Abstract (German)

Diese Arbeit beschreibt die Identifikation und die Rolle von Fussel-15 (Functional smad suppressing element on chromosome 15), Fussel-18 (Functional smad suppressing element on chromosome 18) und TANGO in physiologischen und pathophysiologischen Prozessen. Dabei wurde gezeigt, dass Fussel-15 und Fussel-18, über eine Interaktion mit den Smad Molekülen, inhibitorische Eigenschaften auf Signaltransduktionen des TGF-ß (Transforming Growth Factor) bzw. des BMP (Bone Morphogenetic Protein) Signalwegs besitzen. Die TGF-ß Superfamilie ist neben tumorbiologischen Prozessen an weiteren Vorgängen beteiligt. Ein ausgedehntes Expressions-Screening zeigte, dass Fussel-15 und Fussel-18 ein ganz spezifisches Expressionsmuster hauptsächlich in neuronalen Strukturen des Kleinhirns und Rückenmarks aufzeigten. Für Fussel-15 ließ sich zudem eine Expression in Fibroblasten der frühen Wundheilung und eine Fehlregulation dieses Proteins in Krankheiten mit überschießender Matrixproduktion feststellen.
Zudem wurde die Rolle von TANGO, einem MIA (Melanoma Inhibitory Activity) homologen Protein, im Malignen Melanom, im Kolon- und im Leberzellkarzinom untersucht. Dabei zeigte sich, dass dieses Molekül als Tumorsuppressor wirkt. Die Analyse von TANGO in nicht-tumorösen Zellen ergab eine Induktion dieses Moleküls nach Zelladhäsion. Zusätzlich ließ sich eine Interaktion von TANGO mit dem leukozytenspezifischen Integrin CD11c feststellen, wodurch in Folge die Transmigration von Blutzellen beschleunigt wird.

This study describes the identification and the role of Fussel-15 (Functional smad suppressing element on chromosome 15), Fussel-18 (Functional smad suppressing element on chromosome 18) and TANGO in physiological and pathophysiological processes. It was shown that Fussel-15 and Fussel-18 have inhibitory functions on TGF-ß (transforming growth factor) and BMP (bone morphogenetic protein) signal transductions by interacting with Smad molecules. Beside the role of TGF-ß in tumorbiological processes, the TGF-ß superfamily is also involved in other activities. An extensive expression-screening showed a specific expression pattern of Fussel-15 and Fussel-18 in neuronal structures of the cerebellum and the spinal cord. Furthermore, an expression of Fussel-15 in fibroblasts of the early wound healing and a deregulation of the protein in diseases with increased matrix production was observed.
Additionally, the role of TANGO, a MIA (melanoma inhibitory activity) homologous protein was analysed in malignant melanoma, in colon- and hepatocellular carcinoma. Thereby, it was shown that TANGO acts as a tumor suppressor. The analysis of TANGO in non-tumorous cells presents an induction of TANGO after cell adhesion. In addition, an interaction with the leukocyte specific integrin CD11c was detected, running in an accelerated transmigration of blood monocytes.
