Non-coding RNAs can function as potent gene regulators and consequently, such RNA molecules have been implicated in many diseases including different types of cancer. Circular RNAs (circRNAs) are emerging as a new class of regulatory non-coding RNAs. Several studies have revealed that circRNAs are differentially expressed in several tissues and thus, some circRNAs were reported to play important roles in several diseases such as cancer, cardiovascular disease and neurological disease.
Using an intrasplenic tumor model for colorectal cancer, we have identified several circRNAs differentially expressed in primary tumors and liver metastases suggesting a role of these RNAs in tumor progression. In this study, we have focused on characterization of circular RNA ZNF609 (circZNF609), which is up-regulated in primary tumors as well as liver metastases in our mouse model. We developed methods for knockdown and overexpression of circZNF609 and established several inducible stable cell lines for in vitro and in vivo analyses. Using two xenograft mouse models, we confirmed that circZNF609 can function as oncogene to promote colorectal cancer progression.
Interestingly, we found that circZNF609 encodes for small proteins, which could potentially have function in tumor development. Using Flag reporter constructs and mutagenesis, we identified several AUGs (even exogenous AUGs) which can be used to initiate translation from circZNF609. Moreover, circZNF609 protein localizes to the cytoplasm while the parental ZNF609 protein exclusively localizes to the nucleus. Mechanistically, mass spectrometry analysis identified several proteins of the integrator complex as interaction partners of both circZNF609 and ZNF609 proteins suggesting an interesting mode of circZNF609 functions. Further analysis of the molecular mechanisms of these interactions would shed lights on how circZNF609 functions.

Circular RNAs (circRNAs) sind eine neue Klasse regulatorischer nicht-kodierender RNAs. Mehrere Studien haben gezeigt, dass circRNAs in verschiedenen Geweben differentiell exprimiert werden. Daher wurde berichtet, dass einige circRNAs eine wichtige Rolle bei verschiedenen Krankheiten wie Krebs, Herz-Kreislauf-Erkrankungen und neurologischen Erkrankungen spielen.
Unter Verwendung eines intrasplenalen Tumormodells für kolorektalen Krebs haben wir mehrere circRNAs identifiziert, die differentiell in primären Tumoren und Lebermetastasen exprimiert werden, was auf eine Rolle dieser RNAs bei der Tumorprogression hinweist. In dieser Studie haben wir uns auf die Charakterisierung der zirkulären RNA ZNF609 (circZNF609) konzentriert, die in Primärtumoren und Lebermetastasen in unserem Mausmodell hochreguliert ist. Wir entwickelten Methoden für Knockdown und Überexpression von circZNF609 und etablierten mehrere induzierbare stabile Zelllinien für In-vitro- und In-vivo-Analysen. Mit zwei Xenograft-Mausmodellen haben wir bestätigt, dass circZNF609 als Onkogen fungieren kann, um die Progression des kolorektalen Karzinoms zu fördern.
Interessanterweise fanden wir, dass circZNF609 für kleine Proteine kodiert, die möglicherweise in der Tumorentwicklung eine Rolle spielen könnten. Unter Verwendung von Flag-Reporterkonstrukten und Mutagenese identifizierten wir mehrere AUGs (sogar exogene AUGs), die verwendet werden können, um die Translation von circZNF609 zu initiieren. Außerdem lokalisiert sich das circZNF609-Protein im Cytoplasma, während das parentale ZNF609-Protein ausschließlich im Zellkern lokalisiert ist. Mechanistisch identifizierte die massenspektrometrische Analyse mehrere Proteine des Integratorkomplexes als Interaktionspartner von circZNF609 und ZNF609, was auf eine interessante Funktion von circZNF609 schließen lässt. Eine weitere Analyse der molekularen Mechanismen dieser Wechselwirkungen würde Aufschluss darüber geben, wie circZNF609 funktioniert.
