Die Lungentransplantation ist die einzige etablierte kurative Therapie für Patienten mit nicht malignen Lungenerkrankungen im Endstadium. Eine häufige Komplikation ist die Entstehung einer Bronchiolitis obliterans, bei der es zu narbig-fibrotischen Umbauvorgängen insbesondere der kleinen Atemwege und des Lungeninterstitiums kommt. Daher stehen Rezeptor-Tyrosin-Kinase-Inhibitoren (=RTK-I) als antifibrotische Wirkstoffe im Fokus der Forschung. In der vorliegenden Arbeit soll der Einfluss der RTK-I Imatininb, Nilotinib und Dasatanib auf die Kontraktionskraft von Rattenlungenfibroblasten untersucht werden. Dazu wurde zunächst ein Protokoll zur Kollagenherstellung und ein Kollagen-Kontraktionsassay (=CGC) mit entsprechenden Positiv- und Negativkontrollen etabliert.
Im CGC wurden die Fibroblasten mit Imatinib (100µM; 20µM), Nilotinib (40µM; 20µM) und Dasatanib (10µM; 5µM; 2,5µM; 1µM; 0,5µM und 0,1µM) inkubiert. Nach 24h wurde die Fläche und damit das Kontraktionsausmaß bestimmt und mit den Positiv- und Negativkontrollen verglichen. Bei Imatinib kam es bei beiden Konzentrationen zu einer Kontraktion. In der Vitalitätsfärbung mit Dapi und Calcein sahen wir allerdings nahezu keine vitalen Zellen. Der zytotoxische Effekt steht bei diesen Konzentrationen im Vordergrund. Bei Nilotinib sahen wir ein ähnliches Ergebnis. Die Zellen waren hier noch vital – die fehlende Kontraktionshemmung liegt möglicherweise in einer zu niedrigen Konzentration von Nilotinib. Unter Dasatanib beobachteten wir eine dosisanhängige Kontraktionshemmung. Die maximale Hemmung erreichten wir bei 10µM (57,9% ± 9,8; positiv: 21,8% ± 4,9; 43,0% ±11,2). Die mittlere Hemmkonzentration EC50 lag bei 2,4µM. Die Zellen waren bei allen Konzentrationen ausreichend vital. Dasatanib zeigte in unserer Studie in Anhängigkeit der Dosierung die stärkste Kontraktionshemmung. In der Anwendung ist dieses Medikament gut steuerbar und hat, wie aus der Literatur bekannt, eine positive Wirkung auf senescente Zellen, die unter Dasatanib abgetötet werden (hier nicht untersucht). Demnach könnte diesem Medikament eine wichtige Bedeutung in der Behandlung fibrotischer Erkrankungen, bzw. in unserem Fall, der Bronchiolitis obliterans zukommen.

Lung transplantation is the only established curative therapy for patients with end-stage non-malignant lung disease. A frequent complication is the development of bronchiolitis obliterans, in which there is scarring and fibrotic remodeling, especially of the small airways and the pulmonary interstitium. Therefore, receptor tyrosine kinase inhibitors (= RTK-I) as antifibrotic agents are the focus of research. In the present work, the influence of the RTK-I imatininb, nilotinib and dasatanib on the contraction force of rat lung fibroblasts is to be investigated. For this purpose, a collagen production protocol and a collagen contraction assay (= CGC) with corresponding positive and negative controls were first established.
In the CGC, the fibroblasts were incubated with imatinib (100µM; 20µM), nilotinib (40µM; 20µM) and dasatanib (10µM; 5µM; 2.5µM; 1µM; 0.5µM and 0.1µM). After 24 hours, the area and thus the extent of contraction was determined and compared with the positive and negative controls. Imatinib contracted at both concentrations. In the vitality staining with Dapi and Calcein, however, we saw almost no vital cells. The cytotoxic effect is in the foreground at these concentrations. We saw a similar result with nilotinib. The cells were still vital here - the lack of contraction inhibition may be due to an excessively low concentration of nilotinib. We observed dose-dependent inhibition of contraction with dasatanib. We reached the maximum inhibition at 10µM (57.9% ± 9.8; positive: 21.8% ± 4.9; 43.0% ± 11.2). The mean inhibitory concentration EC50 was 2.4µM. The cells were sufficiently vital at all concentrations. In our study, dasatanib showed the strongest inhibition of contraction, depending on the dose. The use of this drug is easy to control and, as known from the literature, has a positive effect on senescent cells that are killed by dasatanib (not investigated here). Accordingly, this drug could play an important role in the treatment of fibrotic diseases, or in our case, bronchiolitis obliterans.
