zuckerstoffwechsels. Jedoch besitzt TrmBL2 eine Vielzahl an chromosomaler DNA-Bindestellen und auch in vitro starke DNA-Bindeaktivität. Da zudem eine hohe Thermostabilität von TrmBL2 unter Hitzeschock-Bedingungen analysiert wurde, ist nach jetzigem Kenntnisstand davon auszugehen, dass TrmBL2 dem Schutz freier chromosomaler DNA-Bereiche vor Hitzeschädigungen dient.
Da durch einen einzelnen Aminosäureaustausch eine chimäre Rp5H-Untereinheit erfolgreich für Komplementation in einem Rpb5-Hefedeletionsstamm aktiviert werden konnte, wurde ein entsprechender Komplementationsansatz auch in P. furiosus durchgeführt. Durch das genetische System wurde die endogene rpoH-Gensequenz durch eine Variante mit einer Basensubstitution, welche zum Aminosäureaustausch E62K führt, ersetzt. Hingegen ein chimäres Rp5H-Konstrukt aus N-terminalem Anteil von rpb5 aus Hefe und rpoH aus P. furiosus als C-terminaler Anteil konnte anstatt des wt rpoH-Gens nicht etabliert werden. Sowohl der Vergleich der Vitalität von wt P. furiosus und des RpoH E62K-Expressionsstammes, als auch die mit isolierter RpoH-E62K-RNAP durchgeführten in vitro Analysen ergaben keine signifikante Veränderung des Wachstums bzw. der Aktivität der assemblierten RNAPs. Demnach hat der Aminosäureaustausch trotz einer Ladungsumkehrung an dieser Position keinen Einfluss auf die Aktivität der RNAP; mögliche Auswirkungen auf Proteinwechselwirkungen bleiben zu klären.
Zur Optimierung des Chitinabbaus von P. furiosus wurden die im Genom von P. furiosus getrennt vorliegenden kodierenden Sequenzen zweier Chitinase-Einheiten durch eine Basendeletion zu einer kodierenden Sequenz für eine fusionierte Chitinase kombiniert. Wachstumsanalysen und Messungen des Energiegehalts und des Stoffumsatzes der Zellen ergaben ein gesteigertes Wachstum und sichtbaren Abbau des Chitins durch den mutierten P. furiosus-Stamm. Die Ergebnisse bekräftigen die Annahme, dass bei P. furiosus das Gen der Chitinase ursprünglich als eine durchgängige Basensequenz vorlag und erst vor jüngerer Zeit durch die Insertion einer Base zwei getrennte Chitinase-Einheiten erzeugt wurden.

The genetic system developed for P. furiosus was used to analyse various elements of the transcription apparatus as well as the metabolism of P. furiosus. In the course of their application the range of available genetic tools was expanded.
The shuttle vector system was used to express mutated RpoA" subunits of the P. furiosus RNAP. Therefore, even defect and mutated proteins can be expressed in P. furiosus as proven with this study. The simultaneously present wildtype version of the affected protein ensures viability of the cells. Assembled RNAPs with integrated, modified RpoA" subunits could be extracted from the cells and could be used for structure-function analysis. Thereby the inactivation and the reduced activity of the RNAP, caused by the deletion of the tip region of the trigger loop and an amino acid substitution at position 87, were confirmed.
To characterize the putative transcription regulative protein TrmBL2 a P. furiosus deletion strain was established and used for comparative studies of its growth behaviour and gene expression values. In a second P. furiosus strain the chromosomal gene sequence of trmBL2 has been modified by adding a sequence for a protein tag. TrmBL2 purified from the cells was used for in vitro studies of TrmBL2 activity. The generated data gave no hint of a functional participation of TrmBL2 in transcriptional regulation events of sugar metabolism. However, a multitude of DNA-binding sites of TrmBL2 on chromatin of P. furiosus could be defined just as an high DNA-binding activity of TrmBL2 in vitro. Additionally, a high thermal stability of TrmBL2 under heat shock condition was measured. With current state of the art, it can be assumed that TrmBL2 provides the maintenance and prevention of free chromosomal DNA sections from harmful influences of high temperatures.
A single amino acid substitution leads to activation of a chimeric Rp5H subunit for successful complementation in a Rpb5 deficient yeast strain. Therefore, a respective complementation assay was performed in P. furiosus. Using the genetic system, the endogenous rpoH gene sequence was replaced by a version containing a DNA point mutation, leading to the exchange of the glutamate at position 62 against lysine in the expressed protein. In contrast, a chimeric Rp5H construct containing the N-terminal part of rpb5 from yeast and the rpoH gene sequence of P. furiosus as C-terminal part could not be introduced to P. furiosus to replace the wildtype rpoH gene. Both the comparison of the vitality of P. furiosus wildtype and RpoH E62K expression strain as well as the in vitro assays performed with isolated RpoH E62K RNAP indicated no significant changes in the growth behaviour of the strain and the activity of the assembled RNAPs. According to that, despite the inversion of the charge at this position, the exchange of the amino acid has no severe influence to the activity of the RNAP. Possible implications to protein interactions remain to be resolved.
To optimise the chitin degradation of P. furiosus the coding sequences of the chitinase, appearing as two separated gene frames in the genome of P. furiosus, were fused to a single coding sequence by deletion of a DNA base. This leads to expression of a chitinase comparable to the enzyme of T. kodakarensis. Growth analysis as well as the measurement of the energy content and the metabolic rate of the cells resulted in increased growth and distinctly and visibly degradation of chitin by the mutated P. furiosus strain. These results confirm the assumption that the gene sequence of the chitinase in P. furiosus originally existed as one continuous gene sequence and was just separated into two chitinase units by the insertion of a DNA base in the recent past.
