Das Guaninnukleotid-bindende Protein Ras ist als molekularer Schalter in zellulären Signaltransduktionswegen involviert, die zu Proliferation, Differenzierung oder Apoptose von Zellen führen. Dabei wechselt Ras zwischen einer inaktiven, GDP-gebundenen und einer aktiven, GTP-gebundenen Form. In der aktiven Kon-formation kann Ras über die Schalter I-Region mit hoher Affinität verschiedene Effektoren wie die Raf-Kinase oder RalGDS binden. Dadurch werden die Signale weitergeleitet und die entsprechenden Zellantworten bewirkt.
In dieser Arbeit wurden Ras(wt), die onkogene Mutante Ras(G12V) sowie einige Schalter I-Mutanten in Lösung untersucht. Im Gegensatz zu den Ergebnissen aus der Röntgenkristallographie zeigen die hier durchgeführten Mn2+-EPR- und 31P-NMR-Untersuchungen an der inaktiven und aktiven Form von Ras in Lösung, dass sowohl die Koordination des im aktiven Zentrum gebundenen Metallions, als auch die der Phosphate des gebundenen Nukleotids in den unterschiedlichen Ras-Varianten verschieden sind. Aber auch innerhalb den einzelnen Ras-Varianten treten unterschiedliche Konformationen auf.
Ras(wt) kann in der aktiven Konformation in zwei konformationellen Zuständen vorliegen (Zustand 1 und 2), die sich im chemischen Austausch miteinander befinden. Der Zustand 2 wird durch die Bindung von Effektoren stabilisiert und sollte der Effektor-gebundenen Konformation ähnlich sein. Das Gleichgewicht zwischen den beiden Zuständen wird durch die GTP-Analoga GppNHp und GppCH2p, sowie auch durch Mutationen in der Schalter I-Region von Ras zum Zustand 1 verschoben. Auf Kalorimetrie und Fluoreszenz basierende Bindungsstudien dieser Arbeit zeigen, dass diese konformationellen Gleich-gewichte einen direkten Einfluss auf die Ras-Effektorwechselwirkung

The guanine nucleotide binding protein Ras functions as a molecular switch. Ras is involved in cellular signal transduction pathways leading to proliferation, differentiation or apoptosis of cells. By doing this, Ras cycles between an inactive, GDP-bound state and an active GTP-bound state. In the active conformation Ras is able to bind different effectors like Raf-Kinase or RalGDS with high affinity by interacting with its switch I-region. By that signals can be transduced and corresponding cell answers can be affected.
In this work Ras(wt), the oncogenic mutant Ras(G12V) and different switch I-mutants of Ras were investigated in solution. In contrast to the results obtained from X-ray-structure here it is shown by Mn2+-EPR- and 31P-NMR-measurements, that the coordination of both, the bound metal ion and the phosphates of the nucleotide are different in variant active and inactive Ras-nucleotide complexes in solution. Also differerent variants itself exist in more than one conformation.
Ras exists in two different states if bound to triphosphate nucleoside defined as state 1 and state 2, which are in chemical exchange. State 2 should be very similar to the effector-bound state and becomes stabilized if Ras is bound to effectors. Different mutations in switch I-region of Ras or binding to the GTP-analogs GppNHp or GppCH2p shift the equilibrium between the two states towards state 1. Binding studies using calometric and fluorescence-based methods show that these conformational equilibria influence directly the interaction between Ras and its effectors.
