Abstract (German)
Die grüne Kugelalge Volvox carteri zählt mit nur zwei Zelltypen � somatischen und reproduktiven � zu den einfachsten Modellorganismen für das Studium der Zelldifferenzierung. Diese Differenzierung wird von drei Genloci � gls, lag und regA � gesteuert. In dieser Arbeit steht das regA-Gen im Vordergrund der Untersuchungen. Seine Transkription und Expression bestimmen den zellspezifischen und den zeitlichen Verlauf der Differenzierung. Die regulatorische Wirkung des RegA-Proteins und die Regulation des regA-Gens selbst sind die entscheidenden Aspekte der Differenzierung von somatischen Zellen.
Untersuchungen in dieser Arbeit zielten darauf ab, erstens die von RegA ausgeübte Genkontrolle und zweitens die Regulation der Translation des regA-Gens selbst besser zu verstehen. Mit diesen Vorgaben wurden (1) das TATA-Box-Bindeprotein (TBP) von V. carteri charakterisiert und rekombinant exprimiert, (2) ausführliche Versuche zur heterologen Expression und Reinigung von RegA und spezifischen Antiseren unternommen und (3) Kontrollelemente der Translation in der 5�NTR von regA identifiziert.
1. Das tbp-Gen wurde aus V. carteri isoliert und rekombinant exprimiert. Das single-copy Gen enthält sechs Exons und fünf Introns und kodiert für ein Protein mit 340 AS-Resten. Ein Vergleich mit bekannten TBP-Proteinen zeigte, daß die Kernregion hochkonserviert ist. Als Novum gegenüber tierischen und pflanzlichen TBPs trägt es eine 120 AS-Reste lange C-terminale Domäne mit prominenten Glutamin-, Alanin- und Prolin-Clustern, die auf eine regulatorische Funktion (ähnlich den N-terminalen Domänen tierischer TBPs) hindeuten. Die heterologe Expression von tbp cDNA im E. coli-System und die Reinigung von rekombinantem TBP verlief problemlos.
2. Im E. coli-System gelang die Expression eines 393 AS-Reste langen N-terminalen Fragments des RegA-Proteins. Ferner wurden mit dem Baculovirus-Insektenzell-System geringe Mengen von RegA und eine Serie von C-terminal verkürzten Fragmenten gewonnen, die auf unvollständige Translation von regA zurückgeführt wurden. Gegen beide Proteinfraktionen wurden spezifische Antikörper hergestellt und gereinigt. Diese waren genügend empfindlich, um rekombinantes RegA und das RegA-Protein in Überexpressions-Stämmen, nicht aber im WT-Volvox-Stamm, zu detektieren.
3. Das regA-Gen enthält vier nicht-kodierende Exons in der 5�NTR mit acht ATG-Tripletts. Die Bedeutung der ATG-Tripletts für die Translation des regA-Transkripts wurde mittels Komplementation mit ATG nach TGG-Mutationen in einem Volvox-Stamm, der ein defektes regA-Gen enthält, getestet. Der Erfolg eines Komplementations-Experiments wurde durch Wechsel des Phänotyps festgestellt. Aus diesen Analysen resultierten folgende Ergebnisse:
ATG9 oder ATG10 können als Translationsstart für funktionelles RegA dienen.
Gezielte Substitutionen von ATG1 bis ATG7 durch TGG resultierten alle in reduzierten Komplementationsraten. Massive Effekte zeigten aber nur die ATG1- und ATG2-Mutationen.
Die Einführung von zwei nonsense Codons in den längsten Leserahmen der 5�NTR (ORF2) resultierte in einer weiteren Reduktion der Komplementationsrate. Das weist ORF2 eine besondere Bedeutung in der regA-Translationskontrolle zu.
Es konnte außerdem ein eigenes orf2-Transkript nachgewiesen werden, daß Intron 2 im Vergleich zur regA-mRNA noch enthält. Im Vergleich mit den Sequenzen aus drei V. carteri-Vertretern weist die ORF2-Region eine erstaunliche Konservierung der DNA-Sequenzen auf. Dies ist ein starkes Indiz für die Signifikanz der Nukleotidsequenze dieser Region.
Eine computergestützte Analyse ergab eine komplexe Haarnadelstruktur im 5�Breich von ORF2, wie sie in einigen anderen Systemen beim ribosome shunting auftritt. Außerdem tritt am Ende von Exon 3 ein zur 18S rRNA komplementärer Bereich auf, der als Ribosomenbindestelle beim ribosome shunting dienen könnte.
Das postulierte Translationskontroll-Modell des ribosome shunting kann alle Ergebnisse dieser Arbeit erklären.

Translation of the abstract (English)
The green alga Volvox carteri with only two cell types � somatic and reproductive cells � is one of the simplest models for studying cell differentiation. Its differentiation is controlled by three gene loci � gls, lag and regA. In this work the regA gene was examined. Its transcription and expression determine the cell type specific and temporal way of differentiation. The regulatory effects of RegA and the regulation of the regA gene itself are the most important aspects of the differentiation of somatic cells.
The aims of this work were to get a better understanding of the RegA mediated gene control and of the translational regulation of the regA gene itself. For this purpose (1) the TATA box binding protein (TBP) of V. carteri was characterized and recombinantly expressed, (2) RegA was expressed and specific antibodies were raised against and (3) translation control elements were identified in the 5�UTR of regA.
1. The tbp gene from V. carteri was isolated and recombinantly expressed. The single-copy gene consists of six exons and five introns and codes for a protein of 340 aa. The comparison with known TBP proteins shows that the core region is highly conserved, but in contrast, it contains a C-terminal elongation of 120 aa. The glutamine, alanine and proline stretches suggest a regulatory function of this C-terminal region (like the N-terminal region in animals). The heterologous expression of the tbp cDNA in E. coli and the isolation of recombinant TBP were successful.
2. A N-terminal 393 aa fragment of RegA was expressed in E. coli. The whole RegA protein could only be expressed in small quantities in the baculovirus expression system. Most of the isolated RegA was shortened because the translation was incomplete. Against both protein fractions antibodies were raised. The antibodies could detect recombinantly expressed RegA and RegA in a V. carteri overexpression strain, but not in wt V. carteri.
3. The regA gene has four non-coding exons with eight ATGs in its 5�UTR. The importance of these ATGs for the translation of the regA transcript was examined by complementation with ATG to TGG mutations in a Volvox strain, which has a defect regA gene. The following results were obtained:
ATG9 and ATG10 are translation starts for functional RegA.
The substitutions of ATG1 to ATG7 with TGG results in a reduction of the complementation rate. Only the ATG1 and ATG2 mutations show strong effects.
The introduction of two nonsense codons in the largest reading frame of the 5�UTR (ORF2) results in an additional reduction. This shows the importance of ORF2 for the regA translational control.
An own orf2 transcript could be detected. In contrast to the complete regA mRNA it contains the intron 2. When compared with the sequences from three other V. carteri strains, the ORF2 sequence shows a remarkable conservation. This is a strong hint for the significance of the nucleotide sequence of the region.
A computational analysis shows a hairpin structure in the 5� region of ORF2 similar to some other systems with ribosome shunting. Besides there is a complementarity to the 18S rRNA at the end of exon 3 that could serve as a ribosome binding site during ribosome shunting.
The postulated model of translational control - the ribosome shunting - can explain all the results of this work.
