Abstract (German)
In der vorliegenden Dissertation wurde der Zusammenhang zwischen der Quartärstruktur, der thermischen Stabilität und der enzymatischen Aktivität der Anthranilat-Phosphoribosyltransferase (AnPRT) untersucht, die in der Tryptophanbiosynthese den Mg2+-abhängigen Umsatz von Anthranilat (AA) und 5-Phosphoribosyl-α1-pyrophosphat (PRPP) zu Phosphoribosylanthranilat (PRA) und Pyrophosphat (PPi) katalysiert.
Der erste Teil der Arbeit basiert auf dem bekannten Befund, dass die homodimere AnPRT aus dem hyperthermophilen Archaeon Sulfolobus solfataricus (ssAnPRT) durch die beiden Mutationen I36E und M47D am N- und C-Terminus der Helix α3 monomerisiert werden kann. Das resultierende ssAnPRT-M36I+M47D Monomer besitzt eine reduzierte thermische Stabilität bei unveränderter katalytischer Aktivität. Diese Resultate legten die Vermutung nahe, dass wildtypische AnPRTs mit geladenen Resten am N- und C-Terminus der Helix α3 Monomere bilden.
Auf der Basis dieser Hypothese wurden mittels eines multiplen Sequenzalignments (MSA) aus 220 AnPRT-Sequenzen sieben putativ monomere AnPRTs identifiziert und drei Vertreter aus den mesophilen Bakterien Chlorobium luteolum (clAnPRT), Staphylococcus aureus (saAnPRT) und Staphylococcus haemolyticus (shAnPRT) in rekombinanter Form hergestellt und gereinigt. Ihre Charakterisierung mittels analytischer Gelfiltration und analytischer Ultrazentrifugation bestätigte den vermuteten monomeren Assoziationszustand. Steady-state enzymkinetische Messungen bei 25 °C lieferten für diese AnPRTs vergleichbare Wechselzahlen (kcat) und Michaeliskonstanten für Anthranilat (KMAA) sowie 5-Phosphoribosyl-α1-pyrophosphat (KMPRPP). Thermische Auffaltungsexperimente ergaben apparente Schmelztemperaturen (TMapp) von etwa 43 °C (saAnPRT und shAnPRT) bzw. 50 °C (clAnPRT).
In Umkehrung der Monomerisierung von ssAnPRT wurde versucht, die drei nativ monomeren AnPRTs durch den Austausch der geladenen Reste an den Positionen „36“ und „47“ gegen hydrophobe Aminosäuren zu dimerisieren, was jedoch nicht gelang. Um zukünftige Versuche auf eine stabile strukturelle Basis zu stellen, soll die Röntgenstruktur dieser Enzyme bestimmt werden. Die Erfolgsaussichten dafür konnten im Rahmen dieser Arbeit durch die Einführung analoger stabilisierender Punktmutationen (T78I in clAnPRT, V76I in saAnPRT und shAnPRT) erhöht werden.
Dem zweiten Teil der Arbeit lag der frühere Befund zugrunde, dass das durch die zwei Mutationen D83G und F149S in seiner Katalyse aktivierte Monomer ssAnPRT-(I36E+M47D-D83G+F149S) mit einem TMapp-Wert von 70 °C deutlich weniger stabil ist als das nicht aktivierte ssAnPRT-I36E+M47D Monomer mit einem TMapp-Wert von 81 °C.
Die thermische Stabilität von ssAnPRT-(I36E+M47D+D83G+F149S) wurde durch eine Kombination aus Zufallsmutagenese und Selektion in vivo erhöht und anschließend die aus der Stabilisierung resultierenden Konsequenzen für den Assoziationszustand und die Aktivität des Enzyms untersucht. Im ersten Schritt wurde mittels error-prone PCR eine plasmidkodierte Genbank von ssAnPRT-(I36E+M47D+D83G+F149S) in Escherichia coli hergestellt, die etwa 9 ∙ 106 unabhängige Klone mit durchschnittlich 12 Nukleotidaustauschen pro Gen umfasste. Zur Selektion auf erhöhte Stabilität wurde das thermophile und genetisch modifizierbare Bakterium Thermus thermophilus verwendet, welches bei Temperaturen zwischen 55 °C und 80 °C wächst. Dazu wurde eigens ein T. thermophilus Stamm mit einer Deletion der chromosomalen Kopie des trpD-Gens hergestellt. Dieser ΔtrpD Stamm ist auf ein aktives und thermostabiles AnPRT Protein angewiesen, um auf Tryptophan-freiem Medium wachsen zu können. Zur Selektion auf stabilisierte Mutanten wurden Zellen des ΔtrpD-Stammes mit der ssAnPRT-(I36E+M47D+D83G+F149S)-Genbank transformiert, auf Medium ohne Tryptophan ausplattiert und bei 79 °C inkubiert. Nach 36 Stunden waren etwa 100 Klone zu sichtbarer Größe herangewachsen. Davon wurden 20 zufällig ausgewählt und die Plasmidinserts sequenziert, um die in ssAnPRT-(I36E+M47D+D83G+F149S) eingebauten Mutationen zu identifizieren. Anschließend wurden verschiedene Einfach- und Mehrfachmutanten heterolog exprimiert, gereinigt und ihre thermische Stabilität bestimmt.
Es zeigte sich, dass ssAnPRT-(I36E+M47D+D83G+F149S) durch fünf Einzelaustausche stabilisiert wird, wobei der induzierte Anstieg des TMapp-Wertes für T77I etwa 8 °C, für F193S etwa 4 °C und für N109S, I169T und L320M jeweils etwa 1 °C beträgt. Die stabilisierenden Effekte waren additiv, so dass die Kombinations-mutante ssAnPRT-(I36E+M47D+D83G+F149S)+T77I+N109S+I169T+F193S+L320M einen um ca. 13 °C erhöhten TMapp-Wert von etwa 83 °C aufwies. Weiter konnte gezeigt werden, dass die beiden effektivsten Austausche T77I und F193S auch im Hintergrund des nicht aktivierten Monomers ssAnPRT-I36E+M47D und des wildtypischen ssAnPRT-Dimers stabilisierend wirken, wobei jedoch die induzierten TMapp-Steigerungen weniger deutlich ausfallen. Eine Analyse auf der Basis der Röntgenstruktur von wildtypischem ssAnPRT lässt vermuten, dass der T77I-Austausch die Packung im Inneren des Proteins verbessert, während der Austausch F193S einen benachbarten flexiblen loop stabilisieren dürfte.
Mittels analytischer Gelfiltration wurde nachgewiesen, dass alle untersuchten ssAnPRT-(I36E+M47D+D83G+F149S)-Mutanten weiterhin als Monomer vorliegen, d. h. keiner der Austausche führte zu einer „Redimerisierung“ des Proteins. Während die meisten stabilisierenden Mutationen die katalytische Aktivität nur moderat beeinträchtigten, führte die in der Nähe des aktiven Zentrums gelegene N109S-Mutation zu einer etwa vierfachen Erniedrigung des kcat-Wertes und einer etwa sechsfachen Erhöhung des Wertes für KMAA. Generell wurde eine negative Korrelation zwischen den Werten für kcat und TMapp beobachtet, d. h. die Aktivität der ssAnPRT-(I36E+M47D+D83G+F149S)-Mutanten nimmt mit zunehmender Stabilität ab. Diese Befunde stützen die Hypothese, dass es im Zuge der Substratbindung und der Produktfreisetzung zu Konformationsänderungen von ssAnPRT kommt, die eine gewisse Flexibilität des Proteingerüstes voraussetzen und durch stabilisierende Austausche beeinträchtigt werden könnten. Derartige Konformationsübergänge sollen in Zukunft über FRET-Untersuchungen nachgewiesen werden, wofür Fluoreszenzfarbstoffe über gezielt eingeführte Cysteinreste an das Protein gekoppelt werden müssen. Im Rahmen dieser Arbeit wurde das dazu verwendete Protokoll soweit optimiert, dass ein Markierungsgrad von etwa 40 % für die verwendeten Farbstoffe Alexa-488 (FRET-Donor) und Alexa-647 (FRET-Akzeptor) erreicht wurde.
Somit sind die wichtigsten Ergebnisse dieser Arbeit die experimentelle Verifizierung eines bioinformatisch vorhergesagten Assoziationszustandes eines Proteins und die erstmalig gelungene Stabilisierung eines Enzyms durch metabolische Selektion in einem thermophilen Wirt.

The dissertation deals with the correlation of quaternary structure, thermal stability and enzymatic activity of the anthranilate phosphoribosyltransferase (AnPRT), which catalyzes the Mg2+-dependent turnover of anthranilate and 5-phosphoribosyl-α1-pyrophosphate (PRPP) to phosphoribosylanthranilate (PRA) and pyrophosphate (PPi).
The first part of the work was based on the finding, that homodimeric AnPRT from the hyperthermophilic archaeon Sulfolobus solfataricus (ssAnPRT) could be monomerized by the
mutations I36E and M47D at the protein interface. The resulting ssAnPRT-I36E+M47D monomer exhibits a reduced thermal stability but unchanged catalytic efficiencies (Schwab et al., 2008). The results lead to the hypothesis, that AnPRTs with charged residues at corresponding positions are monomeric.
Applying multiple sequence alignments (MSA) lead to the identification of seven putative monomeric AnPRTs, whereby three representatives from the mesophilic bacteria Chlorobium
luteolum (clAnPRT), Staphylococcus aureus (saAnPRT) and Staphylococcus haemolyticus (shAnPRT) were produced recombinant and purified. Analytical size exclusion chromatographie and analytical ultracentrifugation experiments confirmed the assumed monomeric association state. Measurements of the steady-state enzyme kinetics provided comparable enzymatic activities, whereas thermal unfolding showed low apparent melting temperatures for all three enzymes. Due to the low conformational stability, it was not possible to solve the X-ray structure and therefore structural models of the native monomeric enzymes were be generated by homology modeling.
The basis of the second part of the work was the identification of the two mutations D83G and F149S (Schlee et al., 2009), which lead to a catalytically activated monomer ssAnPRTI36E+
M47D+D83G+F149S, which has a lower thermal stability (TMapp = 70 °C) than the not activated monomer ssAnPRT-I36E+M47D (TM app = 81 °C). The thermal stability of ssAnPRTI36E+M47D+D83G+F149S was increased by a combination of gene randomization and selection in vivo. In the first step, the gene was randomized by error-prone PCR and a
plasmid-encoded gene library has been generated in E. coli, which comprised of 9 * 106 individual clones with 12 nucleotide exchanges on average. For selection on increased thermal stability, the thermophilic and genetically modifiable bacterium Thermus thermophilus was used, which grows between 55 and 80 °C. Therefore, a T. thermophilus strain with a deletion of a chromosomal copy of the trpD-gene was generated. The ΔtrpD
strain is dependent on an active and thermostable AnPRT protein to grow on tryptophan free medium. For the selection of stabilized mutants, the ΔtrpD cells were transformed with the
gene library, platted on medium lacking tryptophan and incubated at 79 °C. After 36 hours, there were about 100 colonies, from which 20 were selected and the plasmid inserts were sequenced to determine the introduced amino acid mutations. Afterwards single and combination mutants were produced recombinant, purified and their thermal stability was
determined. By this approach, five mutations could be identified, at which the induced increase of the TMapp-value for T77I is 8 °C, for F193S 4 °C and for N109S, I169T and L320M about 1 °C in each case. The stabilizing effects are additive, leading to a final combination mutant with a TMapp-value of about 83 °C. The structural analysis of the two best mutations leads to the assumption, that T77I improves the packing of the protein interior, while F193S stabilizes an adjacent flexible loop.
All purified mutants were monomeric, which was proved by size exclusion chromatographie. The determination of the enzymatic parameters revealed a negative correlation of turnover numbers and TMapp-values, which means, that activity declines with enhances thermal stability. These results confirm the hypothesis, that during substrate binding and product release there is a conformational change, which requires protein flexibility, that could be compromised by stabilizing mutations. The conformational change will be analyzed by FRETexperiments, whereby the fluorophors have to be coupled to introduced cysteines. In the frame of this work, the labeling and purification protocol was optimized to yield a labeling
degree of 40 % for two Alexa-fluorophors coupled to ssAnPRT.
Hence, the most important results of this work are the experimental validation of bioinformatically predicted association states of three proteins and the first successful
stabilization of an enzyme by metabolic selection in a thermophilic organism.
