Abstract (English)

With the goal of controlling the events at the bone-implant interface, it was the main objective of this thesis to provide a basis for the conjugation of cell stimulating molecules or targeting motifs to the surface of hydroxyapatite ceramic discs and particles. To this end, methods for surface functionalization have been investigated for the attachment of biomolecules. The approach focused on combining three approved and effective principles for enhanced osseointegration of implants: hydroxyapatite coating, surface-immobilized bisphosphonates as anchoring molecules, and localized delivery of BMP 2 by immobilization.
In a cell culture study, the biological activity of immobilized BMP-2 compared to adsorbed BMP-2 was elucidated. In general, immobilized BMP-2 significantly increased the expression of Alkaline Phosphatase in C2C12 cells. The novel bisphosphonate-based method was found to be as efficient as the well-established aminosilanization. To gain deeper insight into the adsorption behavior of the aminobisphosphonate pamidronate on hydroxyapatite surfaces, an adsorption study was performed showing that the number of functional groups on the surface can be controlled.
Additionally, uptake and distribution of nanoparticles in multicellular tumor spheroids was investigated by using confocal laser scanning microscopy in order to elucidate particle transport and diffusion processes in a three dimensional cell culture model.
In conclusion, this thesis contributed to the development of surface modification methods for Ca-rich inorganic materials in order to achieve a selective cell response.

Um die Ereignisse an der Grenzfläche Knochen-Implantat zu kontrollieren, war es das Hauptziel dieser Arbeit, eine Basis für die Anbindung von zellstimulierenden Molekülen oder Targetingsequenzen an die Oberfläche von Hydroxylapatit-Keramikscheiben und Partikeln zu schaffen. Der gewählte Ansatz konzentrierte sich darauf, drei verlässliche und effiziente Prinzipien für die gesteigerte Osteointegration von Implantaten zu kombinieren: Oberflächenbeschichtung mit Hydroxylapatit, immobilisierte Bisphosphonate als Anker-Moleküle und lokale Verfügbarkeit von BMP-2 durch Immobilisierung.
In einer Zellkulturstudie wurde die biologische Aktivität von immobilisiertem BMP-2 im Vergleich zu adsorbiertem BMP-2 untersucht. Im Allgemeinen steigerte immobilisiertes BMP 2 die Expression von Alkalischer Phosphatase in C2C12 Zellen signifikant. Dabei war die neue Bisphosphonat-basierte Methode ebenso effizient wie die wohlbekannte Aminosilanisierung. Um die Adsorption des Aminobisphosphonates Pamidronat an Hydroxylapatitoberflächen besser zu verstehen, wurde eine Adsorptionsstudie durchgeführt, die zeigte, dass die Zahl der funktionellen Gruppen an der Oberfläche kontrolliert werden kann.
Des Weiteren wurden Aufnahme und Verteilung von Nanopartikeln in multizellulären Tumorsphäroiden mittels konfokaler Laserscanning Mikroskopie untersucht, um Transport und Diffusionsprozesse in einem dreidimensionalen Zellkulturmodell näher zu beleuchten.
Abschließend lässt sich sagen, dass diese Arbeit zur Entwicklung von Methoden zur Oberflächenmodifizierung von Ca-reichen anorganischen Materialien beitrug, um eine selektive Zellantwort zu erzielen.
