Die vorliegende Arbeit behandelt die Bedeutung von Endothelinen für die Steuerung
des Reninsystems. Renin stellt in seiner Rolle als Agens im Renin-Angiotensin-
Aldosteron-System und der damit verbundenen Einflussnahme auf den systemischen
Blutdruck, die glomeruläre Filtrationsrate sowie den Wasser- und Elektrolythaushalt
ein entscheidendes Enzym der Niere dar. Neben dem Renin-Angiotensin-
Aldosteron-System ist das Endothelinsystem das stärkste Vasopressorsystem des
Körpers. Zwischen beiden Systemen bestehen zahlreiche Interaktionen. Das Ziel
der vorliegenden Arbeit besteht in der Untersuchung eben jener Einflüsse des
Endothelinsystems auf das Reninsystem. Dazu erfolgte zunächst die Bestimmung
der renalen Endothelinrezeptorlokalisation anhand immunhistochemischer Untersuchungen
von Mäusenieren. Anschließend wurde mithilfe einer pharmakologischen
Blockade von Endothelinrezeptoren an C57BL/6-Mäusen einerseits und an Ren-
1d+/Cre-ETA
fl/fl-Mäusen mit angeborener reninzellspezifischer Endothelinrezeptordefizienz
andererseits untersucht, welche Auswirkungen sich im Hinblick auf Verteilung
und Quantität der Reninzellen und der -produktion sowie der Expression
von Endothelinrezeptoren ergeben. Als Endothelinrezeptor-Antagonisten fanden
die Substanzen BQ610 (ETA-selektiv), BQ788 (ETB-selektiv) sowie Bosentan (dualer
Endothelinrezeptorantagonist) Verwendung. Zur Bestimmung der endothelialen
Beeinflussung der Elektrolythomöostase schloss sich eine atomemissionsspektroskopische
Ermittlung der Kalium- und Natrium-Urin-Konzentration sowie der Urin-
Osmolalität an. Zur Analyse der Einflussnahme eines angeborenen reninzellspezifischen
Endothelinrezeptormangels auf juxtaglomeruläre Zellen auf zellulärer Ebene
erfolgte die Untersuchung der Reninproduktion anhand von Ren-1d+/Cre-ETA
fl/fl-
Mäusen. Zur Klärung der Bedeutung des ETA-Rezeptors für die Stimulation des
Reninsystems schließlich lag der Fokus auf der Differenz der Reninproduktion zwischen Wildtyp- und Knockout-Stämmen. Zusammenfassend lässt sich sagen, dass
die Lokalisation der Endothelinrezeptoren in Mäusenieren weitgehend der Expression
von Endothelinrezeptoren in der Niere anderer Säugetiere entspricht. Abweichungen
ergeben sich insofern, als im Vas afferens sowie im proximalen Tubulus keine
ETB-, im distalen Tubulus und im Sammelrohr hingegen keine ETA-Rezeptoren
nachzuweisen waren. Die Analyse der Elektrolytkonzentrationen förderte keine aufschlussreichen
Erkenntnisse zu Tage, eine endotheliale Beeinflussung der Elektrolythomöostase
kann nicht postuliert werden. Im Hinblick auf die Reninfreisetzung
zeigten die durchgeführten Untersuchungen keine signifikanten Unterschiede. Trotz
der Expression renaler Endothelinrezeptoren scheint sowohl deren Antagonismus
als auch deren reninzellspezifische Abwesenheit in vivo keine signifikanten Einflüsse
auf das Reninsystem nach sich zu ziehen. Diese mangelnde Einflussnahme bei funktioneller
Defizienz der Endothelinrezeptoren lässt mit Blick auf die in der Literatur
beschriebenen Auswirkungen von Endothelinen auf eine Vielzahl renaler Parameter
– einschließlich des Reninsystems – die Existenz kompensatorischer Mechanismen
vermuten. Im Fokus dieser Kompensationsmechanismen könnten möglicherweise
Prostaglandine und NO stehen. Ferner verbleibt die Rolle des ETB-Rezeptors sowie
der Endothelinsubtypen ET2 und ET3 genauer zu eruieren.

Translation of the abstract (English)

The present work deals with the role of endothelins concerning the regulation of
the renin system. In its role as an agent in the renin system and the associated
influence on systemic blood pressure, glomerular filtration rate, and water homeostasis,
renin is of crucial interest for the kidney. Apart from the renin system, the
endothelin system is of importance due to blood pressure regulation. Between both
systems numerous interactions exist. The goal of the present work is to scrutinize
those influences of the endothelin system over the renin system. To this aim the
determination of the localization of renal endothelin receptors using immunohistochemic
methods on the basis of mouse kidneys will be the first step. Subsequent,
using a pharmacologic antagonism of endothelin receptors of C57BL/6 mice on
the one hand and Ren-1d+/Cre-ETA
fl/fl mice with congenital renin cell specific deficience
of endothelin receptors on the other hand, it shall be examined the consequences
regarding the distribution and quantity of renin cells and renin production
as well as the expression of endothelin receptors. As endothelin receptor antagonists,
the substances BQ610 (ETA-selective), BQ788 (ETB-selective) and Bosentan
(dual endothelin receptor antagonist) were used. In order to determine the endothelial
influence on the electrolyte homeostasis, a spectroscopic investigation of
urine potassium and sodium concentration and urine osmolality were performed.
To analyze the exertion of influence on the congenital renin cell specific lack of
endothelin receptors of juxtaglomerular cells, an examination of renin production
using Ren-1d+/Cre-ETA
fl/fl mice took place. To scrutinize the role of ETA receptors
concerning the stimulation of the renin system, we focussed on the difference of the
renin production between wildtype and knockout strains. In summary, the localization
of endothelin receptors in the kidney of mice agrees to a large extent to the
renal expression of endothelin receptors of other mammalians. Analysis of electroly-

te concentrations did not reveal informative discoveries, an endothelial influence on
the electrolyte homeostasis can not be postulated. With regard to renin secretion,
the examinations showed no significant differences. Despite of renal expression of
endothelin receptors, neither antagonism nor renin cell deficience caused significant
influences on the renin system in vivo. This missing exertion of influence strongly
suggests the existence of compensatory mechanisms including prostaglandins and
nitric oxide. Furthermore, on the role of ETB receptors as well as on those of
endothelin isoforms ET-2 and ET-3 should be focused in detail.
