Background: The long-term outcome of lung transplantation (ltx) is majorly impaired by the development of chronic lung allograft dysfunction (CLAD), a disease, for which fibroproliferative changes in the small airways and vessels of the lung are characteristic. Antifibrotic drugs were shown to influence significantly the development of lung fibrosis, in vitro and in vivo. Therefore they are used for therapy.
Objectives: This study was performed to investigate the effect of receptor tyrosine kinase inhibitors (RTKIs) Imatinib, Dasatinib and Nilotinib on the proliferation of fibroblasts in cell culture.
Methods: Minor histoincompatible rats (F344-to-WKY) underwent unilateral, orthotopic, left ltx. The harvest of lungs occured after 60 days. Fibroblasts, from the right native and the left transplanted lung, respectively, were cultured. Cell vitality and viability were tested after 72 h incubation with the antiproliferative RTKIs via MTS (photometrics) and RealTime Glo (luminometrics).
Results: No difference between the viability of fibroblasts originating from the left and right lungs could be detected. Similar was observed between early (p1-2) and late (p3-4) passages. The EC50 of Dasatinib (approx. 0,05 μM) was significantly lower compared to EC50 of Nilotinib and Imatinib (approx. 7 μM). After incubation for 72 h, the fibroblasts treated with the highest concentration of Imatinib showed no vitality. At maximum concentration of Dasatinib fibroblasts were inactive, but still vital. No difference between photometric- and luminometric-based method was observed.
Conclusions: This study demonstrated a clear effect of RTKIs on the proliferation of fibroblasts in cell culture, with Dasatinib being already able to minimize cell growth at lower concentrations than Imatinib or Nilotinib. Our findings provide evidence for using RTKIs as valid candidates to combat CLAD.
