The role and the importance of hyaluronic acid and hyaluronidases in physiological and
pathophysiological processes are largely misunderstood, so that selective and
potent hyaluronidase inhibitors are required. As such compounds are not known so
far, the goal of this project was to identify and to synthesise lead-like
compounds, to optimise the structures and to study the structure-activity
relationships. Therefore, we applied the following strategies: structurally
different oligosaccharides with varying degrees of sulfation were studied for
inhibitory activity on the bovine testicular enzyme, the bacterial hyaluronidase
and the hyaluronidase from bee venom. Furthermore, a series of substituted
flavone derivatives was synthesised and investigated for inhibition of the bovin
e testicular and the bacterial enzyme. For the de novo design of enzyme
inhibitors we followed a structure-based approach. Initially, a model of hylB4755
hyaluronidase was constructed and subsequent LUDI calculation based on this
model resulted in different hits. From this proposals promising compounds were
selected, synthesised and their inhibitory effect on the hyaluronidases were
examined. The aforementioned approaches led to the first hyaluronidase inhibitors
active in the low micromolar range, which can be used as leads for further
structural optimisation.

Die (patho)physiologische Rolle von Hyaluronsäure und deren Abbauprodukte sowie
der Hyaluronidasen ist weitgehend unverstanden, u. a. weil bisher keine potenten
und selektiven Hyaluronidase-Inhibitoren zur Verfügung stehen. Das Ziel der
Dissertationsarbeit war die Identifizierung und die Synthese von Leitstrukturen,
ihre Optimierung sowie die Erstellung von Struktur-Wirkungs-Beziehungen. Dabei
wurden unterschiedliche Strategien verfolgt: Untersuchung von unterschiedlicher
Oligosaccharide mit variierendem Sulfatierungsgrad als Inhibitoren der bovinen
testikulären Hyaluronidase (BTH), der bakteriellen Hyaluronidase (hylB4755) und
der Bienengift-Hyaluronidase (BVH). Des weiteren wurde eine Reihe von substituierten
Flavonen synthetisiert und auf ihre Wirksamkeit gegenüber BTH and hylB4755 untersucht.
Als Schwerpunkt dieser Arbeit wurde ein Struktur-basierter de novo Ansatz
verfolgt; dabei wurde zuerst ein Modell der hylB4755-Hyaluronidase konstruiert.
Anschließende LUDI-Berechnungen führten zu einer Reihe von unterschiedlichen
Strukturvorschlägen, aus denen vielversprechende Verbindungen ausgesucht, synthetisiert
und pharmakologisch untersucht wurden.Im Rahmen dieser Arbeit war es möglich
die ersten Hyaluronidase Inhibitoren mit Wirkung im micromolaren Bereich zu
erhalten, die nun als Leitstrukturen zur weiteren Optimierung verwendet werden
können.
