Abstract (German)

Guaianolide sind sekundäre Pflanzenmetabolite. Sie gehören zur Naturstoffklasse der Sesquiterpenlaktone. In dieser Arbeit wurden stereoselektiv Modellguaianolide synthetisiert um durch Funktionalisierung eine Substanzbibliothek aufzubauen. Guaianolide sind sekundäre Pflanzenmetabolite. Sie gehören zur Naturstoffklasse der Sesquiterpenlaktone. In dieser Arbeit wurden stereoselektiv Modellguaianolide synthetisiert um durch Funktionalisierung eine Substanzbibliothek aufzubauen. Diese Substanzbibliothek wurde Zellassays an MCF-7 Zelllinien unterzogen um einen Zusammenhang zwischen deren Funktionalisierung und deren biologischer Aktivität herzustellen.
Des weiteren wurde eine autokatalytische Re-Elimination einer Dimethylaminogruppe an der alpha-Position des Laktons entdeckt, wenn die C-4 Hydroxylgruppe mesyliert vorlag. Daraus resultierte auch eine Erhöhung der Cytotoxizität.
Um die Selektivität der biologisch aktiven Modellguaianolide zu erhöhen, wurden diese an Derivate von Neuropeptid Y gebunden. Diese Konjugate zeigten allerdings keine biologische Aktivität mehr.
Schließlich wurde eine an Guaianoliden neuartige stereoselektive Homoallyl-Cyclopropylmethyl Umlagerung beschrieben, die durch katalytische Mengen verschiedener Lewis Säuren durchgeführt werden kann, sofern sich zwischen C-6 und C-6a eine C=C Doppelbindung befindet.
Dabei wurden durch Screening verschiedener Modellguaianolide Grenzen und Möglichkeiten dieser neuartigen Umlagerung evaluiert, sowie aufbauend auf diesen Daten, ein plausibler Reaktionsmechanismus postuliert.

Guaianolides are secondary plant metabolites. They belong to the natural product class of sesquiterpene lactones. In this dissertation, model guaianolides were synthesized stereoselectively. Guaianolides are secondary plant metabolites. They belong to the natural product class of sesquiterpene lactones. In this dissertation, model guaianolides were synthesized stereoselectively. By functionalisation, a guaianolide library could be established. This library was used to elucidate a structure activity relationship of the functionalisation of guaianolides and their biological activity by carrying out cell assays with MCF-7 cell lines.
Furthermore a auto catalytically re-elimination of a dimethyl amino group at the alpha-position of the lactone could be discovered, which resulted in a raise of the cytotoxicity levels.
To improve the selectivity of biological active guaianolides, they were linked to derivatives of Neuropeptide Y. However, these adducts did not show any biological activity.
Last but not least a non described novel stereoselective homoallyl-cyclopropylmethyl rearrangement of guaianolides with catalytic amounts of different lewis acids was characterized.
By screening different model guaianolides, limits and possibilities of this novel rearrangement were evaluated and a feasible reaction mechanism could be postulated.
