Abstract (German)
Seit ihrer Entdeckung im Jahre 1946 durch Felix Bloch und Edward Mills Purcell
hielt die Kernspinresonanz-Spektroskopie (NMR, "nuclear magnetic resonance")
erfolgreich Einzug in viele Gebiete der Physik, Chemie, Biologie und Medizin.
Der große Nachteil der NMR liegt in ihrer geringen Sensitivität. Im thermischen Gleichgewicht bei Raumtemperatur liegt der für die Signalintensität entscheidende Besetzungszahlunterschied zwischen zwei Kernspinniveaus bei weniger als 0,001%.
Um dennoch ein ausreichendes Signal-zu-Rausch-Verhältnis zu erhalten, werden heute sehr hohe Magnetfelder verwendet (bis zu 21 Tesla).
Ein anderer Ansatz zur Verbesserung der Sensitivität beruht auf dem optischen Pumpen (Kastler, 1950) eines Alkalimetalldampfes, meist Rubidium. In einer sog. Pumpzelle, die außerdem Helium, Stickstoff und Xenon enthält, werden dabei die Spins der Rubidium-Valenzelektronen durch zirkular polarisiertes Laserlicht (Wellenlänge der Rb-D1-Linie, 794,8 nm) zu über 90% polarisiert. In binären Stößen und spontan gebildeten van-der-Waals-Molekülen übertragen die Rubidiumatome dann Spinpolarisation auf Xe-129-Kerne.
Die Xe-129-Kernspinpolarisation kann über 50%, also etwa das 100 000-fache verglichen mit dem thermischen Gleichgewicht, erreichen, weswegen es als "hyperpolarisiertes Xenon" bezeichnet wird.
In diesem Zusammenhang wird im Rahmen dieser Doktorarbeit zum einen die Herstellung von hyperpolarisiertem Xe-129 in einer Pumpzelle numerisch modelliert und zum anderen werden experimentelle Untersuchungen zur Anwendung von superkritischem Xenon als Lösungsmittel für in inverse Mizellen eingeschlossene Biomoleküle durchgeführt.
Im ersten Teil wird mit Hilfe der Finite-Elemente-Methode (Software FEMLAB 3.1) erstmals ein dreidimensionales mathematisches Modell für den Pumpzellzylinder, in dem der Rubidiumdampf das Laserlicht absorbiert, entwickelt. Dieses Modell basiert auf acht gekoppelten partiellen Differentialgleichungen: dem Navier-Stokes-Gleichungssystem, der Wärmeleitungsgleichung, der Pumpratengleichung, die die Photonenabsorption in der Pumpzelle beschreibt, und den Ratengleichungen für die Rubidium- und Xe-129-Spinpolarisation. Die Relaxation der Spinpolarisation an den Pumpzellwänden wird hierbei über die Randbedingung implementiert und nicht mehr als konstante Rate in den Spinratengleichungen. Dies ermöglicht es, den Einfluss der Konvektion auf die erreichbare Spinpolarisation zu untersuchen. Auf der Basis dieses Modells wurden analytische und numerische Berechnungen zur Pumpzelle mit und ohne kontinuierlichen Gasdurchfluss durchgeführt.
Der Einfluss verschiedener Parameter, wie z.B. der Ofentemperatur, die die Rubidiumdampfdichte bestimmt, des Xenonpartialdrucks, der Laserleistung und der Pumpzellgeometrie, wird charakterisiert und optimiert. Im Vergleich mit von in unserer Arbeitsgruppe durchgeführten Experimenten und Literaturdaten ergibt sich eine gute Übereinstimmung. Daraus folgen wichtige Erkenntnisse für den Aufbau und den Betrieb einer Pumpzelle.
Im zweiten Teil wird zunächst die sog. chemische Verschiebung, die Rückschlüsse auf die chemische Umgebung eines Atomkerns zulässt, von Xe-129 in Abhängigkeit vom Xenondruck bis zu ca. 70 MPa gemessen. Für das in superkritischem Xenon gelöste, amphiphile Tensid AOT (Bis(2-ethyl-1-hexyl)sulfosuccinat) wird mit Hilfe von H-1-NMR-Relaxationszeiten und Magnetfeldgradienten-H-1-NMR-Spektren die Existenz von inversen AOT-Mizellen demonstriert und deren Wassergehalt bestimmt. Erstmals konnte der Einschluss verschiedener Biomoleküle, wie der Aminosäure Glycin und der Proteine Lysozym (14,3 kDa) und Trypsin (23,8 kDa), in diese Mizellen NMR-spektroskopisch nachgewiesen werden, z.B. über ein H-1-C-13-HSQC-Experiment. Ob dies die aufgrund der schlechten spektralen Auflösung schwierige Strukturbestimmung großer Proteine (>20 kDa) erleichtern wird, hängt von zwei Faktoren ab: zum einen, ob der Radius und damit die Korrelationszeit der inversen Mizellen verringert werden kann, zum anderen, ob ein Spinpolarisationsübertrag von hyperpolarisiertem Xe-129 in superkritischem Zustand auf Atomkerne der Proteine möglich ist.

Since its discovery in 1946 by Felix Bloch and Edward Mills Purcell, NMR spectroscopy (nuclear magnetic resonance) has been used in many applications in physics, chemistry, biology, and medicine. A major disadvantage of NMR is its low sensitivity. In thermal equilibrium at room temperature, the occupation number difference between two nuclear spin levels amounts to less than 0.001%.
To achieve sufficient sensitivity, very high magnetic fields (up to 21 Tesla) need to be applied.
A different approach is offered by optically pumping an alkali metal vapour, e.g. Rubidium. In a so called pump cell, which in addition contains Helium, Nitrogen, and Xenon, the Rb valence electron spins are polarised by more than 90% by absorption of circularly polarised laser light (wave length of Rb-D1-line: 794.8 nm). Via binary collisions and formation of van-der-Waals molecules spin polarisation is transferred to the Xe-129 nuclei.
Xe-129 nuclear spin polarisation can amount to more than 50%, which is 100 000 times the value of thermal equlibrium. Therefore, it is called "hyperpolarised Xenon".
Within this context this PHD thesis is concerned with two topics.
On the one hand the production of hyperpolarised Xe-129 in a pump cell is numerically modelled. On the other hand experimental investigations concerning the application of supercritical Xenon as solvent for biomolecules encapsulated in reverse micelles are carried out.
In the first part, a mathematical three dimensional model for the pump cylinder
(in which the Rb vapor absorbs the laser light) is developed, using the finite element method (software FEMLAB 3.1). This model is based on eight coupled partial differential equations: the Navier-Stokes equation system, the heat transfer equation, the pump rate equation describing the absorption of laser photons, and the rate equations for the Rubidium and Xenon-129 spin polarisation. Wall relaxation is implemented via the boundary condition and not any more as constant rate in the equation itself. This allows to determine the influence of convection on spin polarisation. Based on this model analytic and numerical calculations of the pump cell are carried out for the static pump cell and under continuous gas flow conditions.
The influence of different parameters, e.g. oven temperature (which determines Rb vapor density), Xenon partial pressure, laser power, and pump cell geometry, is characterised and optimised. Comparison to experiments of our group and to data from literature yields good agreement so that important guidelines for the construction and operation of a pump cell can be established.
In the second part the NMR chemical shift (which implies information about the chemical environment of a nucleus) of Xe-129 is measured at different Xe pressures up to 70 MPa. After dissolving the amphiphilic surfactant AOT (bis(2-ethyl-1-hexyl)sulfosuccinate) in supercritical Xenon, the existence of reverse AOT micelles is demonstrated via H-1-NMR-relaxation times and pulsed-field-gradient-NMR-spectra, and their water content is determined. For the first time, the encapsulation of different biomolecules, e.g. the amino acid glycine, and the proteins lysozyme (14.3 kDa) and trypsine (23.8 kDa), within these micelles could be demonstrated by means of NMR experiments, e.g. a H-1-C-13-HSQC experiment.
It depends on two factors whether this method can enhance the bad spectral resolution and the difficult structure determination of large proteins (>20 kDa): on the one hand, on the possibility to further reduce the radius of the reverse micelles and, therefore, their correlaton time, on the other hand, on the possibility to transfer spin polarisation from supercritical hyperpolarised Xe-129 to the nuclei of the proteins.
