Bei Hypericum tetrapterum Fr. (Hypericaceae) handelt es sich um eine mehrjährige, krautige Hypericum-Art mit einem vierflügeligen, hohlen Stängel, dessen Verbreitungsgebiet sich über Europa, Westasien und Nordafrika erstreckt. Entsprechend der Unterteilung des Botanikers N.K.B Robson der Hypericum-Arten in 36 Sektionen gegliedert hat, wird die untersuchte Hypericum-Art H. tetrapterum zusammen mit dem wohl prominentesten Vertreter der Hypericaceae, H. perforatum, und weiteren 40 Arten der Sektion 9 Hypericum zugeordnet. Die Sekundärzusammensetzung und die pharmakologische Aktivität der Inhaltsstoffe von H. tetrapterum, insbesondere die der Acylphloroglucinole, ist bisher weitgehend unbekannt. Dagegen ist H. perforatum nicht nur phytochemisch gut untersucht, sondern auch für die antidepressiven, antiviralen, antibakteriellen und antitumoralen Effekte seiner Extrakte bekannt.
Für die Isolierung und anschließende pharmakologische in vitro Testung von Acylphloroglucinolen wurde ein Petrolether-Extrakt aus den getrockneten, oberirdischen Pflanzenteilen von H. tetrapterum hergestellt. Nach ¹H-NMR geleiteter Fraktionierung wurden die Strukturen der 17 erhaltenen Verbindungen mittels ¹H-, ¹³C- und 2D-NMR-Spektroskopie sowie mit Daten aus der Massenspektrometrie aufgeklärt. Die isolierten Substanzen lassen sich in zwei verschiedene Strukturgruppen unterteilen:
Zur ersten Gruppe gehören die monozyklischen Acylphloroglucinole. Die Substanzen 1 bis 8 stellen allesamt O-geranylierte Verbindungen dar, die sich anhand ihrer para- bzw. ortho-Stellung der Geranyl-Einheit unterscheiden. Alle isolierten monozyklischen Verbindungen besitzen durchweg eine aliphatische Acylseitenkette. Sowohl die bereits aus weiteren Hypericum-Arten bekannten Substanzen 3 (Otogirin), 4 (Adotogirin) als auch Substanz 7 (Tetrapterothean) liegen zusätzlich methyliert vor. Substanz 8 (Tetrapteroronjan) weicht strukturell von den übrigen isolierten monozyklischen Verbindungen ab, da sie ähnlich zu den alpha-Bittersäuren des Hopfens, hydroxyliert bzw. methyliert wurde und somit keinen aromatischen Phloroglucin-Ring aufweist.
Die zweite Gruppe stellen die bizyklischen Acylphloroglucinole vom Furan-Typ dar. Die isolierten Substanzen 9 bis 17 weisen ausschließlich aliphatische Acylseitenketten auf. Bei den Substanzen 9 bis 15 fand durch eine vorhergehende Geranylierung in Position 3 am Phloroglucin-Grundkörper ein Ringschluss zum Furan mit der Hydroxygruppe in para-Stellung zur Acylseitenkette statt. Im Gegensatz dazu handelt es sich bei den Substanzen 16 und 17 um eine 5-ortho-Furan-Ringbildung. Im Rahmen dieser Arbeit konnte erstmals der 3-para-Ringschluss zum Furan für bizyklische Acylphloroglucinole beschrieben werden, die aus einer Hypericum-Art isoliert wurden.
Die Kombination aus monozyklischen, O-geranylierten Acylphloroglucinolen und bizyklischen Acylphloroglucinolen vom Furan-Typ werden hier das erste Mal für eine Hypericum-Art beschrieben. Vergleichbare bizyklische Acylphloroglucinole vom Furan-Typ finden sich bisher ausschließlich in der Sektion 9 Hypericum, wohingegen monozyklische, O-geranylierte Verbindungen in den Sektionen 9, 9c, 10, 18, 19, 20, 26 und 30 vorkommen. Um die in der vorliegenden Arbeit angedeuteten chemotaxonomischen Zusammenhänge zu bestätigen, ist es notwendig weitere Hypericum-Arten, insbesondere der Sektion 9, phytochemisch zu untersuchen.
In einem in vitro Testsystem wurden der PE-Extrakt, die Fraktion PE-F1, als auch die Substanzen 1 bis 3, 6, 8 bis 14 sowie 16 und 17 auf ihre zytotoxische Wirkung gegenüber der Zelllinien HeLa, MCF-7, HepG2, PC-3 und SK-MEL-28 untersucht. Hierbei zeigten sich zelllinienabhängige und strukturabhängige Aktivitätsunterschiede zwischen den getesteten monozyklischen Verbindungen und den bizyklischen Acylphloroglucinolen vom Furan-Typ: Die aromatischen, monozyklischen Acylphloroglucinole wiesen niedrigere IC₅₀-Werte (20 bis 60 µM), als die bizyklischen Acylphloroglucinole (36 bis 106 µM) bei der Testung mit HeLa-Zellen auf. Eine Ausnahme bildet Substanz 8, die in keiner der untersuchten Zelllinien zytotoxisch aktiv war. Das monozyklische Acylphloroglucinol 3 wirkte am stärksten zytotoxisch gegenüber allen getesteten Zelllinien. Dadurch kann angenommen werden, dass eine Methylierung allgemein die zytotoxische Aktivität positiv beeinflusst. Die Substitution der Acylseitenkette scheint im untersuchten Substanz-Set keinen nennenswerten Effekt auf die zytotoxische Wirkung zu haben. Durch die fehlende Aktivität der bizyklischen Acylphloroglucinole 9 und 11 gegenüber der Zelllinien MCF-7, HepG2, PC-3 und SK-MEL-28 kann von einer selektiven Wirkung dieses Strukturtypus auf HeLa-Zellen ausgegangen werden. Welche biologische Zielstruktur genau für diese Selektivität verantwortlich ist, konnte im Rahmen dieser Arbeit jedoch nicht eindeutig geklärt werden, vermutlich besteht jedoch ein Zusammenhang mit der Aromatizität des Phloroglucin-Ringes.
Abschließend wurden der PE-Extrakt, Fraktion PE-F1 als auch die Substanzen 3, 4, 6 bis 9 und 11 bis 15 sowie die Substanzen einer in vitro Untersuchung auf ihre antibakterielle Aktivität unterzogen. Bei der Testung gegen den gram-positiven Keim Staphylococcus aureus zeigten die aromatischen, monozyklischen Verbindungen niedrigere MIC-Werte (2 bis 8 mM) als die schwach aktiven bizyklischen Acylphloroglucinole vom Furan-Typ (MIC 128 bis 256 mM). Bei Betrachtung der Aktivitäten der monozyklischen Acylphloroglucinole ergaben sich auch hier wieder strukturell bedingte Unterschiede. Es zeigt sich, dass eine Methylierung einen positiven Effekt auf die antibakterielle Aktivität hat. Dagegen scheint die fehlende Aromatizität des Phloroglucin-Ringes die antibakterielle Wirkung deutlich zu verringern. Innerhalb der bizyklischen Acylphloroglucinole vom Furan-Typ zeigt sich eine Abnahme der antibakteriellen Wirkung mit steigender Lipophilie bzw. Prenylierung der Substanzen. Die Aktivität des PE-Extraktes sowie der Fraktion PE-F1 ist vermutlich auf die monozyklischen Verbindungen zurückzuführen. Die Voruntersuchung des PE-Extraktes und der Fraktion PE-F1 mit dem gram-negativen Testkeim Escherichia coli zeigte keine messbare antibakterielle Wirkung, sodass auch von keiner Wirkung der Reinsubstanzen ausgegangen werden kann.

Hypericum tetrapterum Fr. (Hypericaceae) is a perennial, herbaceous Hypericum species with a four-winged, hollow stem. The distribution area extends across Europe, West Asia, and North Africa. According to the botanist N.K.B Robson's subdivision of Hypericum species into 36 sections, the Hypericum species under investigation, H. tetrapterum, is assigned to section 9 Hypericum together with the most prominent representative of the Hypericaceae, H. perforatum, and a further 40 species. The chemical profile, in particular the composition of the acylphloroglucinols, and pharmacological activity of the constituents of H. tetrapterum, is still largely unknown. In contrast, H. perforatum has not only been well studied phytochemically, but is also known for the antidepressant, antiviral, antibacterial and antitumor effects of its extracts.
For the isolation and subsequent pharmacological in vitro testing of acylphloroglucinols, a petroleum ether extract was prepared from the dried aerial parts of H. tetrapterum. After ¹H-NMR guided fractionation, the structures of the 17 compounds obtained were elucidated using ¹H-, ¹³C- and 2D-NMR spectroscopy as well as data from mass spectrometry. The isolated substances can be divided into two different structural groups:
The monocyclic acylphloroglucinols belong to the first group. Substances 1 to 8 are all O-geranylated compounds, which can be distinguished by the para- or ortho-position of the geranyl unit. All isolated monocyclic compounds have an aliphatic acyl side chain. Substances 3 (otogirin), 4 (adotogirin), which are already known from other Hypericum species, and 7 (tetrapterothean), are also methylated. Substance 8 (tetrapteroronjan) differs structurally from the other isolated monocyclic compounds, as it was hydroxylated and methylated like the alpha-bitter acids of hops and therefore has no aromatic phloroglucin ring.
The second group consists of the bicyclic acylphloroglucinols of the furan type. The isolated substances 9 to 17 have exclusively aliphatic acyl side chains. In the case of substances 9 to 15, a ring closure to the furan with the hydroxy group in the para position to the acyl side chain took place through a previous geranylation in position 3 on the phloroglucin base. In contrast, substances 16 and 17 involve a 5-ortho-furan ring formation. In this work, the 3-para ring closure to furan was described for the first time for bicyclic acylphloroglucinols isolated from a Hypericum species.
The combination of monocyclic, O-geranylated acylphloroglucinols and bicyclic acylphloroglucinols of the furan type is described here for the first time for a Hypericum species. Comparable bicyclic acylphloroglucinols of the furan type have so far been found exclusively in section 9 Hypericum, whereas monocyclic, O-geranylated compounds occur in sections 9, 9c, 10, 18, 19, 20, 26 and 30. To confirm the chemotaxonomic relationships indicated in the present work, it is necessary to phytochemically investigate further Hypericum species, especially those of section 9.
In an in vitro test system, the PE extract, the fraction PE-F1, as well as the substances 1 to 3, 6, 8 to 14, as well as 16 and 17 were evaluated for their cytotoxic effect against the cell lines HeLa, MCF-7, HepG2, PC-3 and SK-MEL-28. This revealed cell line-dependent and structure-dependent differences in activity between the tested monocyclic compounds and the bicyclic acylphloroglucinols of the furan type: The aromatic, monocyclic acylphloroglucinols exhibited lower IC₅₀ values (20 to 60 μM), than the bicyclic acylphloroglucinols (36 μM to 106 μM) when tested with HeLa cells. An exception is substance 8, which was not cytotoxic in any of the cell lines. The monocyclic acylphloroglucinol 3 had the strongest cytotoxic effect on all tested cell lines. It can therefore be assumed that methylation generally has a positive effect on cytotoxic activity. The substitution of the acyl side chain does not appear to have any significant effect on the cytotoxic effect in this set of compounds. Due to the lack of activity of the bicyclic acylphloroglucinols 9 and 11 against the cell lines MCF-7, HepG2, PC-3 and SK-MEL-28, a selective effect of this structure type on HeLa cells can be assumed. However, the exact molecular target responsible for this selectivity could not be clarified in the course of this work, but there is probably a connection with the aromaticity of the phloroglucinol ring.
Finally, the PE extract, fraction PE-F1 as well as substances 3, 4, 6 to 9 and 11 to 15 were subjected to an in vitro test for their antibacterial activity. When tested with the gram-positive germ Staphylococcus aureus, the aromatic, monocyclic compounds showed lower MIC values (2 to 8 mM) than the weakly active bicyclic acylphloroglucinols of the furan type (MIC 128 to 256 mM). When looking at the activities of the monocyclic acylphloroglucinols, structural differences were again determined. The methylation had a positive effect on antibacterial activity. In contrast, the lack of aromaticity of the phloroglucinol ring appeared to significantly reduce the antibacterial effect. Within the bicyclic acylphloroglucinols of the furan type, a decrease in antibacterial activity was observed with increasing lipophilicity or prenylation of the substances. The activity of the PE extract and the PE-F1 fraction is presumably due to the monocyclic compounds. Preliminary testing of the PE extract and the PE-F1 fraction with the gram-negative test germ Escherichia coli showed no measurable antibacterial effect, meaning that no activity of the pure substances can be assumed either.
