T-Zell-Antikörper finden seit Langem Anwendung in der Prävention und Therapie der Graft-versus-Host Disease, der wichtigsten Komplikation der allogenen hämatopoetischen Stammzelltransplantation. Antithymozytenglobulin (ATG) ist ein Medikament, das Antikörper gegen ein breites Spektrum von Antigenen auf T-Zellen, aber auch auf anderen Immunzellen enthält. Alemtuzumab ist ein humanisierter monoklonaler Antikörper gegen CD52, das von Lymphozyten in besonders großer Menge exprimiert wird.
Das Enzym Indolamin-2,3-dioxygenase (IDO) ist verantwortlich für den Abbau der Aminosäure Tryptophan zu Kynurenin. Es gibt Hinweise darauf, dass die Induktion von IDO in dendritischen Zellen ein Mechanismus ist, über den ATG immunsuppressiv wirkt und eine Graft-versus-Host Disease verhindern oder mildern kann.
Wir führten durchflusszytometrische Untersuchungen durch, die ergaben, dass ATG, nicht aber Alemtuzumab in humanen, von Monozyten abstammenden dendritischen Zellen zur Hochregulation von Oberflächenmarkern führt, die auf eine stattgefundene Reifung der dendritischen Zellen hindeuten. Durch quantitative Realtime-PCR konnten wir belegen, dass ATG und in geringerem Ausmaß auch Alemtuzumab zu einer vermehrten Menge an mRNA der Gene IDO-1 und IDO-2 führen. Im Western Blot war das IDO-Protein in Proben nachweisbar, die mit ATG behandelt worden waren. Zudem wurde die IDO-Expression schwächer bei längerer Inkubation mit ATG vor Zellernte. Die Konzentration von Tryptophan und seinen Metaboliten wurde massenspektrometrisch gemessen. Dabei ergaben sich ein Abfall von Tryptophan und ein entsprechender Anstieg von Kynurenin und anderen Abbauprodukten unter dem Einfluss von ATG; unter Alemtuzumab blieben solche Veränderungen aus.
Insgesamt zeigen die für diese Arbeit erhobenen Daten, dass ATG humane dendritische Zellen zur Reifung bringt, in diesen Zellen IDO auf Ebene von RNA und Protein induziert und dass das produzierte IDO enzymatisch aktiv ist.

Antibodies against T-cells have long been used for the prevention and therapy of graft versus host disease, the most important complication of allogeneic hematopoietic stem cell transplantation. The antithymocyte globulin (ATG) drug contains antibodies against a broad spectrum of antigens on T-cells, but also on other immune cells. Alemtuzumab is a humanized monoclonal antibody against CD52, which is abundantly expressed by lymphocytes.
The enzyme indoleamine 2,3-dioxygenase (IDO) is responsible for the degradation of the amino acid tryptophan towards kynurenine. Previous studies suggest that the induction of IDO is a mechanism through which ATG leads to immunosuppression and prevents or reduces graft versus host disease.
Our flow cytometry assays demonstrate that, in human monocyte-derived dendritic cells, ATG, but not alemtuzumab results in the up-regulation of surface markers which indicate the maturation of dendritic cells. We showed by quantitative real-time PCR that ATG and to a smaller extend alemtuzumab lead to an increased quantity of IDO-1 and IDO-2 mRNA. In western blots, IDO protein was detected in samples treated with ATG. Furthermore, the IDO expression was diminished with longer incubations with ATG prior to cell harvesting. The concentration of tryptophan and its metabolites was measured by mass spectrometry, revealing a decrease of tryptophan and a corresponding increase of kynurenine and other catabolites under the influence of ATG; alemtuzumab did not cause such changes.
The data collected for this thesis show that ATG leads to the maturation of human dendritic cells, induces IDO at the level of RNA and protein, and that the produced IDO is enzymatically active.
