Abstract (English)

Phot proteins are homologs of the blue-light receptor phototropin. In this dissertation it was reported that a comparative study of the photocycles of the isolated, light-sensitive domains LOV1 and LOV2 from Chlamydomonas reinhardtii phot protein, as well as the construct LOV1+2 containing both domains. Transient absorption measurements revealed a short lifetime of the LOV2-wt triplet state (500ns), but a long lifetime (287µs) of the triplet in the mutant LOV2-C250S, in which the reactive cysteine is replaced by serine. For LOV1, in comparison, corresponding numbers of 800ns and 4µs for the two conformers in LOV1-wt, and 27µs for LOV1-C57S that has been reported by Tilman Kottke. The triplet decay kinetics in the mixed domains LOV1+2-wt, LOV1+2-C57S, and LOV1+2-C250S can be analyzed as the superposition of the behaviour of the corresponding single domains. The situation is different for the slow, thermal reaction of the photoadduct back to the dark form. Whereas the individual domains LOV1 and LOV2 show two decay components, the mixed domains LOV1+2-C57S and LOV1+2-C250S both show only a single component. The interaction of the two domains does therefore not manifest itself during the lifetime of the triplet states, but changes the decay behaviour of the adduct states. About the mechanism of the adduct formation, we prefer to the neutral radical-pair schemes proposed by Kay et al. according to the simple study on the mutant LOV1-C57G. In order to simulate the photocycle of each LOV domain, several possible models were tried with globle fit, which might help us deeply understand the system of the phot in Chlamydomonas reinhardtii.

Phot-Proteine sind die Homologe des Blaulicht-Rezeptors Phototropin. Diese Doktorarbeit berichtet über die Vergleichsstudie der Photozyklen der isolierten, lichtempfindlichen Domänen LOV1 und LOV2 des phot-Proteins aus Chlamydomonas reinhardtii und über den Aufbau von LOV1+2, die beide Domänen beinhaltet. Transient-Absorption-Messungen zeigten eine kurze Lebensdauer des Triplettzustandes von LOV2-wt (500 ns), aber eine lange Lebensdauer des Triplettzustandes des LOV2-C250S-Mutantes (287 ms), in dem das reaktive Cystein durch das Serin ersetzt wurde. Über das Verhalten von LOV1 wurde von Tilman Kottke berichtet. Zum Vergleich die Daten für die zwei Konformere von LOV1-wt betragen 800 ns und 4 ms und für den LOV1-C57S-Mutanten 27 ms. Die Kinetik des Triplettabklingen der gemischten LOV1+2-wt, LOV1+2-C57S und LOV1+2-C250S Domänen wurde als die Superposition des Verhaltens der zugehörigen Domäne analysiert. Ein anderes Verhalten findet man für langsame thermale Rückreaktion des Photoaddukts zum Ausgangsprodukt (dark form). Während die einzelnen Domänen LOV1 und LOV2 einen Zweikomponentenabfall aufweisen, zeigen die beiden gemischten Domänen LOV1+2-C57S und LOV1+2-C250S einen Einkomponentenabfall. Daraus lässt sich schließen, dass die Wechselwirkung zwischen dieser zwei Domänen nicht das Verhalten des Triplettzustandes beeinflusst, sondern das Verhalten des Photoaddukts. Als Mechanismus der Adduktbildung, befürworten wir den �neutral radical-pair� Mechanismus, vorgeschlagen von Kay et al. nach der Studie des LOV1-C57G-Mutantes. Um den Photozyklus jeder LOV Domäne simulieren zu können, wurden mehrere mögliche Modelle ausprobiert, die uns ermöglicht haben, die Vorgänge des phot-Proteins in Chlamydomonas reinhardtii besser zu verstehen.
