Abstract (English)

NMR-guided fractionation of the petrolium ether- and ethyl acetate extracts of the bark of Garcinia fusca Pierre led to the isolation of thirty one phenolic compounds using various chromatographic methods such as TLC, flash column chromatography, gel permeation chromatography, centrifugal partition chromatography and semi-preparative HPLC. The structures of these compounds were elucidated by spectroscopic techniques such as UV, HR-MS, 1D and 2D NMR as well as the optical rotation, comprising a tocotrienol, a benzaldehyde derivative, four biflavonoids and twenty five prenylated xanthone derivatives, eight of which are new. New xanthones exhibited cytotoxicity against HeLa cells with IC50 values in the range of 19.1 – 45.9 µM. Furthermore, a bisxanthone, griffipavixanthone was evaluated for anti-angiogenic activity by in vitro proliferation assay on HEMC-1 cell line and it showed a greatly strong anti-proliferative effect with an IC50 value of 0.15 ± 0.0085 µM after 72h incubation.

NMR-geleitete Fraktionierung eines Petrolether- und Ethylacetatextraktes der Rinde von Garcinia fusca PIERRE führte zur Isolierung von 31 phenolischen Verbindungen unter zu Hilfenahme verschiedener chromatographischer Methoden, wie DC, Flash-Chromatographie, Gel-Chromatographie, Centrifugal Partition Chromatographie und semi-präparative HPLC. Die Strukturaufklärung wurde durch spektroskopische Techniken, wie UV, HR-MS, 1D- und 2D-NMR, und durch die optische Drehung bewerkstelligt. Das führte zu einem Tocotrienol, einem Derivat eines Benzaldehyds, vier Biflavanoiden und 25 prenylierten Xanthonen, von denen acht zum ersten Mal beschrieben wurden. Die neuen Xanthone zeigten eine Zytotoxizität gegen HeLa-Zellen mit IC50-Werten in einem Bereich von 19.1 – 45.9 µM. Außerdem wurde Griffipavixanthone, ein Bisxanthon, für die Bestimmung der antiangiogenetischen Aktivität durch einen in vitro Proliferationsassay an der HMEC-1-Zelllinie ausgewählt und zeigte einen großen anti-proliferativen Effekt mit einem IC50-Wert von 0.15 ± 0.0085 µM nach einer Inkubationszeit von 72 h.
