Abstract (English)
Conjugated linoleic acid (CLA) isomers are dietary fatty acids that modulate gene expression in many cell types. The aim of this work was to examine the function of the single c9,t11-, t10,c12- and t9,t11 CLA isomers on gene expression in human macrophages. Therefore, in vitro MCSF differentiated monocyte derived macrophages from three healthy donors and THP-1 macrophages were incubated with these CLA-isomers and whole genome transcripts were analyzed with DNA-microarrays and realtime RT-PCR. T9,t11-CLA, but not c9,t11- and t10,c12-CLA activated target genes of SREBP, SREBP-1c and ABCG1. This led to the hypothesis that ABCG1 is activated via SREBP-1c. Gene reporter assays with deletion constructs of the ABCG1 regulatory region and cotransfections with a SREBP-1c expression plasmid in RAW 264.7 macrophages confirmed that t9,t11-CLA activates ABCG1 via SREBP-1c. EMSAs showed that SREBP-1c binds to a SREBP responsive element in the promoter of ABCG1. To answer the question by which mechanism this CLA isomer induces SREBP-1c, transcription of liver X receptor (LXR), which is a major regulator of SREBP-1c, and its target genes was analyzed in t9,t11-CLA stimulated macrophages with RT-PCR. A significant induction of LXR and its target genes was found. Gene reporter assays with a plasmid containing the LXR regulatory region of the SREBP-1c promoter confirmed that SREBP-1c is activated via LXR by t9,t11-CLA. To determine the concentration range of t9,t11-CLA, human macrophages were treated with various doses of CLA and mRNA expression of target genes was analyzed. Incubation of human macrophages with 5 and 10µM t9,t11-CLA, which can be considered as physiological levels, led to a significant modulation of LXR, SREBP-1c, ABCG1 and ABCA1. Finally the effects of 10µM t9,t11-CLA on ABCA1 and ABCG1 expression were found to parallel with the functional response of cholesterol efflux in human macrophages.

Konjugierte Linolsäuren (CLAs) sind Positions- und geometrische Isomere der essentiellen Fettsäure Linolsäure und finden sich überwiegend in Rindfleisch und Milchprodukten. Ziel dieser Arbeit war es den Einfluß der einzelnen cis-9,trans-10-, trans-10,cis-12- und trans-9,trans-11-CLA Isomere auf die Genexpression von Makrophagen zu untersuchen. Dafür wurden in vitro mit MCSF (Macrophage Colony Stimulating Factor) differenzierte Makrophagen von drei gesunden Spender mit den einzelnen CLA-Isomeren inkubiert und daraufhin die mRNA Expression mit DNA-Microarrays und Real-Time (RT)-PCR untersucht. Es zeigte sich, daß trans-9,trans-11-CLA, jedoch nicht die anderen beiden Isomere, den nukleären Transkriptionsfaktor SREBP (Sterol Regulatory Element Binding Protein)-1c und den ABC-Transporter ABCG1 signifikant induzierten. Zur detaillierten Analyse der trans-9,trans-11-CLA vermittelten Aktivierung von ABCG1 wurde die Aktivität von verschiedenen Luciferase-Konstrukten der regulatorischen Region von ABCG1 bestimmt. Desweiteren wurden Kotransfektions-Experimente mit SREBP-1c Überexpressionskonstrukten und Gelretardations- Experimente durchgeführt. Es konnte gezeigt werden, daß trans-9,trans-11-CLA SREBP-1c aktiviert und damit zur SREBP-1c abhängigen Induktion des ABCG1 Promotors führt. Um zu untersuchen über welchen Mechanismus trans-9,trans-11-CLA SREBP-1c aktiviert, wurde die Expression des Transkriptionsfaktor Liver X Receptor (LXR), einem wichtigen SREBP-1c Aktivator, in trans-9,trans-11 CLA behandelten humanen Makrophagen mit RT-PCR analysiert. Es stellte sich heraus, daß das CLA Isomer LXR direkt induziert. Weiterführende Luciferase Experimente mit SREBP-1c Promotor-Konstrukten konnten bestätigen, dass trans-9,trans-11-CLA zuerst LXR und daraufhin SREBP-1c aktiviert. Eine Behandlung von humanen Makrophagen mit verschiedenen Konzentrationen trans-9,trans-11-CLA und eine anschließende Expressionsanalyse von LXR, SREBP-1c und ABCG1 mittels RT-PCR ergab, daß bereits Konzentrationen von 5µM und 10µM erhebliche transkriptionelle Effekte haben. Diese Daten konnten ebenfalls auf funktioneller Ebene bestätigt werden, indem ein signifikanter Anstieg des ABCG1 vermittelten Cholesterin Exports in humanen Makrophagen gezeigt wurde.
