Abstract (German)

In der vorliegenden Dissertation wurde die Bedeutung der oxidativ aktivierten CaMKII für die Kardiotoxizität von Doxorubicin in der isolierten Herzmuskelzelle erforscht. Hierfür wurden Messungen am Epifluoreszenzmikroskop an wildtyp-Mäusen und Oxiation-resistenten CaMKIIdelta Knock-in Mäusen durchgeführt. In der vorliegenden Dissertation wurde die Bedeutung der oxidativ aktivierten CaMKII für die Kardiotoxizität von Doxorubicin in der isolierten Herzmuskelzelle erforscht. Hierfür wurden Messungen am Epifluoreszenzmikroskop an wildtyp-Mäusen und Oxiation-resistenten CaMKIIdelta Knock-in Mäusen durchgeführt. Hier wurde ein messbarer negativ inotroper Effekt zwischen den Gruppen erst ab einer Dosis von 50 µM Doxorubicin nachgewiesen. Der Ausmaß der Inotropieeinschränkung zwischen wt Zellen und CaMKIIdelta Met281/282Val Zellen unterscheidet jedoch nicht. Zudem fand sich, dass die fehlende oxidative Aktivierbarkeit der CaMKIIdelta mit einer geringeren diastolischen Calciumüberladung und einem erhaltenen SR Calciumgehalt bei einer Doxorubicinkonzentration von 10 μM einhergeht, was in Zusammenschau mit der unveränderten Relaxationskinetik hinweisgebend auf ein SR Calciumleck ist, welches durch eine oxidativ aktivierte CaMKIIdelta unterhalten wird.

Translation of the abstract (English)

Objective of the present dissertation was the role of the oxidative activated CaMKII for the cardiotoxicity of doxorubicin in the isolated heart cell. For this have been made measures at the epifluorescence microscope of wildtype and oxidation resistant CaMKII delta knock-in mice. Objective of the present dissertation was the role of the oxidative activated CaMKII for the cardiotoxicity of doxorubicin in the isolated heart cell. For this have been made measures at the epifluorescence microscope of wildtype and oxidation resistant CaMKII delta knock-in mice. A negative inotrope effect between des groups have been found at a dose of 50 µM doxorubicin. The dimension of the restriction between wt cells and CaMKIIdelta Met 281/282Val cells wasn’t significant different. In addition, the data has shown that the missing activation through oxidation comes with a less diastolic calcium overload with a unchanged SR calcium load at a concentration of doxorubicin of 10 µM. In summary with the unchanged kinetic of relaxation these findings indicative a SR calcium leak.
